Approaches to improving the biocompatibility of porous perfluoropolyethers for ophthalmic applications

Abstract: Porous perfluoropolyether (PFPE) membranes for ophthalmic applications were prepared with a zwitterion monomer, 3-[[2-(methacryloxy) ethyl](N,N-dimethyl)ammonio]-propane-1-sulphonate, copolymerized in weight ratios of 0-10%. The polymer s les were assessed for a range of physical properties, including equilibrium water content, bovine serum albumin permeability, transparency, refractive index and the ability to support corneal epithelial cell and tissue attachment, growth and migration. In vitro assessment of the polymers using bovine corneal epithelial cells and tissue showed that a zwitterion incorporation level of between 0% and 6% in the PFPE membranes supported the migration of an intact sheet of epithelial tissue without compromising epithelial cell attachment and growth, with 4-6% being the optimal level for these properties. Binding patterns of the cell adhesion glycoprotein fibronectin were also found to reflect the cell and tissue response. Effective nutrient permeability, refractive index and optical transparency were also maintained by the porous PFPE polymers containing this concentration of zwitterionic monomer. The presence of amounts of zwitterion greater than 6% was inhibitory to both tissue migration and cell growth and was associated with increased optical haze. These results demonstrated that it is possible to achieve the potential for increased biocompatibility in zwitterion-containing PFPE polymers without compromising existing beneficial characteristics.

Surface Modification of Multiwalled Carbon Nanotubes with Engineered Self-Assembled RAFT Diblock Coatings

Abstract: A facile method to modify the surface of multiwalled carbon nanotubes (MWCNTs) via electrostatic interactions between polyelectrolytes and oxidized MWCNTs was developed. Diblock copolymers containing poly[2-(methacryloyloxy)ethyltrimethylammonium chloride] (PMETAC), a positively charged block, and poly(ethylene glycol) methacrylate (PEGMA), a neutral block, with tailored molecular weights and low polydispersities were synthesized by reversible addition–fragmentation chain transfer (RAFT) polymerization. Acid treated-MWCNTs were coated with the RAFT diblock copolymers to improve their dispersibility in aqueous phosphate buffered saline (PBS) solution. The short positively charged PMETAC block was designed to attach the block copolymers to the surface of MWCNTs via electrostatic interactions, whereas the PEGMA block improved dispersibility of the MWCNTs in aqueous solutions. Extensive screening of the diblock copolymers with different degrees of polymerization (DP) showed that the dispersion stability of the polymer-coated MWCNTs in PBS was greatly improved with increasing chain length of the PEGMA block. In particular, the MWCNTs coated with a diblock copolymer containing PEGMA (DP = 118, the longest block investigated) showed superior dispersion stability in both water and PBS solution.

Highly Ordered Honeycomb Film Formation of Linear Polymers by the Breath Figure Technique

Abstract: Highly ordered, porous honeycomb (HC) films were prepared by the breath figure technique from linear polymers poly(methyl methacrylate) (PMMA) and polystyrene (PS). Typically HC films are difficult to form from such simple linear polymers. The addition of a novel fluorinated polymer (FP) additive with as little as 1 wt-% to PMMA or 5 wt-% to PS was required to obtain regular porous HC films. Through investigation of the influence of the additive on the polymer properties, three parameters based on interfacial tension, polymer solution viscosity, and polymer solidification rate were identified as key factors affecting the ability of polymer systems to form regular porous HC films. A new hypothesis was subsequently developed based on the relationships of these parameters to explain the unusual behaviour associated with HC film formation from linear PMMA and PS with addition of FP additive. This work will provide a new tool to guide the formation of HC films and will greatly broaden the range of polymers used to form HC films in the future.
Azobenzene moiety variation directing self-assembly and photoresponsive behavior of azo-surfactants

Abstract: We report two photoresponsive hiphiles with comparable structures consisting of hydrophilic oligooxyethylene units, a hydrophobic alkyl chain and a light-sensitive azobenzene moiety, demonstrating that self-assembly and photoresponsive behaviour is extremely sensitive to the position of the photoactive moiety within the surfactant molecular architecture.

Abstract: Novel syntheses of 4,5-dihydro-1,2,4-triazin-6(1H)-ones were developed by use of imidoyl chlorides. The overall yield of 4,5-dihydro-1,2,4-triazin-6(1H)-ones prepared from amino acid imidates and hydrazines was improved by the development of a much more efficient synthesis of the imidates. The new 1,2-, 1,4- and 5,5-dimethyl dihydro-1,2,4-triazin-6(1H)-ones have been synthesized by cyclocondensation/cycloaddition pathways. Base-catalysed methylation of 3-phenyl-4,5-dihydro-1,2,4-triazin-6(1H)-one (15) gave the 1- methylated derivative (8) under similar conditions 2-methyl-3-phenyl-2,5-dihydro-1,2,4-triazin-6(1H)-one (9) afforded the 1,2-dimethyl derivative (7).
Water-dispersible magnetic carbon nanotubes as T2-weighted MRI contrast agents

Abstract: An efficient MRI T2-weighted contrast agent incorporating a potential liver targeting functionality was synthesized via the combination of superparamagnetic iron oxide (SPIO) nanoparticles with multiwalled carbon nanotubes (MWCNTs). Poly(diallyldimethylammonium chloride) (PDDA) was coated on the surface of acid treated MWCNTs via electrostatic interactions and SPIO nanoparticles modified with a potential targeting agent, lactose-glycine adduct (Lac-Gly), were subsequently immobilized on the surface of the PDDA-MWCNTs. A narrow magnetic hysteresis loop indicated that the product displayed superparamagnetism at room temperature which was further confirmed by ZFC (zero field cooling)/FC (field cooling) curves measured by SQUID. The multifunctional MWCNT-based magnetic nanocomposites showed low cytotoxicity in vitro to HEK293 and Huh7 cell lines. Enhanced T2 relaxivities were observed for the hybrid material (186 mM(-1) s(-1)) in comparison with the pure magnetic nanoparticles (92 mM(-1) s(-1)) due to the capacity of the MWCNTs to "carry" more nanoparticles as clusters. More importantly, after administration of the composite material to an in vivo liver cancer model in mice, a significant increase in tumor to liver contrast ratio (277%) was observed in T2 weighted magnetic resonance images.
A simple way to track single gold-loaded alginate microcapsules using x-ray CT in small animal longitudinal studies

Abstract: The use of alginate based microcapsules to deliver drugs and cells with a minimal host interaction is increasingly being proposed. A proficient method to track the position of the microcapsules during such therapies, particularly if they are amenable to commonly used instrumentation, would greatly help the development of such treatments. Here we propose to label the microcapsules with gold nanoparticles to provide a bright contrast on small animal x-ray micro-CT systems enabling single microcapsule detection. The microcapsules preparation is based on a simple protocol using inexpensive compounds. This, combined with the widespread availability of micro-CT apparatus, renders our method more accessible compared with other methods. Our labeled microcapsules showed good mechanical stability and low cytotoxicity in-vitro. Our post-mortem rodent model data strongly suggest that the high signal intensity generated by the labeled microcapsules permits the use of a reduced radiation dose yielding a method fully compatible with longitudinal in-vivo studies. The authors of this study report the development of a micro-CT based tracking method of alginate-based microcapsules by incorporating gold nanoparticles in the microcapsules. They demonstrate the feasibility of this system in rodent models, where due to the high signal intensity, even reduced radiation dose is sufficient to track these particles, providing a simple and effective method to track these commonly used microcapsules and allowing longitudinal studies.

Photo‐Modulated Therapeutic Protein Release from a Hydrogel Depot Using Visible Light

Abstract: The use of biomacromolecular therapeutics has revolutionized disease treatment, but frequent injections are required owing to their short half-life in vivo. Thus there is a need for a drug delivery system that acts as a reservoir and releases the drug remotely "on demand". Here we demonstrate a simple light-triggered local drug delivery system through photo-thermal interactions of polymer-coated gold nanoparticles (AuNPs) inside an agarose hydrogel as therapeutic depot. Localized temperature increase induced by the visible light exposure caused reversible softening of the hydrogel matrix to release the pre-loaded therapeutics. The release profile can be adjusted by AuNPs and agarose concentrations, light intensity and exposure time. Importantly, the biological activity of the released bevacizumab was highly retained. In this study we demonstrate the potential application of this facile AuNPs/hydrogel system for ocular therapeutics delivery through its versatility to release multiple biologics, compatibility to ocular cells and spatiotemporal control using visible light.
The effect of silicone hydrogel contact lens composition on dexamethasone release

Abstract: The relationship between the delivery of dexamethasone and the composition of silicone hydrogel materials was investigated. Two hydrophilic monomers (2-hydroxyethyl methacrylate or N,N-dimethylacrylamide), a siloxy methacrylate-based monomer (1-(Bis(trimethylsiloxy)methylsilyl)propoxy-3-metacryloxy-2-propanol, a polysiloxane (monomethacryloxypropyl-terminated polydimethylsiloxane) and a polymerizable silicone surfactant (Silmer ACR A008-UP) were used to synthesize silicone hydrogels of variable composition. The materials properties, such as surface wettability and equilibrium water content, were highly dependent on polymer composition. All dexamethasone-loaded hydrogels showed uptake that was driven primarily by sorption to the polymer phase. Furthermore, a positive correlation between loading mass and equilibrium water content was established. The duration of drug release from the hydrogels ranged from one to greater than two weeks depending on the monomer composition and relative contribution of hydrophilic and hydrophobic monomers. Higuchi model rate constants for the release showed strong correlation with the equilibrium water content, signifying that the release is likely controlled by aqueous phase diffusion.

Copolymerization of Pentafluorophenylmethacrylate with Hydrophilic Methacrylamide Monomers Induces Premature Hydrolytic Cleavage

Abstract: Active ester polymers are commonly used for fast development of novel polymer libraries, but they require post-polymerization modification, which is not atom-efficient or economical. In order to more efficiently produce 2-hydroxypropyl methacrylamide (HPMAm) libraries, it would be advantageous to perform a direct copolymerization with active ester monomers. In this work, the synthesis of copolymer libraries of pentafluorophenyl methacrylate (PFPMA) and the hydrophilic monomer HPMAm is investigated. Surprisingly, HPMAm induces premature hydrolytic cleavage of PFPMA, which occurs during polymerization and depends on the HPMAm/PFPMA feed ratio. Copolymerization of PFPMA with N-isopropylmethacrylamide and the methacrylate monomers 2-hydroxypropylmethacrylate and N-isopropylmethacrylate reveals that the hydrolytic cleavage is promoted by copolymerization with methacrylamides only. By switching from a thermal- to a light-based initiator and lowering the reaction temperature, premature hydrolytic cleavage of PFPMA is avoided and allows direct copolymerization of HPMAm together with PFPMA to create polymer libraries for biomaterial screening.

Multistimuli responsive organogels based on a reactive azobenzene gelator

Abstract: A reactive azobenzene based super organogelator was found to rapidly and reversibly transform a range of hydrophobic solvents from gels to solutions upon changes in temperature, light and shear force. More specifically they formed gels at concentrations as low as 0.08 wt%. Upon heating, exposure to UV light, or application of shear, the π-π bonding was disrupted which resulted in a rapid drop of both modulus and viscosity. This was confirmed by (1)H NMR, SAXS, and rheological measurements. Although many ex les of organogelators are known in the literature, this is the first time that a reactive group, a benzoyl chloride, has been incorporated in a supramolecular organogel structure. Moreover, this group is available for subsequent synthetic modifications. The presence of benzoyl chloride groups showed a remarkable effect on the formation and properties of the gels. Compared with other approaches, this strategy is advantageous in terms of structural design since it not only produces a multi-responsive soft material but also allows facile modifications which may expand the applications of organogels to other fields.

Reversible Photorheological Lyotropic Liquid Crystals

Abstract: We describe novel lyotropic liquid-crystalline (LLC) materials based on photoresponsive hiphiles that exhibit rapid photoswitchable rheological properties of unprecedented magnitude between solidlike and liquidlike states. This was achieved through the synthesis of a novel azobenzene-containing surfactant (azo-surfactant) that actuates the transition between different LLC forms depending on illumination conditions. Initially, the azo-surfactant/water mixtures formed highly ordered and viscous LLC phases at 20-55 wt % water content. Spectroscopic, microscopic, and rheological analysis confirmed that UV irradiation induced the trans to cis isomerization of the azo-surfactant, leading to the disruption of the ordered LLC phases and a dramatic, rapid decrease in the viscosity and modulus resulting in a 3 orders of magnitude change from a solid (20,000 Pa) to a liquid (50 Pa) at rate of 13,500 Pa/s. Subsequent exposure to visible light reverses the transition, returning the viscosity essentially to its initial state. Such large, rapid, and reversible changes in rheological properties within this LLC system may open a door to new applications for photorheological fluids.

Abstract: Polymerizing nanodroplets at solid-liquid interfaces is a facile solution-based approach to the functionalization of large surface areas with polymeric lens-shaped nanostructures. In this work, we have applied a one-pot approach to obtain polymeric nanolenses with controlled sizes and densities. We take advantage of the formation mechanism by the direct adsorption of nanodroplets from a surfactant-free microemulsion onto an immersed hydrophobic substrate. The interfacial nanodroplets were photopolymerized to produce polymeric nanolenses on the substrate surface. The surfactant-free microemulsion of the monomer nanodroplets was obtained through the spontaneous emulsification (i.e., ouzo effect) in the tertiary system of ethanol, water, and precusor monomer. The size of nanolenses on the surface was adjusted by the nanodroplet size, following a linear relationship with the ratio of the components in the microemulsion. This simple approach is applicable to produce nanolenses over the entire surface area or on any specific area at will by depositing a drop of the microemulsion. Possessing high optical transparency, the resulting substrates may have potential application as functional biomedical supporting materials or effective light-harvesting coatings.

Honeycomb Films from Perfluoropolyether-Based Star and Micelle Architectures

Abstract: A perfluoropolyether-b-poly(t-butyl acrylate) (PFPE-b-PtBA) block copolymer macroinitiator was used to prepare both core cross-linked star (CCS) polymers and micelles, whereby the outer shell and core, respectively, are comprised of fluorinated segments. The star polymer complete with PFPE outer shell was synthesised via atom transfer radical polymerisation (ATRP) and the arm-first approach, through cross-linking of the PFPE-b-PtBA macroinitiator with ethylene glycol diacrylate (EGDA). Alternatively, the PFPE-b-PtBA block copolymer could be self-assembled in benzene to form micelles with a PtBA shell and PFPE core. Both the micelle and CCS polymer were subsequently fabricated into non-cracking honeycomb (HC) patterned films on both planar and non-planar surfaces via the ‘Breath Figure’ (BF) technique using a static casting system.

Colloidally Stabilized Magnetic Carbon Nanotubes Providing MRI Contrast in Mouse Liver Tumors

Abstract: The use of medical imaging contrast agents may lead to improved patient prognosis by potentially enabling an earlier detection of diseases and therefore an earlier initiation of treatments. In this study, we fabricated superparamagnetic iron oxide (SPIO) nanoparticles within the inner cavity of multiwalled carbon nanotubes (MWCNTs) for the first time thereby ensuring high mechanical stability of the nanoparticles. A simple, but effective, self-assembled coating with RAFT diblock copolymers ensured the SPIO-MWCNTs have a high dispersion stability under physiological conditions. In vivo acute tolerance testing in mice showed a high tolerance dose up to 100 mg kg(-1). Most importantly, after administration of the material a 55% increase in tumor to liver contrast ratio was observed with in vivo MRI measurements compared to the preinjection image enhancing the detection of the tumor.
A Novel Route to Prepare Highly Reactive and Versatile Chromatographic Monoliths

Abstract: The copolymer poly(isocyanatoethyl methacrylate-co-methyl methacrylate-co-ethylene glycol dimethacrylate) (poly(IEM-co-MMA-co-EGDMA)) was developed as a novel, facile, highly reactive and versatile monolithic matrix, which was amenable to surface functionalization with a variety of nucleophilic modifiers based on the reactive isocyanate group, producing monoliths of various chemistries suitable for chromatography. The specific surface area, pore size distribution, porosity and morphology of the monolithic matrix were characterized using a mercury intrusion porosimeter and scanning electron microscopy (SEM), respectively. Thermal analysis results revealed that the monolith was thermally stable up to 307 °C. The success of the chemical modification of the monolithic matrix was confirmed by FT-IR, solid state (13) C NMR and XPS elemental analysis, showing the high ligand density of the modified monoliths. A ligand density of up to 2.33 mmol·mL(-1) was obtained for the 1-octanol modified monolith (M1) with an isocyanate group conversion of 96.9%, indicating a high efficiency of the modification reaction. The potential application of the monoliths was demonstrated by the separation of a series of compounds. The novel monolithic columns exhibited high mechanical stability, column efficiency and good repeatability and reproducibility.

Variation in Hydrogel Formation and Network Structure for Telo-, Atelo- and Methacrylated Collagens

Abstract: As the most abundant protein in the extracellular matrix, collagen has become widely studied in the fields of tissue engineering and regenerative medicine. Of the various collagen types, collagen type I is the most commonly utilised in laboratory studies. In tissues, collagen type I forms into fibrils that provide an extended fibrillar network. In tissue engineering and regenerative medicine, little emphasis has been placed on the nature of the network that is formed. Various factors could affect the network structure, including the method used to extract collagen from native tissue, since this may remove the telopeptides, and the nature and extent of any chemical modifications and crosslinking moieties. The structure of any fibril network affects cellular proliferation and differentiation, as well as the overall modulus of hydrogels. In this study, the network-forming properties of two distinct forms of collagen (telo- and atelo-collagen) and their methacrylated derivatives were compared. The presence of the telopeptides facilitated fibril formation in the unmodified s les, but this benefit was substantially reduced by subsequent methacrylation, leading to a loss in the native self-assembly potential. Furthermore, the impact of the methacrylation of the collagen, which enables rapid crosslinking and makes it suitable for use in 3D printing, was investigated. The crosslinking of the methacrylated s les (both telo- and atelo-) was seen to improve the fibril-like network compared to the non-crosslinked s les. This contrasted with the s les of methacrylated gelatin, which showed little, if any, fibrillar or ordered network structure, regardless of whether they were crosslinked.

Gelatin-Based Photocurable Hydrogels for Corneal Wound Repair

Abstract: In this study, an injectable, photocurable gelatin system, consisting of acrylated gelatin and thiolated gelatin, with tunable mechanical, biodegradation, and biological properties was used as a potential cell-supportive scaffold for the repair of focal corneal wounds. The mechanical property of hydrogels can be readily modified (postcure shear modulus of between 0.3 and 22 kPa) by varying the ratio of acrylate to thiol groups, photointensity, and solid content, and the biodegradation times also varied with the change of solid content. More importantly, the generated hydrogels exhibited excellent cell viability in both cell seeding and cell encapsulation experiments. Furthermore, the hydrogels were found to be biocompatible with rabbit cornea and aided the regeneration of a new tissue under a focal corneal wound (exhibiting epithelial wound coverage in <3d), and ultraviolet irradiation did not have any obvious harmful effect on the cornea and posterior eye segment tissues. Along with their injectability and tunable mechanical properties, the photocurable thiol-acrylate hydrogels showed promise as corneal substitutes or substrates to construct a new corneal tissue.
Silicone Microemulsion Structures Are Maintained During Polymerization with Reactive Surfactants

Abstract: Bicontinuous microemulsions exhibit domain structures on the nanoscale (<20 nm). Normally, such fine details are lost during the conversion from a fluid microemulsion to solid elastomeric materials, as a consequence of interfacial destabilization via polymerization of either the oil phase or monomers in the aqueous phase. Very little is known about the polymerization of silicone microemulsions and the morphological changes that occur upon transition from a nanostructured liquid to a solid matrix. Silicone microemulsions polymerized by free radical (aqueous phase) and condensation (silicone phase) processes, respectively, were characterized by small-angle X-ray scattering and transmission electron microscopy. It was found that cross-linking of the silicone phase alone led, over time, to large increase of the size of the microemulsion nanodomains. By contrast, photoinduced polymerization of a reactive surfactant and acrylic monomers in the aqueous phase was effective at retaining bicontinuous nanomorphology, irrespective of the degree of cross-linking of the silicone phase.

Abstract: 1-Methyl-3-phenyl-4,5-dihydro-1,2,4-triazin-6(1H)-one (1) undergoes aerial oxidation to give a mixture of 1- methyl-3-phenyl-1,2,4-triazin-6(1H)-one (2) and 1-methyl-3-phenyl-1,4-dihydro-1,2,4-triazine-5,6-dione (3). The dehydro derivative (2) was cleanly prepared by the oxidation of (1) with 2,3-dichloro-5,6-dicyano-1,4- benzoquinone (ddq). The dehydro derivative (2) underwent a surprising rearrangement to the triazole (12) upon oxidation with OxoneR. Several attempts at unambiguous synthesis of the α-dicarbonyl derivative (3) were unsuccessful it was obtained, together with the 1,4-dimethyl derivative (13) by methylation of 3-phenyl-1,4- dihydro-1,2,4-triazine-5,6-dione (4) with sodium hydride and methyl iodide.

Influence of Polymer Elasticity on the Formation of Non‐Cracking Honeycomb Films

Abstract: Non-planar non-cracking honeycomb (HC) structures are prepared from star polymers with high glass transition temperatures (T(g) ) and relatively low Young's moduli (E). This study demonstrates that the Young's modulus of a polymer is a more important factor than the glass transition temperature for determining the occurrence of cracking during HC film formation on non-planar surfaces.
Preparation of monomeric and polymeric β-cyclodextrin functionalized monoliths for rapid isolation and purification of puerarin from Radix puerariae

Abstract: Monomeric and epichlorohydrin polymerized β‐CD functionalized monoliths were prepared for the rapid isolation and purification of the isoflavonoid puerarin, a well‐known traditional Chinese drug, from a crude extract of Radix puerariae (root of the plant Pueraria lobata ). Two copolymers poly(isocyanatoethyl methacrylate‐ co ‐methyl methacrylate‐ co ‐ethylene dimethacrylate) (poly(IEM‐ co ‐MMA‐ co ‐EDMA)) and poly(glycidyl methacrylate‐ co ‐EDMA) (poly(GMA‐ co ‐EDMA)) were developed as facile, highly reactive and versatile monolithic matrix. SEM characterization demonstrated that the modified monoliths had homogenous porous structure and morphology. The success of the chemical modification of the monolithic matrix was confirmed by Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), solid‐state 13 C NMR and elemental analysis. It was demonstrated that polymeric β‐CD modified monoliths had better separation and selectivity for puerarin, recovering puerarin with a purity of 96% (m%) and a yield of 93% (m%). Compared with poly(glycidyl methacrylate‐ co ‐EDMA), poly(isocyanatoethyl methacrylate‐ co ‐methyl methacrylate‐ co ‐EDMA) monolithic matrix had higher reactivity, which significantly improved the β‐CD ligand density and thus the selectivity of the monoliths. Puerarin with a purity of 96% (m%) and with a yield of 89% (m%) was recovered on the monolith.

Correlation between the Level of Physical Activity as Measured by Accelerometer and the Harris Hip Score

Abstract: The most widely used method to assess the outcome of total hip arthroplasty (THA) is the Harris Hip Score (HHS). Patients’ expectations about the benefits of hip arthroplasty are increasing and are no longer limited to pain reduction. Patients believe they will be able to do recreational activity and sport after surgery. It is also essential to assess the level of physical activity after arthroplasty insofar as it is associated with early failure of the components. The purpose of this study is to explore correlation of the HHS with physical activity in patients with THA. This is a cross-sectional study on 47 patients with THA. Correlation of HHS with the results of physical activity obtained objectively using accelerometer worn for a week and subjectively using the IPAQ questionnaire and the UCLA scale. There was no correlation between the HHS and the activity measured using accelerometers, or with the IPAQ activity questionnaire. The HHS reported moderately significant correlations with the UCLA scale. The HHS may not be as discriminatory as other instruments at assessing patient activity levels after THA.

Encapsulated pancreatic progenitors derived from human embryonic stem cells as a therapy for insulin‐dependent diabetes

Abstract: Cellular-based therapies for insulin-dependent diabetes are potential means of achieving and maintaining normal blood glucose levels (BGL) without the need for insulin administration. Islets isolated from donor pancreases have been the most common tissue used to date, but supply is a limiting factor. The use of human embryonic stem cells (hESC) as a therapy became a possibility with the report that these cells could be differentiated to pancreatic progenitors (PP) over 12 days in vitro. Conversion of PP to glucose-responsive insulin-secreting cells can be achieved by transplanting the progenitors in vivo where cell maturation occurs. To date this step has not been shown under in vitro conditions. Prior to transplanting, cells are encapsulated in alginate to prevent the immune cells of recipient attacking the graft. The alginate capsules have pores with a molecular weight cut-off of 250 kDa. These are too small to allow entry of immune cells, but large enough for passage of nutrients and insulin. Encapsulated insulin-producing cells survive and function when transplanted, and have been shown to normalize BGL when allografted into diabetic mice. As few as 750 encapsulated human islets are sufficient to normalize BGL of diabetic non-obese diabetic severe combined immunodeficient (NOD/SCID) recipient mice for at least 2 months. The safety of transplanting encapsulated human islets as demonstrated by the lack of major adverse events and infection was recently shown in a first-in-human clinical trial. Finally, fetal porcine islet-like cell clusters, which are akin to PP derived from ESC, mature and normalize BGL of diabetic recipient mice with the same efficiency as non-encapsulated clusters placed under the kidney capsule. Transplanting encapsulated PP, derived from hESCs, into diabetic recipients is the strategy that is now being explored in the Australia Diabetes Therapy Project.
Preparation of hydrogels via ultrasonic polymerization

Abstract: Several acrylic hydrogels were prepared via ultrasonic polymerization of water soluble monomers and macromonomers. Ultrasound was used to create initiating radicals in viscous aqueous monomer solutions using the additives glycerol, sorbitol or glucose in an open system at 37 degrees C. The water soluble additives were essential for the hydrogel production, glycerol being the most effective. Hydrogels were prepared from the monomers 2-hydroxyethyl methacrylate, poly(ethylene glycol) dimethacrylate, dextran methacrylate, acrylic acid/ethylene glycol dimethacrylate and acrylamide/bis-acrylamide. For ex le a 5% w/w solution of dextran methacrylate formed a hydrogel in 6.5min in a 70% w/w solution of glycerol in water at 37 degrees C with 20kHz ultrasound, 56Wcm(-2). The ultrasonic polymerization method described here has a wide range of applications such a biomaterial synthesis where initiators are not desired.
The development of photochemically crosslinked native fibrinogen as a rapidly formed and mechanically strong surgical tissue sealant

Abstract: We recently reported the generation of a highly elastic, crosslinked protein biomaterial via a rapid photochemical process using visible light illumination. In light of these findings, we predicted that other unmodified, tyrosine-rich, self-associating proteins might also be susceptible to this covalent crosslinking method. Here we show that unmodified native fibrinogen can also be photochemically crosslinked into an elastic hydrogel biomaterial through the rapid formation of intermolecular dityrosine. Photochemically crosslinked fibrinogen forms tissue sealant bonds at least 5-fold stronger than commercial fibrin glue and is capable of producing maximum bond strength within 20s. In vitro studies showed that components of the photochemical crosslinking reaction are non-toxic to cells. This material will find useful application in various surgical procedures where rapid curing for high strength tissue sealing is required.
