Abstract: Biomolecule environments can enhance chemistries with the potential to mediate and modulate self-modification (e.g., self-cleavage). While these enhanced modes are found in certain biomolecules (e.g., RNA ribozymes), it is more rare in proteins. Targeted proteolytic cleavage is vital to physiology, biotechnology, and even emerging therapy. Yet, purely chemically induced methods for the site-selective cleavage of proteins remain scarce. Here, as a proof of principle, we designed and tested a system intended to combine protein-enhanced chemistry with tag modification to enable synthetic reductive protein chemistries promoted by diboron. This reductively driven, single-electron chemistry now enables an operationally simple, site-selective cleavage protocol for proteins directed to readily accessible dehydroalanine (Dha) residues as tags under aqueous conditions and in cell lysates. In this way, a mild, efficient, enzyme-free method now allows not only precise chemical proteolysis but also simultaneous use in the removal of affinity tags and/or protein-terminus editing to create altered N- and C-termini such as protein amidation (─CONH 2 ).

Abstract: Many pathogens exploit host cell-surface glycans. However, precise analyses of glycan ligands binding with heavily modified pathogen proteins can be confounded by overlapping sugar signals and/or compounded with known experimental constraints. Universal saturation transfer analysis (uSTA) builds on existing nuclear magnetic resonance spectroscopy to provide an automated workflow for quantitating protein-ligand interactions. uSTA reveals that early-pandemic, B-origin-lineage severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike trimer binds sialoside sugars in an “end-on” manner. uSTA-guided modeling and a high-resolution cryo–electron microscopy structure implicate the spike N-terminal domain (NTD) and confirm end-on binding. This finding rationalizes the effect of NTD mutations that abolish sugar binding in SARS-CoV-2 variants of concern. Together with genetic variance analyses in early pandemic patient cohorts, this binding implicates a sialylated polylactosamine motif found on tetraantennary N-linked glycoproteins deep in the human lung as potentially relevant to virulence and/or zoonosis.

Abstract: From 2002 to 2014, the Portland Police Bureau reported large reductions in complaints against officers and use of force indicators. The purpose of this paper is to develop a case study to document these changes and explore possible influences. The paper maps the changes in conduct indicators against the developing relationship between the Bureau and the Portland Independent Police Review Division, and changes in policies and procedures. Public complaints reduced by 54.4 per cent, while the rate of specific allegations per officer fell by 70.1 per cent. Quarterly use of force incident reports were reduced by 65.4 per cent between 2008 and 2014. Annual average shootings decreased from a high of nine per year across 1997-2002 to just below four per year in 2009-2014. Fatal shootings also trended downward but remained two per year in the last three years on record. Reforms instituted during this period that may have influenced these trends include a more rigorous complaints process, an early intervention system (EIS), enhanced external and internal review mechanisms, policy changes and training initiatives. The researchers were unable to control for a range of additional variables that may have influenced the findings, including police deployments and changes in officer demographics. The study provides support for strategies to improve police conduct including external oversight, diagnostic research, training focussed on de-escalation and minimal force, and complaint profiling and EISs. There are very few studies available showing large long-term reductions in adverse police conduct indicators.

Abstract: Understanding and engaging with assessment feedback is a crucial step in contributing to a student's development of academic skills and content knowledge. However, in order for feedback to be effective, students need to be aware of its importance and how to engage with it. Preliminary benchmarking of the tertiary education sector in Australia demonstrates that publicly and openly accessible student resources on feedback are not available. Rather, most student resources at universities focus on common academic and study skills. At the University of Southern Queensland, a student resource was developed to inform students on the value of feedback and how to engage with it through a three-step process. This resource was embedded in first-year courses as part of a suite of academic and study skills resources. This chapter explores how student feedback resources have the potential to positively contribute to student feedback literacy, learning and development.
