Abstract: Plasmid DNA molecules with unique loop structures have widespread bioapplications, in many cases relying heavily on delivery vehicles to introduce them into cells and achieve their functions. Herein, we demonstrate that control over delicate nanotopography of silica nanoparticles as plasmid DNA vectors has significant impact on the transfection efficacy. For silica nanoparticles with rambutan-, raspberry-, and flower-like morphologies composed of spike-, hemisphere-, and bowl-type subunit nanotopographies, respectively, the rambutan-like nanoparticles with spiky surfaces demonstrate the highest plasmid DNA binding capability and transfection efficacy of 88%, higher than those reported for silica-based nanovectors. Moreover, it is shown that the surface spikes of rambutan nanoparticles provide a continuous open space to bind DNA chains via multivalent interactions and protect the gene molecules sheltered in the spiky layer against nuclease degradation, exhibiting no significant transfection decay. This unique protection feature is in great contrast to a commercial transfection agent with similar transfection performance but poor protection capability against enzymatic cleavage. Our study provides new understandings in the rational design of nonviral vectors for efficient gene delivery.

Abstract: The cooperative self-assembly of organic–inorganic siliceous composite structures has been studied from the aspect of inorganic precursors. We reveal that the vesicular or mesostructured materials can be obtained selectively by just changing the silica sources in one templating system. For poly(ethylene oxide)-type block copolymers with either poly(propylene oxide) or poly(butylene oxide) as the hydrophobic moieties, when the other synthesis parameters are exactly the same, the use of tetramethyl orthosilicate (TMOS) as a silica source gives rise to highly ordered mesostructures, while the use of tetraethyl orthosilicate (TEOS) leads to vesicles or foams. The attenuated total reflection Fourier transform infrared (ATR-FTIR) technique is used to monitor the silicate species derived from the hydrolysis and condensation of TMOS and TEOS as a function of the reaction time. On the basis of the ATR-FTIR results, we propose a “differentiating effect” at relatively high pH (4.7) to interpret the influence of different silica sources on the self-organized composite structures. For comparison, a “leveling effect” at relatively low pH (strong acidic conditions) is revealed to explain that both TMOS and TEOS lead to the same mesostructures. Our contribution provides a feasible and designable method to synthesize from conventional ordered mesostructures to novel vesicular structures, which are significant for their future practical applications.

Abstract: Zeolitic imidazolate framework-8 (ZIF-8) and its composites have erse applications. However, ZIF-8-based nanocomposites are mainly used as carriers in biomolecular delivery, with the functions of metal ions and ligands rarely used to modulate the biofunctions. In this work, dendritic mesoporous organosilica nanoparticles (DMONs) with tetrasulfide bond were used to confine ZIF-8 growth partially inside mesopores as a novel nanocomposite for mRNA delivery. Each component in the resultant DMONs-ZIF-8 contributed to mRNA delivery applications, including high loading benefitting from positively charged ZIF-8 and large mesopores of DMONs, endosomal escape promoted by the imidazole ring of ZIF-8, and long-term glutathione depletion mediated by both zinc ions and tetrasulfide bond. Combined together, DMONs-ZIF-8 demonstrated enhanced mRNA translation and better transfection efficiency than commercial products and toxic polymer-modified DMONs in vitro and in vivo.

Abstract: Accumulating evidence supports the concept that cancer stem cells (CSCs) are responsible for the tumor recurrence and metastasis, the two major causes of cancer-related death. Therefore, CSC-targeted cancer therapy is important for the future development of more effective and advanced cancer therapy. One of the approaches is to specifically silence oncogene expression in CSCs and inhibit their growth. The significance of this approach is its specificity and ability to avoid multi-drug resistance of CSCs. Hierarchical Porous Nitrogen‐Doped Spray‐Dried Graphene for High Performance Capacitive Deionization

Abstract: Graphene foam materials have attracted particular attention for capacitive deionization (CDI) applications due to their robust conductive framework and open three‐dimensional porous structures. However, their salt adsorption capacities (SACs) are limited by the bland macropores and poor compatibility with aqueous environments. Herein, hierarchical porous nitrogen‐doped spray‐dried graphene (N‐SDG) is prepared through a spray‐drying method followed by low‐temperature nitrogen doping. The resulting material exhibits hierarchical porosity and a nitrogen‐rich carbon framework which demonstrates an improved SAC of 19.6 mg g −1 in 500 mg L −1 NaCl solution, amongst the highest reported for graphene‐based CDI materials. The method herein demonstrates a commercially attractive approach for the production of N‐doped porous graphene material for CDI applications. These findings are expected to underpin new developments in low‐cost graphene‐based materials for a range of applications.

Low-cost and large-scale synthesis of functional porous materials for phosphate removal with high performance

Abstract: A facile spray drying technique has been developed for large-scale and template-free production of nanoporous silica with controlled morphology, large pore size, and high pore volume, using commercially available fumed silica, Aerosil 200, as a sole precursor. This approach can be applied to the preparation of functional nanoporous materials, in this study, lanthanum oxide functionalised silica microspheres by introducing lanthanum nitrate in situ during the spray drying process and followed by a post-calcination process. The resultant lanthanum functionalised Aerosil microspheres manifest high phosphate adsorption capacity (up to 2.317 mmol g(-1)), fast kinetics, and excellent adsorption performance at a low phosphate concentration (1 mg L(-1)). In virtue of the easy and scalable synthesis method, low cost and high performances of the product, the materials we reported here are promising for water treatment. Our approach may be general and extended to the synthesis of other functional nanoporous materials with versatile applications.

One-pot synthesis of hierarchical FeZSM-5 zeolites from natural aluminosilicates for selective catalytic reduction of NO by NH3

Abstract: Iron-modified ZSM-5 zeolites (FeZSM-5s) have been considered to be a promising catalyst system to reduce nitrogen oxide emissions, one of the most important global environmental issues, but their synthesis faces enormous economic and environmental challenges. Herein we report a cheap and green strategy to fabricate hierarchical FeZSM-5 zeolites from natural aluminosilicate minerals via a nanoscale depolymerization-reorganization method. Our strategy is featured by neither using any aluminum-, silicon-, or iron-containing inorganic chemical nor involving any mesoscale template and any post-synthetic modification. Compared with the conventional FeZSM-5 synthesized from inorganic chemicals with the similar Fe content, the resulting hierarchical FeZSM-5 with highly-dispersed iron species showed superior catalytic activity in the selective catalytic reduction of NO by NH 3 .

Comparison of Two Commercial ELISA Kits for the Detection of Anti-Dengue IgM for Routine Dengue Diagnosis in Laos

Abstract: The endemicity of Dengue virus (DENV) infection remains a major public health problem in Lao PDR. In this study, we compared two commercial anti-dengue IgM ELISA kits, Panbio® Dengue IgM Capture ELISA (Panbio Kit, Alere, Waltham, MA, USA) and DEN DetectTM MAC-ELISA (InBios kit, InBios International, Inc., Seattle, WA, USA), in the context of diagnosis of patients admitted to hospital with clinical dengue presentation. Two panels of paired blood s les were tested. Panel A was composed of 54 dengue confirmed patients (by DENV real-time RT-PCR) and 11 non-dengue dengue patients (other infections confirmed by corresponding PCR results). Panel B included 74 patients randomly selected from consecutive patients admitted to Mahosot Hospital in 2008 with suspicion of dengue fever according to WHO criteria. Results from panel A showed significantly better sensitivity for Panbio kit (64.8% 95%CI: 50.6–77.3%) than for InBios kit (18.5% 95%CI: 9.3–31.4%) when testing admission sera. Sensitivity was increased for both kits when combining results from admission and convalescent sera. Concordant results were obtained from panel B with fair agreement (κ = 0.29) between both kits when testing single admission s les, and moderate agreement (κ = 0.5) when combining results from admission and convalescent sera.

Silica nanopollens enhance adhesion for long-term bacterial inhibition

Abstract: Nature's creations with spiky topological features typically exhibit intriguing surface adhesive properties. From micrometer-sized pollen grains that can easily stick to hairy insects for pollination to nanoscale virus particles that are highly infectious toward host cells, multivalent interactions are formed taking advantage of rough surfaces. Herein, this nature-inspired concept is employed to develop novel drug delivery nanocarriers for antimicrobial applications. A facile new approach is developed to fabricate silica nanopollens (mesoporous silica nanospheres with rough surfaces), which show enhanced adhesion toward bacteria surfaces compared to their counterparts with smooth surfaces. Lysozyme, a natural antimicrobial enzyme, is loaded into silica nanopollens and shows sustained release behavior, potent antimicrobial activity, and long-term total bacterial inhibition up to 3 days toward Escherichia coli. The potent antibacterial activity of lysozyme-loaded silica nanopollens is further demonstrated ex vivo by using a small-intestine infection model. Our strategy provides a novel pathway in the rational design of nanocarriers for efficient drug delivery.

Synergistic Effect of Two Nanotechnologies Enhances the Protective Capacity of the Theileria parva Sporozoite p67C Antigen in Cattle

Abstract: East Coast fever (ECF), caused by Theileria parva, is the most important tick-borne disease of cattle in sub-Saharan Africa. Practical disadvantages associated with the currently used live-parasite vaccine could be overcome by subunit vaccines. An 80-aa polypeptide derived from the C-terminal portion of p67, a sporozoite surface Ag and target of neutralizing Abs, was the focus of the efforts on subunit vaccines against ECF and subjected to several vaccine trials with very promising results. However, the vaccination regimen was far from optimized, involving three inoculations of 450 μg of soluble p67C (s-p67C) Ag formulated in the Seppic adjuvant Montanide ISA 206 VG. Hence, an improved formulation of this polypeptide Ag is needed. In this study, we report on two nanotechnologies that enhance the bovine immune responses to p67C. In idually, HBcAg-p67C (chimeric hepatitis B core Ag virus-like particles displaying p67C) and silica vesicle (SV)–p67C (s-p67C adsorbed to SV-140-C18, octadecyl-modified SVs) adjuvanted with ISA 206 VG primed strong Ab and T cell responses to p67C in cattle, respectively. Coimmunization of cattle (Bos taurus) with HBcAg-p67C and SV-p67C resulted in stimulation of both high Ab titers and CD4 T cell response to p67C, leading to the highest subunit vaccine efficacy we have achieved to date with the p67C immunogen. These results offer the much-needed research depth on the innovative platforms for developing effective novel protein-based bovine vaccines to further the advancement.

Glutathione-depletion mesoporous organosilica nanoparticles as a self-adjuvant and Co-delivery platform for enhanced cancer immunotherapy

Abstract: Silica based nanoparticles have emerged as a promising vaccine delivery system for cancer immunotherapy, but their bio-degradability, adjuvanticity and the resultant antitumor activity remain to be largely improved. In this study, we report biodegradable glutathione-depletion dendritic mesoporous organosilica nanoparticles (GDMON) with a tetrasulfide-incorporated framework as a novel co-delivery platform in cancer immunotherapy. Functionalized GDMON are capable of co-delivering an antigen protein (ovalbumin) and a toll-like receptor 9 (TLR9) agonist into antigen presenting cells (APCs) and inducing endosome escape. Moreover, decreasing the intracellular glutathione (GSH) level through the -S-S-/GSH redox chemistry increases the ROS generation level both in vitro and in vivo, facilitating cytotoxic T lymphocyte (CTL) proliferation and reducing tumour growth in an aggressive B16-OVA melanoma tumour model. Our results have shown the potential of GDMON as a novel self-adjuvant and co-delivery nanocarrier for cancer vaccine.

Effect of Surface Functionality of Silica Nanoparticles on Cellular Uptake and Cytotoxicity

Abstract: Mesoporous silica nanoparticles (MCM-41) with different surface chemistry were used as carrier system to study its influence on drug delivery and anticancer activity of curcumin (CUR). CUR was encapsulated in pristine MCM-41 (hydrophilic and negatively charged), amino functionalized MCM-41 (MCM-41-NH2 which is hydrophilic and positively charged), and methyl functionalized MCM-41 (MCM-41-CH3 which is hydrophobic and negatively charged) and evaluated for in vitro release and cell cytotoxicity in human squamous cell carcinoma cell line (SCC25). Various techniques were employed to evaluate the performance of these materials on cellular uptake and anticancer activity in the SCC25 cell line. Both positively and negatively charged surfaces demonstrated enhanced drug release and anticancer activity compared to pure CUR. Positively charged nanoparticles showed higher cell uptake compared to negatively charged nanoparticles owing to its electrostatic interaction with cells. However, hydrophobic surface modified nanoparticles (MCM-41-CH3) showed no improvement in drug release and anticancer activity due to its poor wetting effect. Cell cycle analysis and cell apoptosis studies revealed different pathway mechanisms followed by the positively and negatively charged nanoparticles but exhibiting similar anticancer activity in SCC25 cells.

Modulating in vitro release and solubility of griseofulvin using functionalized mesoporous silica nanoparticles

Abstract: Mesoporous silica nanoparticles (MCM-41) were used as a carrier system to study the influence of surface charge and hydrophobicity on solubility and in-vitro drug release behavior of Griseofulvin, a potent antifungal drug with low water solubility. Bare MCM-41 with a pure silica composition, MCM-41 after amino functionalization (MCM-41-NH2) and methyl functionalization (MCM-41-CH3) were used in this study followed by encapsulation of griseofulvin. Various characterization techniques have been employed to confirm the successful drug loading inside the nanopores. The surface functionalization on MCM-41 is found to have significant effect on griseofulvin's in vitro release and solubility. Both negatively and positively charged surface showed enhancement in solubility and drug release of griseofulvin. However, the hydrophobic modification led to a retarded drug release, which is caused by the poor wetting effect in the case of MCM-41-CH3 nanoparticles.

Periodic Mesoporous Organosilicas with Controlled Pore Symmetries for Peptides Enrichment

Abstract: Periodic mesoporous organosilicas (PMOs) with controlled structures have been synthesized by using cetyltrimethylammonium bromide (CTAB) and sodium perfluorooctanoate (PFONa) as co-templates, 1,2-bis (triethoxysilyl)ethane (BTEE) as an organosilica precursor. By increasing the weight ratio of PFONa/CTAB, a structure transformation from a cubic (Pm-3n) to a two-dimensional hexagonal (p6m) mesostructure and then to multilamellar vesicles can be observed. The cubic and hexagonal s les have similar particle size (200-750 nm), pore size (2.6 and 2.8 nm, respectively), total pore volume (approximately 0.7 cm3/g), and surface area (approximately 900 m2/g), providing ideal candidates to study the peptide enrichment performance influenced simply by pore symmetries. Matrix-assisted laser desorption ionization time-of-flight mass spectroscopy (MALDI-TOF MS) analysis indicates that PMO with a cubic (Pm-3n) structure is more effective in small molecular weight peptides enrichment compared with PMO with a hexagonal structure, showing the importance of mesostructural control for targeted applications. The phenomena can be attributed to the cage-type structure of the Pm-3n symmetry, which possesses cages with a relatively larger pore size and connectivity with a relatively smaller size. It is suggested that the pore entrances with small size are responsible for entrapping small molecular weight peptides. Our study may shed light on the designed synthesis of functional porous materials with controlled structures and enhanced performance in peptides enrichment.

Antibiotic‐Free Antibacterial Strategies Enabled by Nanomaterials: Progress and Perspectives

Abstract: Bacterial infection is one of the top ten leading causes of death globally and the worst killer in low-income countries. The overuse of antibiotics leads to ever-increasing antibiotic resistance, posing a severe threat to human health. Recent advances in nanotechnology provide new opportunities to address the challenges in bacterial infection by killing germs without using antibiotics. Antibiotic-free antibacterial strategies enabled by advanced nanomaterials are presented. Nanomaterials are classified on the basis of their mode of action: nanomaterials with intrinsic or light-mediated bactericidal properties and others that serve as vehicles for the delivery of natural antibacterial compounds. Specific attention is given to antibacterial mechanisms and the structure-performance relationship. Practical antibacterial applications employing these antibiotic-free strategies are also introduced. Current challenges in this field and future perspectives are presented to stimulate new technologies and their translation to fight against bacterial infection.

Abstract: Anisotropy plays a unique role in the structural regulation of metal–organic frameworks (MOFs) and their composites, especially at the micro‐ and nanoscale. However, there is a lack of a understanding of MOF micro/nanoparticles (MNPs) from the perspective of morphological anisotropy. In this Minireview, recent advances in anisotropic MOF MNPs are summarized, with a focus on how morphological anisotropy leads to innovative structures and modulates properties. First, anisotropic pristine MOF MNPs with erse morphologies are introduced and classified by their morphology‐dependent and morphology‐independent anisotropy. Secondly, the anisotropy‐enabled site‐selective higher‐order construction of MOF‐based materials is highlighted. Finally, challenges and prospects for anisotropic MOFs are discussed, aiming to provide inspiration for further developments in this interesting research field.

Abstract: The packing structures of macroporous ordered siliceous foams (MOSFs) are systematically investigated by using a 3D electron tomography technique and the nanostructural characteristics for layered MOSFs are resolved. MOSF materials adopt an ordered 2D hexagonal arrangement in single-layered areas, regular honeycomb patterns in double-layered s les, and polyhedric cells similar to a Weaire-Phelan structure in multilayered areas, all following the principle of minimizing surface area, which is well understood in soap foams at the macroscopic scale. In surfactant-templated materials, liquid-crystal templating is generally applied, but here it is revealed that the surface-area-minimization principle can also be applied, which facilitates the design and synthesis of novel macroporous materials using surfactant molecules as templates.

Abstract: Innovations in nanofabrication have expedited advances in hollow-structured nanomaterials with increasing complexity, which, at the same time, set challenges for the precise determination of their intriguing and complicated 3D configurations. Conventional transmission electron microscopy (TEM) analysis typically yields 2D projections of 3D objects, which in some cases is insufficient to reflect the genuine architectures of these 3D nano-objects, providing misleading information. Advanced analytical approaches such as focused ion beam (FIB) and ultramicrotomy enable the real slicing of nanomaterials, realizing the direct observation of inner structures but with limited spatial discrimination. Electron tomography (ET) is a technique that retrieves spatial information from a series of 2D electron projections at different tilt angles. As a unique and powerful tool kit, this technique has experienced great advances in its application in materials science, resolving the intricate 3D nanostructures. Here, the exceptional capability of the ET technique in the structural, chemical, and quantitative analysis of hollow-structured nanomaterials is discussed in detail. The distinct information derived from ET analysis is highlighted and compared with conventional analysis methods. Along with the advances in microscopy technologies, the state-of-the-art ET technique offers great opportunities and promise in the development of hollow nanomaterials.

Mesoporous silica nanoparticles as antigen carriers and adjuvants for vaccine delivery

Abstract: Vaccines have been at the forefront of improving human health for over two centuries. The challenges faced in developing effective vaccines flow from complexities associated with the immune system and requirement of an efficient and safe adjuvant to induce a strong adaptive immune response. Development of an efficient vaccine formulation requires careful selection of a potent antigen, efficient adjuvant and route of delivery. Adjuvants are immunological agents that activate the antigen presenting cells (APCs) and elicit a strong immune response. In the past decade, the use of mesoporous silica nanoparticles (MSNs) has gained significant attention as potential delivery vehicles for various biomolecules. In this review, we aim to highlight the potential of MSNs as vaccine delivery vehicles and their ability to act as adjuvants. We have provided an overview on the latest progress on synthesis, adsorption and release kinetics and biocompatibility of MSNs as next generation antigen carriers and adjuvants. A comprehensive summary on the ability of MSNs to deliver antigens and elicit both humoral and cellular immune responses is provided. Finally, we give insight on fundamental challenges and some future prospects of these nanoparticles as adjuvants.

Abstract: Accumulating evidence supports the concept that cancer stem cells (CSCs) are responsible for tumor initiation and maintenance. They are also considered as an attractive target for advanced cancer therapy. Using a sphere culture method that favors the growth of self-renewal cells, we have isolated sphere-forming cells (SFCs) from cervical cancer cell lines HeLa and SiHa. HeLa-SFCs were resistant to multiple chemotherapeutic drugs and were more tumorigenic, as evidenced by the growth of tumors following injection of immunodeficient mice with 1 × 10(4) cells, compared with 1 × 10(6) parental HeLa cells required to grow tumors of similar size in the same time frame. These cells showed an expression pattern of CD44(high)/CD24(low) that resembles the CSC surface biomarker of breast cancer. We further demonstrated that HeLa-SFCs expressed a higher level (6.9-fold) of the human papillomavirus oncogene E6, compared with that of parental HeLa cells. Gene silencing of E6 with a lentiviral-short-hairpin RNA (shRNA) profoundly inhibited HeLa-SFC sphere formation and cell growth. The inhibition of cell growth was even greater than that for sphere formation after E6 silence, suggesting that the loss of self-renewing ability may be more important. We then measured the expression of self-renewal genes, transformation growth factor-beta (TGF-β) and leukemia-inhibitory factor (LIF), in shRNA-transduced HeLa-SFCs and found that expression of all three TGF-β isoforms was significantly downregulated while LIF remained unchanged. Expression of the Ras gene (a downstream component of TGF-β) was also markedly decreased, suggesting that the growth-inhibitory effect could be via the TGF-β pathway. The above data indicate RNA interference-based therapy may offer a new approach for CSC-targeted cancer therapy.

A Smart Glycol‐Directed Nanodevice from Rationally Designed Macroporous Materials

Abstract: We have developed a smart nanodevice for the highly efficient and selective detection of glycoproteins. This polyfunctional device is fabricated through the rational functionalization of macroporous silica foam (MOSF) materials with a boron species (B-MOSF) and amino groups (NH(2)-MOSF), and then the integration of MOSF, B-MOSF and NH(2)-MOSF materials. In such a device, a macroporous structure with very large-pore sizes (diameters approximately 100 nm) and high-pore volumes (>0.65 cm(3) g(-1)) is advantageous to efficiently fasten the enzymatic reaction. The targeted specific glycopeptides of the products can be selectively isolated and enriched in B-MOSF through the chemo-affinity between boronic acid and glycol groups, while the non-specific peptides are released to the solutions, or further purified by MOSF and NH(2)-MOSF, which have opposite charges. As a result, the protein digestion and glycol-peptide isolation can be simultaneously achieved in the functionalized macroporous materials in one step, which is a great advantage compared to conventional multi-procedure and time-consuming techniques.

Abstract: Effective messenger RNA (mRNA) transfection in hard-to-transfect cells delivered by vectors is a long-standing challenge. Now it is hypothesized that the high intracellular glutathione level is associated with suppressed mRNA translation. This theory leads to a new design principle of next-generation mRNA vectors: nanoparticles with glutathione depletion chemistry upregulate mRNA translation and enhance transfection, which is beneficial for mRNA delivery in hard-to-transfect cells in vitro and in vivo.

Mesoporous silica nanocarriers encapsulated antimalarials with high therapeutic performance

Abstract: The use of nanocarriers in drug delivery is a breakeven research and has received a clarion call in biomedicine globally. Herein, two newly nano-biomaterials: MCM-41 encapsulated quinine (MCM-41 ⊃ QN) ( 1 ) and 3-phenylpropyl silane functionalized MCM-41 loaded QN (pMCM-41 ⊃ QN) ( 2 ) were synthesized and well characterized. 1 and 2 along with our two already reported nano-antimalarial drugs (MCM-41 ⊃ ATS) ( 3 ) and 3-aminopropyl silane functionalized MCM-41 contained ATS (aMCM-41 ⊃ ATS) ( 4 ) were screened in vitro for their activity against P. falciparium W2 strain, cytotoxicity against BGM cells and in vivo for their activity against Plasmodium bergheiNK65 . 1 has the highest antimalarial activity in vivo against P. berghei NK65, (ED 50 : 0.0625 mg/kg body weight) and higher mean survival time compared to the other nano biomaterials or unencapsulated drugs at doses higher than 0.0625 mg/kg body weight. This encapsulation strategy of MCM-41 ⊃ QN ( 1 ) stands very useful and effective in delivering the drug to the target cells compared to other delivery systems and therefore, this encapsulated drug may be considered for rational drug design.

Abstract: In this paper, a targeted drug delivery system has been developed based on hyaluronic acid (HA) modified mesoporous silica nanoparticles (MSNs). HA-MSNs possess a specific affinity to CD44 over-expressed on the surface of a specific cancer cell line, HCT-116 (human colon cancer cells). The cellular uptake performance of fluorescently labelled MSNs with and without HA modification has been evaluated by confocal microscopy and fluorescence-activated cell sorter (FACS) analysis. Compared to bare MSNs, HA-MSNs exhibit a higher cellular uptake via HA receptor mediated endocytosis. An anticancer drug, doxorubicin hydrochloride (Dox), has been loaded into MSNs and HA-MSNs as drug delivery vehicles. Dox loaded HA-MSNs show greater cytotoxicity to HCT-116 cells than free Dox and Dox-MSNs due to the enhanced cell internalization behavior of HA-MSNs. It is expected that HA-MSNs have a great potential in targeted delivery of anticancer drugs to CD44 over-expressing tumors.

Harnessing bioactive nanomaterials in modulating tumor glycolysis-associated metabolism

Abstract: Glycolytic reprogramming is emerging as a hallmark of various cancers and a promising therapeutic target. Nanotechnology is revolutionizing the anti-tumor therapeutic approaches associated with glycolysis. Finely controlled chemical composition and nanostructure provide nanomaterials unique advantages, enabling an excellent platform for integrated drug delivery, biochemical modulation and combination therapy. Recent studies have shown promising potential of nanotherapeutic strategies in modulating tumor glycolytic metabolism alone or in combination with other treatments such as chemotherapy, radiotherapy and immunotherapy. To foster more innovation in this cutting-edge and interdisciplinary field, this review summarizes recent understandings of the origin and development of tumor glycolysis, then provides the latest advances in how nanomaterials modulate tumor glycolysis-related metabolism. The interplay of nanochemistry, metabolism and immunity is highlighted. Ultimately, the challenges and opportunities are presented.

Advances in silica based nanoparticles for targeted cancer therapy

Abstract: Targeted delivery of anticancer drug specifically to tumor site without damaging normal tissues has been the dream of all scientists fighting against cancer for decades. Recent breakthrough on nanotechnology based medicines has provided a possible tool to solve this puzzle. Among erse nanomaterials that are under development and extensive study, silica based nanoparticles with vast advantages have attracted great attention. In this review, we concentrate on the recent progress using silica based nanoparticles, particularly mesoporous silica nanoparticles (MSNs), for targeted drug delivery applications. First, we discuss the passive targeting capability of silica based nanoparticles in relation to their physiochemical properties. Then, we focus on the recent advances of active targeting strategies involving tumor cell targeting, vascular targeting, nuclear targeting and multistage targeting, followed by an introduction to magnetic field directed targeting approach. We conclude with our personal perspectives on the remaining challenges and the possible future directions. Chemotherapy has been one of the mainstays of cancer treatment. The advances in nanotechnology has allowed the development of novel carrier systems for the delivery of anticancer drugs. Mesoporous silica has shown great promise in this respect. In this review article, the authors provided a comprehensive overview of the use of this nanoparticle in both passive, as well as active targeting in the field of oncology. The advantages of this particle were further discussed.

New Insight into Ordered Cage-Type Mesostructures and Their Pore Size Determination by Electron Tomography

Abstract: In this work, a new approach based on electron tomography (ET) has been developed to measure the pore size, through which new insight into cage-type ordered mesostructures and their pore size determination has been obtained. It is demonstrated that the accurate pore diameter, especially for cage-type cubic mesoporous materials, can be determined only through our ET approach by considering that the pore geometry is a real 3D space. We use the established ET method to revisit the applicability of different models for the pore size calculation in nitrogen adsorption analysis. Different from the overwhelming understanding that the nonlocal density functional theory (NLDFT) and Derjaguin-Broekhoff-de Boer (BdB) model are recommended to calculate the pore size of cage-type cubic mesoporous materials while the Barret-Joyner-Halenda (BJH) model should not be used, a new understanding is gained through this study. The choice of a suitable model for pore size determination depends on the precise pore structure. For a cage-type cubic mesoporous material with fcc symmetry and a large entrance connecting the cages, the BJH model is more accurate while the other two methods overestimate the pore size (by up to 40%). The DFT model is more appropriate when the pore shape is a perfect sphere than the BJH model, which underestimates the pore size, and the BdB model, which overestimates the pore size. It is our opinion that the unique ET approach should be used to revisit a vast number of large-pore cubic mesoporous materials to provide genuine structural information.

Asymmetric Silica Nanoparticles with Tunable Head–Tail Structures Enhance Hemocompatibility and Maturation of Immune Cells

Abstract: Asymmetric mesoporous silica nanoparticles (MSNs) with controllable head-tail structures have been successfully synthesized. The head particle type is tunable (solid or porous), and the tail has dendritic large pores. The tail length and tail coverage on head particles are adjustable. Compared to spherical silica nanoparticles with a solid structure (Stöber spheres) or large-pore symmetrical MSNs with fully covered tails, asymmetrical head-tail MSNs (HTMSNs) show superior hemocompatibility due to reduced membrane deformation of red blood cells and decreased level of reactive oxygen species. Moreover, compared to Stöber spheres, asymmetrical HTMSNs exhibit a higher level of uptake and in vitro maturation of immune cells including dendritic cells and macrophage. This study has provided a new family of nanocarriers with potential applications in vaccine development and immunotherapy.

Submicron-sized vermiculite assisted oregano oil for controlled release and long-term bacterial inhibition

Abstract: Oregano essential oil (OEO) is a natural compound consisting of potent antibiotic molecules. Its volatility is the major obstacle against the transportation and anti-bacterial performance. In this work, submicron-sized vermiculite (SMV) particles were prepared from Australian vermiculite clay by ball milling, and tested as a potential particulate-carrier for OEO. The loading of OEO by SMV can be easily achieved by mechanical mixing. Compared to raw vermiculite and free OEO, the OEO-loaded SMV displayed sustained isothermal release behaviour of OEO and demonstrated enhanced antibacterial performance in in vitro antibacterial tests against Escherichia coli (E. coli) and Staphylococcus epidermidis (S. epidermidis). This study provides a facile and commercially viable approach in designing advantageous carriers for volatile actives in antimicrobial applications.

Metal‐Organic Frameworks Derived Titanium Oxides via Soft Interface Adaptive Transformation

Abstract: High‐temperature treatment of metal‐organic frameworks (MOFs) is a promising route to prepare a variety of functional materials with erse applications. However, it is challenging to prepare highly crystalline metal oxides such as TiO 2 with controllable structures and morphologies via direct calcination of MOFs. Here, we report a soft interface adaptive transformation strategy for the synthesis of MOFs derived TiO 2 nanoparticles with elaborate structures. The soft interface is constructed by coating a thin silica layer on the external surface of Ti‐MOFs with an octahedral morphology. This soft interface can partially adapt to the significant shrinkage and counteract the contraction force during the MOFs‐to‐TiO 2 conversion. As a result, TiO 2 hollow octahedrons are formed with a concave morphology, high specific surface area, abundant oxygen vacancy sites, and consequently enhanced photocatalytic activity, which cannot be obtained by direct calcination or coating a thick/rigid silica layer. The opposite impact of the interface on MOFs derived metal oxides and carbon materials is further elucidated. This contribution provides new understandings in the synthesis of MOFs derived functional materials.

Abstract: Characterization and application of graphene sheets modified glassy carbon electrodes (graphene/GC) have been presented for the electrochemical bio-sensing. A probe molecule, potassium ferricyanide is employed to study the electrochemical response at the graphene/GC electrode, which shows better electron transfer than graphite modified (graphite/GC) and bare glassy carbon (GC) electrodes. Based on the highly enhanced electrochemical activity of NADH, alcohol dehydrogenase (ADH) is immobilized on the graphene modified electrode and displays a more desirable analytical performance in the detection of ethanol, compared with graphite/GC or GC based bio-electrodes. It also exhibits good performance of ethanol detection in the real s les. From the results of electrochemical investigation, graphene sheets with a favorable electrochemical activity could be an advanced carbon electrode materials for the design of electrochemical sensors and biosensors.

Prevalence and Molecular Characterization of Rickettsia spp. from Wild Small Mammals in Public Parks and Urban Areas of Bangkok Metropolitan, Thailand

Abstract: Rural areas usually show a higher prevalence of rickettsial infection than urban areas. However, information on the rickettsial infection status in urban settings (e.g., built-up areas and city parks) is still limited, particularly in the Bangkok metropolitan area. In this study, we performed a molecular rickettsial survey of spleen s les of small mammals caught in public parks and built-up areas of Bangkok. Out of 198 s les, the Rattus rattus complex was found to be most prevalent. The lification of rickettsial gltA fragment gene (338 bp) by nested PCR assay revealed positive results in four s les, yielding a low prevalence of infection of 2.02%. DNA sequencing results confirmed that three s les were matched with Rickettsia typhi, and one was identified as R. felis. It is noteworthy that this is the first report of the occurrence of R. felis DNA in rodents in Southeast Asia.

Combination of Microporous Hollow Carbon Spheres and Nafion for the Individual Metal-free Stripping Detection of Pb2+ and Cd2+

Abstract: Here, the combination of Nafion with microporous hollow carbon spheres (MHCS) is first proposed to fabricate a disposable metal-free electrode for heavy metal stripping sensing. The MHCS-Nafion composite film electrode is prepared by drop-casting a mixture of MHCS and Nafion onto the lab-made screen-printed carbon electrode (SPCE(*)). Results demonstrate that the interfusion of MHCS into Nafion offers enhanced performance for the electro-enrichment and stripping of lead and cadmium over the only Nafion film: 1) abundant MHCS immobilized on the electrode surface serve as effective nucleation sites for metal ion reduction 2) the mixing of MHCS into Nafion enlarges the active surface of negative-charged Nafion for the electrostatic adsorption of metal cations. The proposed MHCS-Nafion/SPCE(*) provides linear responses for Pb(2+) and Cd(2+) in the range of 2 - 200 μg/L, with a detection limit of 1.37 and 1.63 μg/L, respectively. Practical applications of the sensor in water s le detection with good accuracy have also been confirmed.

Abstract: Fungi and bacteria afflict humans with innumerous pathogen-related infections and ailments. Most of the commonly employed microbicidal agents target commensal and pathogenic microorganisms without discrimination. To distinguish and fight the pathogenic species out of the microflora, novel antimicrobials have been developed that selectively target specific bacteria and fungi. The cell wall features and antimicrobial mechanisms that these microorganisms involved in are highlighted in the present review. This is followed by reviewing the design of antimicrobials that selectively combat a specific community of microbes including Gram-positive and Gram-negative bacterial strains as well as fungi. Finally, recent advances in the antimicrobial immunomodulation strategy that enables treating microorganism infections with high specificity are reviewed. These basic tenets will enable the avid reader to design novel approaches and compounds for antibacterial and antifungal applications.

Abstract: Since the extension of the Stöber method traditionally used in the synthesis of silica nanoparticles into the fabrication of resorcinol-formaldehyde nanospheres in 2011, significant progresses have been achieved in this exiting area of nano-resole-enabled synthesis of elegant nanostructures with versatile compositions and promising applications in various fields. To date, there are few reviews focused on this topic. In this minireview, we aim to provide an overview on recent developments, with the emphasis on nano-resoles as an enabling strategy in the synthesis of innovative materials. The history of nano-resoles and their distinct roles classified into four functions will be introduced. Clear understanding into this research field is vital for achieving rational design and controllable synthesis of a new generation of nano-resoles and their derived nanostructures.

Macroporous Materials as Novel Catalysts for Efficient and Controllable Proteolysis

Abstract: A novel nanopore based digestion strategy has been developed by directly adding a macroporous material as catalyst to the conventional in-solution reaction system. Without increasing the enzyme or protein concentrations, this simple digestion approach exhibits high proteolysis efficiency and selectivity due to the in situ fast adsorption of both enzymes and proteins from bulk solution into the macropores of the catalysts, where the target substrates and enzymes are greatly concentrated and confined in the nanospace to realize a quick digestion. Based on the electrostatic interaction matching between the biomolecules and catalysts, selective extraction and digestion of proteins with different isoelectric points can be achieved by adjusting the surface charge of the catalysts. This nanoporous reaction system has been successfully applied to the analysis of a complex biological s le, where 293 proteins are identified, while only 100 proteins are obtained by the standard overnight in-solution digestion. The present nanospace confined digestion strategy will lead to promising advances not only in proteomics but also in other applications where enzymatic reactions are involved.

Surfactant-Free Assembly of Mesoporous Carbon Hollow Spheres with Large Tunable Pore Sizes

Abstract: Mesoporous carbon hollow spheres (MCHS) have wide applications, including catalysis, absorption, and energy storage/conversion. Herein, we report a one-pot, surfactant-free synthesis of MCHS using three molecules: resorcinol, formaldehyde, and tetrapropyl orthosilicate. The co-condensation process between the in situ generated silica primary particles and the polymer oligomers is regulated, leading to monodispersed MCHS with adjustable pore sizes from micropores to 13.9 nm. The resultant MCHS shows excellent performance for electrochemical double-layer capacitors with high capacitance (310 F g(-1) at 1 A g(-1)), excellent rate capability (157 F g(-1) at 50 A g(-1)), and outstanding cycling stability (98.6% capacity retention after 10 000 cycles at 10 A g(-1)). Our one-pot synthesis strategy is versatile and can be extended to fabricate metal oxide@mesoporous carbon yolk-shell structures in the absence of surfactant, paving the way toward designed synthesis of nanostructured mesoporous carbon composites for various applications.

Functionalized hollow siliceous spheres for VOCs removal with high efficiency and stability

Abstract: Functionalized hollow siliceous spheres (HSSs) have been prepared by surface modification with trimethylchlorosilane (TMCS) for the removal of volatile organic compounds (VOCs). The resultant HSSs-TMCS possesses a uniform and well-dispersed hollow spherical structure, high surface area, large total pore volume, high VOCs adsorption capacity, and small water vapor adsorption capacity. The adsorption and desorption performance of HSSs-TMCS under static (n-hexane and 93# gasoline) and dynamic (n-hexane) conditions was investigated. Compared with commercial silica gel (SG) and activated carbon (AC), HSSs-TMCS show higher capacity of adsorbing n-hexane and 93# gasoline with good stability and low water vapor adsorption capacity under static adsorption conditions, higher dynamic adsorption capacity and stable breakthrough time under dynamic adsorption conditions. The high efficiency and stability of functionalized HSSs are associated with their unique hollow morphology and structure parameters. The designed HSSs-TMCS with high VOCs removal capacity and recyclability are promising candidates for the treatment of air pollution.

A Pacman‐Like Titanium‐Doped Cobalt Sulfide Hollow Superstructure for Electrocatalytic Oxygen Evolution

Abstract: Transition‐metal sulfides (TMSs) are attractive oxygen evolution reaction (OER) electrocatalysts. Developing new strategies to improve their electrochemical performance of TMSs is of great significance. Herein, a unique pacman‐like titanium‐doped cobalt sulfide hollow superstructure (Ti‐CoSx HSS) is fabricated as an OER electrocatalyst. Using a prearranged metal–organic framework (MOF)‐on‐MOF heterostructure as a precursor treated by one‐pot sulfidation, a sequential structural conversion process leads to the formation of Ti‐CoSx HSS, which is assembled by interconnected Ti‐doped CoSx nanocages around a cake‐like cavity. Benefiting from the architecture and compositional advantages, Ti‐CoSx HSS exhibits excellent OER performance with an overpotential of 249 mV at 10 mA cm −2 and Tafel slope of 45.5 mV dec −1 due to increased active site exposure, enhanced electron and mass transfer. This strategy enabled by MOF‐on‐MOF paves the way toward innovative MOF derivatives for various applications.

Sensitive Detection of Human Insulin Using a Designed Combined Pore Approach

Abstract: A unique combined pore approach to the sensitive detection of human insulin is developed. Through a systematic study to understand the impact of pore size and surface chemistry of nanoporous materials on their enrichment and purification performance, the advantages of selected porous materials are integrated to enhance detection sensitivity in a unified two-step process. In the first purification step, a rationally designed large pore material (ca. 100 nm in diameter) is chosen to repel the interferences from nontarget molecules. In the second enrichment step, a hydrophobically modified mesoporous material with a pore size of 5 nm is selected to enrich insulin molecules. A low detection limit of 0.05 ng mL(-1) in artificial urine is achieved by this advanced approach, similar to most antibody-based analysis protocols. This designer approach is efficient and low cost, and thus has great potential in the sensitive detection of biomolecules in complex biological systems.

Designer Anticancer Nanoprodrugs with Self‐Toxification Activity Realized by Acid‐triggered Biodegradation and In Situ Fragment Complexation

Abstract: Prodrugs that allow in situ chemical conversion of less toxic precursors into active drugs in response to certain stimuli are promising anticancer candidates. Herein, we present a novel design of nanoprodrugs with a “degradation‐mediated self‐toxification” strategy, which realizes intracellular synthesis of anticancer agents using the nanoparticles’ own degradation fragments as the precursors. To fulfill this concept, a metal complexing dicyclohexylphosphine (DCP) organosilane is carefully screened out from various ligands to conjugate onto Pd(OH) 2 nanodots confined hollow silica nanospheres (PD‐HSN). This constructed nanoprodrug shows acid‐triggered degradation in lysosomes and neutralizes protons to induce lysosomes rupturing, generating predesigned less toxic fragments (Pd 2+ and DCP‐silicates) that complex into DCP/Pd complex in situ for inducing DNA damage, leading to enhanced anticancer activity against various cancer cell lines as well as in a xenograft tumour model.

Abattoir-based serological surveillance for transboundary and zoonotic diseases in cattle and swine in Cambodia: a pilot study in Phnom Penh province during 2019 and 2020

Abstract: A pilot animal disease surveillance program was implemented at four abattoirs in Phnom Penh, Cambodia, between October 2019 and January 2020. A total of 1141 s les were collected from 477 cattle and 664 swine. Serological testing was performed using commercial antibody ELISA kits for zoonotic and high-impact animal diseases, namely brucellosis, Q fever, classical swine fever (CSF), porcine reproductive and respiratory syndrome (PRRS) and African swine fever (ASF). Only two s les tested positive for Brucella antibodies (0.2%, 95% CI 0.4–0.6, n = 1141). The seroprevalence of Q fever was 0.8% (95% CI 0.3–2.1, n = 477) in the cattle s les, while CSF, PRRS and ASF in pigs were 55.4% (95% CI 51.6–59.2, n = 655), 81.2% (95% CI 78.1–84.0, n = 655) and 2.6% (95% CI 1.6–4.1, n = 664), respectively. All 38 doubtful and 17 positive ASF antibody ELISA s les were negative when tested by real-time PCR. Univariate analyses demonstrated that the factor significantly associated with positive results of ASF was the abattoir location ( p -value = 0.002). Based on logistic regression models, significant risk factors for CSF were province of origin ( p -value = 1.7 × 10 −6 ), abattoir ( p -value = 3.6 × 10 −11 ) and PRRS positivity ( p -value = 0.004), and for PRRS were province of origin ( p -value = 0.0004) and CSF positivity ( p -value = 0.001). In conclusion, the seroprevalences of zoonotic diseases in this study were very low. The high prevalence of CSF and PRRS antibodies were most likely the result of vaccination. All ASF seropositive pigs, including those that gave equivocal results, originated from large-scale Cambodian-based commercial farms, as well as Thailand, which raises questions about possible illegal vaccination or low-pathogenicity ASF variants. The pilot abattoir serological surveillance program described here has the potential to provide a sentinel for incursions of novel and endemic pathogens, although further work is required to demonstrate its capacity to provide information on the longitudinal disease trends.

Understanding the interactions at the silica and PEI interface is important for successful DNA delivery and transfection, especially for silica with different surface functionality. Herein, we report that a higher content of hydrogen boning formed between PEI molecules and phosphonate modified silica nanoparticles could slow down the PEI dissolution from the freeze-dried solid composites into aqueous solution than the bare silica counterpart. The pronounced PEI retention ability through phosphonation of silica nanoparticles effectively improves the transfection efficiency due to the high DNA binding affinity extracellularly, effective lysosome escape and high nuclear entry of both PEI and DNA intracellularly. Our study provides a fundamental understanding on designing effective silica-PEI-based nano-vectors for DNA delivery applications.

Abstract: Recently, dendritic mesoporous silica nanoparticles with widespread applications have attracted great interest. Despite many publications ( ), the terminology “dendritic” is ambiguous. Understanding what possible “dendritic structures” are, their formation mechanisms and the underlying structure–property relationship is fundamentally important. With the advance of characterization techniques such as electron tomography, two types of tree‐branch‐like and flower‐like structures can be distinguished, both described as “dendritic” in the literature. In this Review, we start with the definition of “dendritic”, then provide critical analysis of reported dendritic silica nanoparticles according to their structural classification. We update the understandings of the formation mechanisms of two types of “dendritic” nanoparticles, highlighting how to control their structural parameters. Applications of dendritic mesoporous nanoparticles are also reviewed with a focus on the biomedical field, providing new insights into the structure–property relationship in this family of nanomaterials.

Abstract: Fiber shaped supercapacitors are promising candidates for wearable electronics because they are flexible and light-weight. However, a critical challenge of the widespread application of these energy storage devices is their low cell voltages and low energy densities, resulting in limited run-time of the electronics. Here, we demonstrate a 1.5 V high cell voltage and high volumetric energy density asymmetric fiber supercapacitor in aqueous electrolyte. The lightweight (0.24 g cm −3 ), highly conductive (39 S cm −1 ) and mechanically robust (221 MPa) graphene fibers were firstly fabricated and then coated by NiCo 2 S 4 nanoparticles (GF/NiCo 2 S 4 ) via the solvothermal deposition method. The GF/NiCo 2 S 4 display high volumetric capacitance up to 388 F cm −3 at 2 mV s −1 in a three-electrode cell and 300 F cm −3 at 175.7 mA cm −3 (568 mF cm −2 at 0.5 mA cm −2 ) in a two-electrode cell. The electrochemical characterizations show 1000% higher capacitance of the GF/NiCo 2 S 4 as compared to that of neat graphene fibers. The fabricated device achieves high energy density up to 12.3 mWh cm −3 with a maximum power density of 1600 mW cm −3 , outperforming the thin-film lithium battery. Therefore, these supercapacitors are promising for the next generation flexible and wearable electronic devices.

Trimetallic Sulfide Hollow Superstructures with Engineered d‐Band Center for Oxygen Reduction to Hydrogen Peroxide in Alkaline Solution

Abstract: High‐performance transition metal chalcogenides (TMCs) as electrocatalysts for two‐electron oxygen reduction reaction (2e‐ORR) in alkaline medium are promising for hydrogen peroxide (H 2 O 2 ) production, but their synthesis remains challenging. In this work, a titanium‐doped zinc–cobalt sulfide hollow superstructure (Ti–ZnCoS HSS) is rationally designed as an efficient electrocatalyst for H 2 O 2 electrosynthesis. Synthesized by using hybrid metal–organic frameworks (MOFs) as precursors after sulfidation treatment, the resultant Ti–ZnCoS HSS exhibits a hollow‐on‐hollow superstructure with small nanocages assembled around a large cake‐like cavity. Both experimental and simulation results demonstrate that the polymetallic composition tailors the d‐band center and binding energy with oxygen species. Moreover, the hollow superstructure provides abundant active sites and promotes mass and electron transfer. The synergistic d‐band center and superstructure engineering at both atomic and nanoscale levels lead to the remarkable 2e‐ORR performance of Ti–ZnCoS HSS with a high selectivity of 98%, activity (potential at 1 mA cm −2 of 0.774 V vs reversible hydrogen electrode (RHE)), a H 2 O 2 production rate of 675 mmol h –1 g cat –1 , and long‐term stability in alkaline condition, among the best 2e‐ORR electrocatalysts reported to date. This strategy paves the way toward the rational design of polymetallic TMCs as advanced 2e‐ORR catalysts.

Abattoir-Based Serological Surveillance and Spatial Risk Analysis of Foot-and-Mouth Disease, Brucellosis, and Q Fever in Lao PDR Large Ruminants

Abstract: A national animal disease surveillance network initiated by the Lao PDR government is adopted and reinforced by a joint research project between the National Animal Health Laboratory (NAHL), the Department of Livestock and Fisheries (DLF), and the Mahidol Oxford Tropical Medicine Research Unit (MORU). The network is strengthened by staff training and practical exercises and is utilised to provide zoonotic or high-impact disease information on a national scale. Between January and December 2020, large ruminant s les are collected monthly from 18 abattoirs, one in each province, by provincial and district agriculture and forestry officers. The surveillance network collected a total of 4247 serum s les (1316 buffaloes and 2931 cattle) over this period. S les are tested for antibodies against Brucella spp., Coxiella burnetii (Q fever) and Foot-and-Mouth Disease Non-Structural Protein (FMD NSP) using commercial ELISA kits and the Rose Bengal test. Seroprevalences of Q fever and brucellosis in large ruminants are low at 1.7% (95% CI: 1.3, 2.1) and 0.7% (95% CI: 0.5, 1.0) respectively, while for FMD NSP it is 50.5% (95% CI: 49.0, 52.0). Univariate analyses show differences in seroprevalences of Q fever between destination (abattoir) province (p-value = 0.005), province of origin (p-value = 0.005), animal type (buffalo or cattle) (p-value = 0.0008), and collection month (p-value = 3.4 × 10−6). Similar to Q fever, seroprevalences of brucellosis were significantly different for destination province (p-value 0.00001), province of origin (p-value 0.00001), animal type (p-value = 9.9 × 10−5) and collection month (p-value 0.00001), plus body condition score (p-value = 0.003), and age (p-value = 0.007). Additionally, risk factors of the FMD NSP dataset include the destination province (p-value 0.00001), province of origin (p-value 0.00001), sex (p-value = 7.97 × 10−8), age (p-value = 0.009), collection date (p-value 0.00001), and collection month (p-value 0.00001). Spatial analyses revealed that there is no spatial correlation of FMD NSP seropositive animals. High-risk areas for Q fever and brucellosis are identified by spatial analyses. Further investigation of the higher risk areas would provide a better epidemiological understanding of both diseases in Lao PDR. In conclusion, the abattoir serological survey provides useful information about disease exposure and potential risk factors. The network is a good base for field and laboratory staff training in practical technical skills. However, the sustainability of such a surveillance activity is relatively low without an external source of funding, given the operational costs and insufficient government budget. The cost-effectiveness of the abattoir survey could be increased by targeting hotspot areas, reducing fixed costs, and extending the focus to cover more diseases.

Risk factors analysis for neglected human rickettsioses in rural communities in Nan province, Thailand: A community-based observational study along a landscape gradient

Abstract: In this study, we estimated exposure for Scrub typhus (STG), Typhus (TG) and Spotted fever groups (SFG) rickettsia using serology at a fine scale (a whole sub-district administration level) of local communities in Nan Province, Thailand. Geographical characteristics of the sub-district were ided into two landscape types: lowland agricultural area in an urbanized setting (lowland-urbanized area) and upland agricultural area located close to a protected area of National Park (upland-forested area). This provided an ideal contrast between the two landscapes with low and high levels of human-altered habitats to study in differences in disease ecology. In total, 824 serum s les of participants residing in the eight villages were tested by screening IgG ELISA, and subsequently confirmed by the gold standard IgG Immunofluorescent Assay (IFA). STG and TG IgG positivity were highest with seroprevalence of 9.8% and 9.0%, respectively whereas SFG positivity was lower at 6.9%. Inhabitants from the villages located in upland-forested area demonstrated significantly higher STG exposure, compared to those villages in the lowland-urbanized area (chi-square = 51.97, p 0.0001). In contrast, TG exposure was significantly higher in those villagers living in lowland-urbanized area (chi-square = 28.26, p 0.0001). In addition to the effect of landscape types, generalized linear model (GLM) analysis identified socio-demographic parameters, i.e., gender, occupation, age, educational level, domestic animal ownership (dog, cattle and poultry) as influential factors to explain the level of rickettsial exposure (antibody titers) in the communities. Our findings raise the public health awareness of rickettsiosis as a cause of undiagnosed febrile illness in the communities.

A S‐Scheme MOF‐on‐MOF Heterostructure

Abstract: Constructing MOF‐on‐MOF heterojunction with elaborate charge transfer mechanism and interface is a promising strategy for improving the photocatalytic properties of MOFs. Herein, a Step‐scheme (S‐scheme) MIL‐125‐NH 2 @CoFe Prussian blue analogue (PBA) heterojunction is reported for the first time. The MOF‐on‐MOF heterostructure exhibits a sandwich‐like morphology with hollow CoFe PBA nanocages selectively assembled on the top‐down surfaces of MIL‐125‐NH 2 nanocakes. Experimental findings and theoretical simulation results reveal the formation of internal electric field via interfacial TiOCo bonds at the heterojunction, providing driving force and atomic transportation highway for accelerating the S‐scheme charge transfer and enhancing the redox performance. Contributed further by the hollow sandwich‐like structures with increased active site exposure, the designed MOF‐on‐MOF heterojunction exhibits significantly enhanced photocatalytic activity for degradation of various organic pollutants. This study provides insights toward the rational design of semiconducting MOF‐based heterojunctions with improved properties.

Rickettsial Infections Are Neglected Causes of Acute Febrile Illness in Teluk Intan, Peninsular Malaysia

Abstract: Rickettsial infections are among the leading etiologies of acute febrile illness in Southeast Asia. However, recent data from Malaysia are limited. This prospective study was conducted in Teluk Intan, Peninsular Malaysia, during January to December 2016. We recruited 309 hospitalized adult patients with acute febrile illness. Clinical and biochemistry data were obtained, and patients were stratified into mild and severe infections based on the sepsis-related organ failure (qSOFA) scoring system. Diagnostic assays including blood cultures, real-time PCR, and serology (IFA and MAT) were performed. In this study, pathogens were identified in 214 (69%) patients, of which 199 (93%) patients had a single etiology, and 15 (5%) patients had etiologies. The top three causes of febrile illness requiring hospitalization in this Malaysian study were leptospirosis (68 (32%)), dengue (58 (27%)), and rickettsioses (42 (19%)). Fifty-five (18%) patients presented with severe disease with a qSOFA score of ≥2. Mortality was documented in 38 (12%) patients, with the highest seen in leptospirosis (16 (42%)) followed by rickettsiosis (4 (11%)). While the significance of leptospirosis and dengue are recognized, the impact of rickettsial infections in Peninsular Malaysia remains under appreciated. Management guidelines for in-patient care with acute febrile illness in Peninsular Malaysia are needed.

Small Mesoporous Silica Nanoparticles as Carriers for Enhanced Photodynamic Therapy

Abstract: Small mesoporous silica nanoparticles (MSNs ca. 37 nm in diameter) have a high loading capacity for a hydrophobic photosensitizer, SiPcCl(2) (82.6% in weight), and excellent endocytosis properties. As a result, the amount of SiPcCl(2) being delivered to cancer cells is increased by approximately two orders of magnitude compared to pure SiPcCl(2) at the same dosage, and the photodynamic therapy (PDT) efficiency is enhanced by over fourfold. Our method can be widely used to increase the dosage of hydrophobic anti-cancer drugs in cancer cells and therefore increase the cytotoxicity of the drugs.

A Use of 56-kDa Recombinant Protein of Orientia tsutsugamushi Karp Serotype in Serodiagnosis of Scrub Typhus by Enzyme-Linked Immunosorbent Assay in Thais

Abstract: Scrub typhus is a mite-borne disease caused by a Gram-negative obligately intracellular bacillus, Orientia tsutsugamushi. The disease is endemic in the Asia–Australia–Pacific region, including Thailand. Scrub typhus generally manifests as acute undifferentiated febrile fever along with myalgia, rash, and lymphadenopathy. An eschar can be a valuable diagnostic clue, but this skin lesion may be missed in some patients. The disease symptoms resemble those of other febrile illnesses such as leptospirosis, typhoid, murine typhus, malaria, and dengue fever, making a laboratory diagnosis necessary for the definitive diagnosis. In this study, we expressed a recombinant protein derived from 56-kDa type-specific antigen of O. tsutsugamushi Karp serotype and tested its ability to detect and differentiate scrub typhus infection. IgM and IgG antibodies were determined in sera from scrub typhus (n = 92) and other febrile illness patients (murine typhus (n = 25), melioidosis (n = 36), leptospirosis (n = 42), and dengue (n = 35)) from Thailand. Sensitivities of 87.0% and 59.8% with a specified assay cut-off were obtained for IgM and IgG indirect ELISAs, respectively, with a specificity of 100% in both tests. The sensitivity was increased to 95.7% when a combination of IgM and IgG ELISAs results was considered. Our study suggested a potential of the 56-kDa recombinant protein for further development and evaluation for use in scrub typhus serodiagnosis.

Abstract: We report a new approach to produce macroporous ( approximately 110 nm in diameter) ordered siliceous foams (MOSF) by using block copolymers as templates in the absence of any organic cosolvent. The fine three-dimensional honeycomb structure of MOSF was determined by electron tomography. A formation mechanism of MOSF that spans from the atomic to macroscopic scale is proposed, which involves the cooperative self-assembly of unilamellar vesicles followed by the supra-assembly of vesicles. The fusion of soft vesicles finally leads to MOSF with well-ordered and defined honeycomb structures.

Production diseases in smallholder pig systems in rural Lao PDR

Abstract: Pigs in Lao People's Democratic Republic are important for income and food security, particularly in rural households. The majority of pigs are reared in smallholder systems, which may challenge the implementation of any disease control strategies. To investigate risk factors for pig production diseases in such farming systems in the country a serological survey was conducted during 2011. A total of 647 pigs were s led, accounting for 294 households in Luang Prabang and 353 in Savannakhet province representing upland and lowland, respectively. The results demonstrated that pigs in Lao PDR had antibodies against erysipelas (45.2%), CSF (11.2%), PRRS (8.6%), FMD O (17.2%) and FMD Asia 1, (3.5%). Differences in the housing systems influenced disease risk, for ex le, penned pigs had reduced odds of FMD and CSF, compared to those in scavenger systems. Pigs owned by farms using a sanaam (a communal area where pigs are kept for some time of the year) had 3.93 (95% confidence interval (CI): 1.09-14.7) times the odds of having pigs seropositive for FMD. Farms on which sudden piglet deaths had been experienced were more likely to have pigs seropositive for FMD O and erysipelas. These diseases constrain the development of village farming and the wider livestock industry due to their impact on productivity and trade. Vaccination coverage for FMD and CSF was low and there was a lack of national funding for livestock disease control at the time of the study. Further investigation into sustainable low-cost control strategies for these pathogens is warranted.

Abstract: Anisotropy plays a unique role in the structural regulation of metal–organic frameworks (MOFs) and their composites, especially at the micro‐ and nanoscale. However, there is a lack of a understanding of MOF micro/nanoparticles (MNPs) from the perspective of morphological anisotropy. In this Minireview, recent advances in anisotropic MOF MNPs are summarized, with a focus on how morphological anisotropy leads to innovative structures and modulates properties. First, anisotropic pristine MOF MNPs with erse morphologies are introduced and classified by their morphology‐dependent and morphology‐independent anisotropy. Secondly, the anisotropy‐enabled site‐selective higher‐order construction of MOF‐based materials is highlighted. Finally, challenges and prospects for anisotropic MOFs are discussed, aiming to provide inspiration for further developments in this interesting research field.

A spatio-temporal analysis of scrub typhus and murine typhus in Laos; implications from changing landscapes and climate

Abstract: Scrub typhus (ST) and murine typhus (MT) are common but poorly understood causes of fever in Laos. We examined the spatial and temporal distribution of ST and MT, with the intent of informing interventions to prevent and control both diseases. This study included s les submitted from 2003 to 2017 to Mahosot Hospital, Vientiane, for ST and MT investigation. Serum s les were tested using IgM rapid diagnostic tests. Patient demographic data along with meteorological and environmental data from Laos were analysed. Approximately 17% of patients were positive for either ST (1,337/8,150 patients tested) or MT (1,283/7,552 patients tested). While both diseases occurred in inhabitants from Vientiane Capital, from the univariable analysis MT was positively and ST negatively associated with residence in Vientiane Capital. ST was highly seasonal, with cases two times more likely to occur during the wet season months of July-September compared to the dry season whilst MT peaked in the dry season. Multivariable regression analysis linked ST incidence to fluctuations in relative humidity whereas MT was linked to variation in temperature. Patients with ST infection were more likely to come from villages with higher levels of surface flooding and vegetation in the 16 days leading up to diagnosis. The data suggest that as cities expand, high risk areas for MT will also expand. With global heating and risks of attendant higher precipitation, these data suggest that the incidence and spatial distribution of both MT and ST will increase.

Abstract: Lateral flow assays (LFAs) have attracted much attention as rapid and affordable point-of-care devices for medical diagnostics. The global SARS-CoV-2 pandemic has further highlighted the importance of LFAs. Many efforts have been made to enhance the sensitivity of LFAs. In recent years, silica nanomaterials have been used to either lify the signal of label materials or provide stability, resulting in better detection performance. In this review, the recent progress of silica-nanomaterial-assisted LFAs is summarized. The impact of the structure of silica nanomaterials on LFA performance, the challenges and prospects in this research area are also discussed.

Abstract: Photocatalytic H 2 O 2 production by conversion of O 2 in aqueous solution is often challenged by the use of sacrificial agents, the separation of powdery photocatalysts, solution, and contaminants, and low activity of photocatalyst. Herein, a membrane of covalent furan‐benzimidazole‐linked polymer (Furan‐BILP) with both O‐ and N‐containing heterocycles bonded via OCCN is reported for the first time as a photocatalyst to harvest clean H 2 O 2 in pure water with high‐performance. A coordination‐polymer hard template strategy is developed to produce Furan‐BILP hollow microfibers that can be further assembled into membranes with desired sizes. The resultant Furan‐BILP membrane directly delivers clean H 2 O 2 solution as the product with a high H 2 O 2 production rate of 2200 µmol g −1 h −1 in pure water. Density functional theory calculations and experiment results indicate that the C atom from Furan ring on the linkage binds to the adsorbed OOH * , the H atom of OOH * forms a hydrogen bond with the N atom in the benzimidazole ring, thus the intermediate six‐membered ring structure stabilizes the OOH * and favors 2e‐ORR. The strategy using both molecular engineering to tune the electronic structure and macrostructural engineering to shape the morphology may be applied to design other coordination organic polymer photocatalysts with further improved performance.

Abstract: Aluminum‐containing adjuvants used in vaccine formulations suffer from low cellular immunity, severe aggregation, and accumulation in the brain. Conventional aluminosilicates widely used in the chemical industry focus mainly on acidic sites for catalytic applications, but they are rarely used as adjuvants. Reported here is an innovative “ligand‐assisted steric hindrance” strategy to create a high density of six‐coordinate VI Al−OH groups with basicity on dendritic mesoporous silica nanoparticles as new nanoadjuvants. Compared to four‐coordinate IV Al‐modified counterparts, VI Al−OH‐rich aluminosilicate nanoadjuvants enhance cellular delivery of antigens and provoke stronger cellular immunity. Moreover, the aluminum accumulation in the brain is more reduced than that with a commercial adjuvant. These results show that coordination chemistry can be used to control the adjuvanticity, providing new understanding in the development of next‐generation vaccine adjuvants.

Abstract: This study investigated the comparative accuracy of a recombinant 56-kDa type-specific antigen-based rapid diagnostic test (RDT) for scrub typhus for the detection of IgM antibodies by using conventional serology in well-characterized serum s les from undifferentiated febrile illness patients. The RDT showed high specificity and promising comparative accuracy, with 82% sensitivity and 98% specificity for s les defined positive at an IgM indirect immunofluorescence assay positivity cutoff titer of ≥1:1,600 versus 92% and 95% at ≥1:6,400, respectively.

Spatiotemporal Drivers of the African Swine Fever Epidemic in Lao PDR

Abstract: African swine fever (ASF) is a devastating transboundary disease of swine. Following the first report of ASF in China in August 2018, ASF spread through South-East Asian nations in 2019. Without control and containment measures, ASF can decimate smallholder pig holdings and disrupt value chains. This study aimed to describe the spatiotemporal spread of the 2019 Lao PDR ASF epidemic in domestic pigs, identify environmental (protected areas and forests), production (pig ownership), transportation (roads), and social (poverty and ethnicity) risk factors, and recommend measures that could reduce ASF spread. A retrospective spatiotemporal study was conducted at the village level. Information on the date that ASF was first reported from each case location was collected, and the outcome variable of interest “epidemic day” was created. Risk factor information from different sources was extracted for each case location. The association or correlation between epidemic day and risk factors for the spread of ASF was investigated using Kruskal–Wallis tests and Spearman rank correlation statistics. The epidemic started on June 16 and lasted for 190 days, displaying a right-skewed epidemic curve. The directional distribution was rotated approximately 305°, from Southeast to Northwest Laos. Significant risk factors for ASF associated with epidemic day were location in terms of distance from the closest protected natural area P = 0.020 , pig ownership P = 0.005 , road networks P = 0.003 , and poverty indices P 0.001 . Cases were reported earlier in this epidemic at locations that were closer to protected natural areas, of higher pig ownership, more connected via the national road network, and which experienced elevated poverty. The spatiotemporal pattern described suggests that ASF was introduced via infected pigs from Vietnam. Based on study findings, recommendations for smallholder pig production in Southeast Asia include improving knowledge of swine value chains to inform disease risk and control, monitoring pig transportation, implementing stricter biosecurity measures on the domestic pig population, and providing biosecurity support and education to smallholder pig farmers in poverty.

Superstructured Macroporous Carbon Rods Composed of Defective Graphitic Nanosheets for Efficient Oxygen Reduction Reaction

Abstract: Rationally designed carbon materials with superstructures are promising candidates in applications such as electrocatalysis. However, the synthesis of highly porous carbon superstructures with macropores and carbon defects from a simple crystalline solid remains challenging. In this work, superstructured macroporous carbon rods composed of defective graphitic nanosheets are synthesized by direct carbonization of crystalline poly tannic acid (PTA) rods as precursors. During carbonization, PTA rods with a highly ordered lamellar structure induce a spatially confined two‐step localized contraction that takes place in different dimensions and directions in each step. The unexpected contraction behavior results in the sponge‐like macroporous carbon superstructure with large surface area, high porosity, and abundant defects, thus showing a superior electrocatalytic performance with high activity and selectivity for oxygen reduction reaction. The study provides new understandings in the design of functional carbon materials with distinctive structures and applications.

Manganese-Doped Silica-Based Nanoparticles Promote the Efficacy of Antigen-Specific Immunotherapy

Abstract: Prophylactic human papillomavirus (HPV) vaccines are commercially available for prevention of infection with cancerogenic HPV genotypes but are not able to combat pre-existing HPV-associated disease. In this study, we designed a nanomaterial-based therapeutic HPV vaccine, comprising manganese (Mn4+)-doped silica nanoparticles (Mn4+-SNPs) and the viral neoantigen peptide GF001 derived from the HPV16 E7 oncoprotein. We show in mice that Mn4+-SNPs act as self-adjuvants by activating the inflammatory signaling pathway via generation of reactive oxygen species, resulting in immune cell recruitment to the immunization site and dendritic cell maturation. Mn4+-SNPs further serve as Ag carriers by facilitating endo/lysosomal escape via depletion of protons in acidic endocytic compartments and subsequent Ag delivery to the cytosol for cross-presentation. The Mn4+-SNPs+GF001 nanovaccine induced strong E7-specific CD8+ T cell responses, leading to remission of established murine HPV16 E7-expressing solid TC-1 tumors and E7-expressing transgenic skin grafts. This vaccine construct offers a simple and general strategy for therapeutic HPV and potentially other cancer vaccines.

Silica vesicles as nanocarriers and adjuvants for generating both antibody and T-cell mediated immune resposes to Bovine Viral Diarrhoea Virus E2 protein

Abstract: Bovine Viral Diarrhoea Virus (BVDV) is widely distributed in cattle industries and causes significant economic losses worldwide annually. A limiting factor in the development of subunit vaccines for BVDV is the need to elicit both antibody and T-cell-mediated immunity as well as addressing the toxicity of adjuvants. In this study, we have prepared novel silica vesicles (SV) as the new generation antigen carriers and adjuvants. With small particle size of 50 nm, thin wall (~6 nm), large cavity (~40 nm) and large entrance size (5.9 nm for SV-100 and 16 nm for SV-140), the SV showed high loading capacity (∼ 250 μg/mg) and controlled release of codon-optimised E2 (oE2) protein, a major immunogenic determinant of BVDV. The in vivo functionality of the system was validated in mice immunisation trials comparing oE2 plus Quil A (50 μg of oE2 plus 10 μg of Quil A, a conventional adjuvant) to the oE2/SV-140 (50 μg of oE2 adsorbed to 250 μg of SV-140) or oE2/SV-140 together with 10 μg of Quil A. Compared to the oE2 plus Quil A, which generated BVDV specific antibody responses at a titre of 10(4), the oE2/SV-140 group induced a 10 times higher antibody response. In addition, the cell-mediated response, which is essential to recognise and eliminate the invading pathogens, was also found to be higher [1954-2628 spot forming units (SFU)/million cells] in mice immunised with oE2/SV-140 in comparison to oE2 plus Quil A (512-1369 SFU/million cells). Our study has demonstrated that SV can be used as the next-generation nanocarriers and adjuvants for enhanced veterinary vaccine delivery.

Abstract: Here we estimated the accuracy of the InBios Scrub Typhus Detect™ immunoglobulin M (IgM) ELISA to determine the optimal optical density (OD) cut-off values for the diagnosis of scrub typhus. Patients with undifferentiated febrile illness from Chittagong, Bangladesh, provided s les for reference testing using (i) qPCR using the Orientia spp. 47-kDa htra gene, (ii) IFA ≥1:3200 on admission, (iii) immunofluorescence assay (IFA) ≥1:3200 on admission or 4-fold rise to ≥3200, and (iv) combination of PCR and IFA positivity. For sero-epidemiological purposes (ELISA vs. IFA ≥1:3200 on admission or 4-fold rise to ≥3200), the OD cut-off for admission s les was ≥1.25, resulting in a sensitivity (Sn) of 91.5 (95% confidence interval (95% CI: 96.8–82.5) and a specificity (Sp) of 92.4 (95% CI: 95.0–89.0), while for convalescent s les the OD cut-off was ≥1.50 with Sn of 66.0 (95% CI: 78.5–51.7) and Sp of 96.0 (95% CI: 98.3–92.3). Comparisons against comparator reference tests (ELISA vs. all tests including PCR) indicated the most appropriate cut-off OD to be within the range of 0.75–1.25. For admission s les, the best Sn/Sp compromise was at 1.25 OD (Sn 91.5%, Sp 92.4%) and for convalescent s les at 0.75 OD (Sn 69.8%, Sp 89.5%). A relatively high (stringent) diagnostic cut-off value provides increased diagnostic accuracy with high sensitivity and specificity in the majority of cases, while lowering the cut-off runs the risk of false positivity. This study underlines the need for regional assessment of new diagnostic tests according to the level of endemicity of the disease given the high levels of residual or cross-reacting antibodies in the general population.

Abstract: Murine typhus is a rarely diagnosed cause of acute febrile illness in Malaysia, and its true disease burden is unknown. We report a case of an acute murine typhus infection in a patient living in a small city in Peninsular Malaysia, presenting with fever, rash, and headache. Unresponsive to the initial empirical treatment for leptospirosis, he showed a rapid response to doxycycline when murine typhus was diagnosed later. This case highlights the importance of considering murine typhus as a diagnostic in cases of acute febrile illness in urban and sub-urban areas, such as that of in Peninsular Malaysia.

Confinement of Chemisorbed Phosphates in a Controlled Nanospace with Three-Dimensional Mesostructures

Abstract: The confinement of phosphates inside adsorbents is important for not only entrophication control, but also the recovery of phosphorous, a depleting natural resource. However, the behaviour of chemisorbed phosphates inside nanoporous materials has not been systematically studied. Here, the confinement of chemisorbed phosphates in a three-dimensional cubic mesoporous material with adjustable structural parameters is systematically investigated. By taking advantage of advanced electron tomography techniques, the relationship between the growth of chemisorbed phosphates, the overall phosphate-adsorption performance and the mesostructural parameters is revealed. Cubic cage-type FDU-12 materials with tuneable entrance sizes were prepared and functionalised with different amounts of lanthanum oxide. When the entrance size is smaller than approximately 5 nm, phosphates are found only in isolated cages, thereby leading to low lanthanum (La) usage efficiency and phosphate removal capacity. When the entrance size is increased, chemisorption occurs in both cages and entrances, thus forming crystalline LaPO4 nanorods and increasing both the La usage efficiency and the phosphate removal capacity. In addition, the LaPO4 nanorods show a preferential orientation along the [110] direction. This study provides new insights in the rational design of phosphate adsorbents with controlled structures and high performance.

Hierarchical Cu4V2.15O9.38 micro-/nanostructures: a lithium intercalating electrode material

Abstract: Hierarchical Cu4V2.15O9.38 micro-/nanostructures have been prepared by a facile "forced hydrolysis" method, from an aqueous peroxovanadate and cupric nitrate solution in the presence of urea. The hierarchical architectures with diameters of 10-20 µm are assembled from flexible nanosheets and rigid nanoplates with widths of 2-4 µm and lengths of 5-10 µm in a radiative way. The preliminary electrochemical properties of Cu4V2.15O9.38 have been investigated for the first time and correlated with its structure. This material delivers a large discharge capacity of 471 mA h g(-1) above 1.5 V, thus making it an interesting electrode material for primary lithium ion batteries used in implantable cardioverter defibrillators.

Abstract: Here, ZnFe2 O4 double-shell hollow microspheres are designed to accommodate the large volume expansion during lithiation. A facile and efficient vapor-phase polymerization method has been developed to coat the ZnFe2 O4 hollow spheres with polypyrrole (PPY). The thin PPY coating improves not only the electronic conductivity but also the structural integrity, and thus the cycling stability of the ZnFe2 O4 hollow spheres. Our work sheds light on how to enhance the electrochemical performance of transition metal oxide-based anode materials by designing delicate nanostructures.

Abstract: Immunosuppressive tumors generally exhibit poor response to immune checkpoint blockade based cancer immunotherapy. Rationally designed hybrid nanoreactors are now presented that have integrated functions as Fenton catalysts and glutathione depletion agents for lifying the immunogenic cell death and activating immune cells. A simple physical mixture of nanoreactors and chemodrugs in combination with immune checkpoint blockades show synergistically and concurrently enhanced chemo-immunotherapy efficacy, inhibiting the growth of both treated primary immunosuppressive tumors and untreated distant tumors. The off-the-shelf strategy uses tumor antigens generated in situ and avoids cargo loading, and is thus a substantial advance in personalized nanomedicine for clinical translation.

Abstract: Osteoarthritis (OA) is a common joint degenerative disease that causes pain, joint damage, and dysfunction. External hyaluronic acid (HA) supplement is a common method for the management of osteoarthritis which requires multi-injections. It is demonstrated that biodegradable mesoporous silica nanoparticles successfully deliver an enzyme, hyaluronan synthase type 2 (HAS2), into synoviocytes from the temporomandibular joint (TMJ) and generate endogenous HA with high molecular weights. In a rat TMJ osteoarthritis inflammation model, this strategy promotes endogenous HA production and inhibits the synovial inflammation of OA for more than 3 weeks with one-shot administration. Such nanotherapy also helps repairing the bone defects in a rat OA bone defect model.

Abstract: Anaplasma marginale is a devastating tick-borne pathogen causing anaplasmosis in cattle and results in significant economic loss to the cattle industry worldwide. Currently, there is no widely accepted vaccine against A. marginale. New generation subunit vaccines against A. marginale, which are much safer, more efficient and cost-effective, are in great need. The A. marginale outer membrane protein VirB9-1 is a promising antigen for vaccination. We previously have shown that soluble recombinant VirB9-1 protein can be expressed and purified from Escherichia coli and induce a high level of humoral and cellular immunity in mice. In this study, we re-formulated the nanovaccines using the partially-purified VirB9-1 protein as the antigen and hollow nano-size silica vesicles (SV-100) as the adjuvant. We simplified the purification method to obtain the partially-purified antigen VirB9-1 with a six-fold higher yield. The new formulations using the partially-purified VirB9-1 protein achieved higher antibody and cell-mediated immune responses compared to the purified ones. This finding suggests that the partially-purified VirB9-1 protein performs better than the purified ones in the vaccination against A. marginale, and a certain level of contaminants in the protein antigen can be self-adjuvant and boost immunogenicity together with the nanoparticle adjuvant. This may lead to finding a "Goldilocks" level of contaminants. The new nanovaccine formulation using partially-purified antigens along with nanoparticle adjuvants offers an alternative strategy for making cheaper veterinary vaccines.

Abstract: Iron oxide nanoparticles (IONPs) have emerging anticancer applications via polarizing tumor-associated macrophages from tumor-promoting phenotype (M2) to tumor-suppressing phenotype (M1). However, the underlying mechanism and structure-function relationship remain unclear. We report magnetite IONPs are more effective compared to hematite in M1 polarization and tumor suppression. Moreover, magnetite IONPs specifically rely on interferon regulatory factor 5 signaling pathway for M1 polarization and down-regulate M2-assoicated arginase-1. This study provides new understandings and paves the way for designing advanced iron-based anticancer technologies.

Functionalized Periodic Mesoporous Organosilicas for Enhanced and Selective Peptide Enrichment

Abstract: The analysis of peptides by the mass spectrometry (MS) technique is important in modern life science. The enrichment of peptides can increase the detection efficiency and is sometimes indispensable for collecting the information on proteins with low-abundance. Herein, we first report that functionalized periodic mesoporous organosilica (PMO) materials have a superior peptide enrichment property. It is demonstrated that the PMO materials with an organo-bridged (-CH(2)-) hybrid wall composition display a highly enhanced peptide enrichment ability compared to the pure silica material (SBA-15) with similar mesostructured parameters and morphology. More importantly, by surface modification of PMO with amino groups (denoted NH(2)-PMO), PMO and NH(2)-PMO with opposite charged surfaces (-25.2 and +39.0 mV, respectively) show selective affinities for positively and negatively charged peptides, respectively. By directly adding PMO, NH(2)-PMO as well as pure silica materials to the peptides solution with a low concentration (1-2 fmol/microL), 36 and 28 peptides can be detected from the BSA digestion in the presence of PMO and NH(2)-PMO, respectively, while only 6 and 4 are monitored in the case of SBA-15 enrichment and from solution without enrichment, respectively. Moreover, 69.4% (25 of 36) of enriched peptides by PMO have pI > or = 6 and 80% (21 of 28) of enriched peptides by NH(2)-PMO possess pI < or = 6. Combining the results from the NH(2)-PMO and PMO enrichment together, 51 peptides can be identified with a MOWSE score of 333. It is also noted that similar conclusions can also be obtained from the peptides solution originated from other proteins. This might be an important contribution to the understanding of the interaction between peptides and porous hosts, and the proposed method is promising for the development of both material science and biotechnology.

Abstract: Recently, dendritic mesoporous silica nanoparticles with widespread applications have attracted great interest. Despite many publications ( ), the terminology “dendritic” is ambiguous. Understanding what possible “dendritic structures” are, their formation mechanisms and the underlying structure–property relationship is fundamentally important. With the advance of characterization techniques such as electron tomography, two types of tree‐branch‐like and flower‐like structures can be distinguished, both described as “dendritic” in the literature. In this Review, we start with the definition of “dendritic”, then provide critical analysis of reported dendritic silica nanoparticles according to their structural classification. We update the understandings of the formation mechanisms of two types of “dendritic” nanoparticles, highlighting how to control their structural parameters. Applications of dendritic mesoporous nanoparticles are also reviewed with a focus on the biomedical field, providing new insights into the structure–property relationship in this family of nanomaterials.

Optimal Cutoff and Accuracy of an IgM Enzyme-Linked Immunosorbent Assay for Diagnosis of Acute Scrub Typhus in Northern Thailand: an Alternative Reference Method to the IgM Immunofluorescence Assay.

Abstract: The enzyme-linked immunosorbent assay (ELISA) has been proposed as an alternative serologic diagnostic test to the indirect immunofluorescence assay (IFA) for scrub typhus. Here, we systematically determine the optimal s le dilution and cutoff optical density (OD) and estimate the accuracy of IgM ELISA using Bayesian latent class models (LCMs). Data from 135 patients with undifferentiated fever were reevaluated using Bayesian LCMs. Every patient was evaluated for the presence of an eschar and tested with a blood culture for Orientia tsutsugamushi , three different PCR assays, and an IgM IFA. The IgM ELISA was performed for every s le at s le dilutions from 1:100 to 1:102,400 using crude whole-cell antigens of the Karp, Kato, and Gilliam strains of O. tsutsugamushi developed by the Naval Medical Research Center. We used Bayesian LCMs to generate unbiased receiver operating characteristic curves and found that the s le dilution of 1:400 was optimal for the IgM ELISA. With the optimal cutoff OD of 1.474 at a s le dilution of 1:400, the IgM ELISA had a sensitivity of 85.7% (95% credible interval [CrI], 77.4% to 86.7%) and a specificity of 98.1% (95% CrI, 97.2% to 100%) using paired s les. For the ELISA, the OD could be determined objectively and quickly, in contrast to the reading of IFA slides, which was both subjective and labor-intensive. The IgM ELISA for scrub typhus has high diagnostic accuracy and is less subjective than the IgM IFA. We suggest that the IgM ELISA may be used as an alternative reference test to the IgM IFA for the serological diagnosis of scrub typhus.

Abstract: Rechargeable aluminium batteries are a promising alternative battery technology compared to lithium-ion batteries, because of the high theoretical capacity, low cost and high safety of aluminium. The past decade has witnessed the rapid development of rechargeable aluminium battery technology with the focus on exploring high performance cathode materials and investigating their charge storage mechanisms. However, the challenges in the cathode research including inadequate capacity, sluggish reaction kinetics and inferior cycling stability still remain. Various strategies have been attempted to address these challenges to realize the advantages of rechargeable aluminium batteries. The present review aims to collect the comprehensive body of research performed in the literature hitherto to develop interaction/conversion/coordination type cathodes for rechargeable aluminium batteries. Future research directions and prospects in rechargeable aluminium battery field are also proposed.

Mesoporous Organosilica Nanoparticles with Tetrasulphide Bond to Enhance Plasmid DNA Delivery

Abstract: Cellular delivery of plasmid DNA (pDNA) specifically into dendritic cells (DCs) has provoked wide attention in various applications. However, delivery tools that achieve effective pDNA transfection in DCs are rare. Herein, we report that tetrasulphide bridged mesoporous organosilica nanoparticles (MONs) have enhanced pDNA transfection performance in DC cell lines compared to conventional mesoporous silica nanoparticles (MSNs). The mechanism of enhanced pDNA delivery efficacy is attributed to the glutathione (GSH) depletion capability of MONs. Reduction of initially high GSH levels in DCs further increases the mammalian target of rapamycin complex 1 (mTORc1) pathway activation, enhancing translation and protein expression. The mechanism was further validated by showing that the increased transfection efficiency was apparent in high GSH cell lines but not in low GSH ones. Our findings may provide a new design principle of nano delivery systems where the pDNA delivery to DCs is important.

Synthesis of silica vesicles with controlled entrance size for high loading, sustained release, and cellular delivery of therapeutical proteins

Abstract: A rationally designed two-step synthesis of silica vesicles is developed with the formation of vesicular structure in the first step and fine control over the entrance size by tuning the temperature in the second step. The silica vesicles have a uniform size of ≈50 nm with excellent cellular uptake performance. When the entrance size is equal to the wall thickness, silica vesicles after hydrophobic modification show the highest loading amount (563 mg/g) towards Ribonuclease A with a sustained release behavior. Consequently, the silica vesicles are excellent nano-carriers for cellular delivery applications of therapeutical biomolecules.

Rational Design of Multifunctional Dendritic Mesoporous Silica Nanoparticles to Load Curcumin and Enhance Efficacy for Breast Cancer Therapy

Abstract: Breast cancer is the primary reason for cancer-related death in women worldwide and the development of new formulations to treat breast cancer patients is crucial. Curcumin (Cur), a natural product, exerts promising anticancer activities against various cancer types. However, its therapeutic efficacy is hindered as a result of poor water solubility, instability, and low bioavailability. The aim of this work is to assess the curative effect of a novel nanoformulation, i.e., Cur-loaded and calcium-doped dendritic mesoporous silica nanoparticles modified with folic acid (Cur-Ca@DMSNs-FA) for breast cancer therapy. The results manifested that Cur-Ca@DMSNs-FA dispersed very well in aqueous solution, released Cur with a pH-responsible profile, and targeted efficiently to human breast cancer MCF-7 cells. Further investigations indicated that Cur-Ca@DMSNs-FA effectively inhibited cell proliferation, increased intracellular ROS generation, decreased mitochondrial membrane potential, and enhanced cell cycle retardation at G2/M phase, leading to a higher apoptosis rate in MCF-7 compared to free Cur. Moreover, the Western blotting analysis demonstrated that Cur-Ca@DMSNs-FA were more active than free Cur through suppression of PI3K/AKT/mTOR and Wnt/β-catenin signaling, and activation of the mitochondria-mediated apoptosis pathway. In addition, hemolysis assay showed that the Ca@DMSNs-FA exhibited good biocompatibility. Last, in vivo studies indicated that when Cur was encapsulated in Ca@DMSNs-FA, the Cur concentration in blood serum and tumor tissues was increased after 1 h intraperitoneal injection. In conclusion, Cur-Ca@DMSNs-FA might act as a potential anticancer drug formulation for breast cancer therapy.

Abstract: Cancer is one of the fatal diseases in the history of humankind. In this regard, cancer immunotherapeutic strategies have revolutionized the traditional mode of cancer treatment. Silica based nano‐platforms have been extensively applied in nanomedicine including cancer immunotherapy. Mesoporous silica nanoparticles (MSN) and mesoporous organosilica nanoparticles (MON) are attractive candidates due to the ease in controlling the structural parameters as needed for the targeted immunotherapeutic applications. Especially, the MON provide an additional advantage of controlling the composition and modulating the biological functions to actively synergize with other immunotherapeutic strategies. In this review, the applications of MSN, MON, and metal‐doped MSN/MON in the field of cancer immunotherapy and tumor microenvironment regulation are comprehensively summarized by highlighting the structural and compositional attributes of the silica‐based nanoplatforms.

Abstract: The synthesis of ultrasmall, well-dispersed, hollow siliceous spheres (HSSs) by using a block copolymer as the template and tetraethoxysilane as a silica source under acidic conditions is reported. After removing the surfactant core of as-synthesized, spherical, silica-coated block-copolymer micelles, HSSs with a uniform particle size of 24.7 nm, a cavity diameter of 11.7 nm, and a wall thickness of 6.5 nm are obtained. It is shown that by surface functionalization of HSSs with methyl groups during synthesis, HSSs can be further dispersed in solvents such as water or ethanol to form a stable sol. Moreover, the hollow cavities are accessible for further loading of functional components. In addition, it is demonstrated that HSSs possess superior endocytosis properties for HeLa cells compared to those of conventional mesoporous silica nanoparticles. A feasible and designable strategy for synthesizing novel well-dispersed hollow structures with ultrasmall diameters instead of conventional ordered mesostructures is provided. It is expected that HSSs may find broad applications in bionanotechnology, such as drug carriers, cell imaging, and targeted therapy.

Synthesis of multi-functional large pore mesoporous silica nanoparticles as gene carriers

Abstract: The development of functional nanocarriers that can enhance the cellular delivery of a variety of nucleic acid agents is important in many biomedical applications such as siRNA therapy. We report the synthesis of large pore mesoporous silica nanoparticles (LPMSN) loaded with iron oxide and covalently modified by polyethyleneimine (denoted PEI-Fe-LPMSN) as carriers for gene delivery. The LPMSN have a particle size of ∼200 nm and a large pore size of 11 nm. The large pore size is essential for the formation of large iron oxide nanoparticles to increase the magnetic properties and the adsorption capacity of siRNA molecules. The magnetic property facilitates the cellular uptake of nanocarriers under an external magnetic field. PEI is covalently grafted on the silica surface to enhance the nanocarriers' affinity against siRNA molecules and to improve gene silencing performance. The PEI-Fe-LPMSN delivered siRNA-PLK1 effectively into osteosarcoma cancer cells, leading to cell viability inhibition of 80%, higher compared to the 50% reduction when the same dose of siRNA was delivered by a commercial product, oligofectamine.

Abstract: In the present work, a simple method was used to develop composite curcumin-amine functionalized mesoporous silica nanoparticles (MSN). The nanoparticles were used to improve the bioavailability of curcumin in mice through oral administration. We investigated the effect of particle size on the release profile, solubility and oral bioavailability of curcumin in mice, including amine functionalized mesoporous silica micron-sized-particles (MSM) and MSN (100-200 nm). Curcumin loaded within amine functionalized MSN (MSN-A-Cur) had a better release profile and a higher solubility compared to amine MSM (MSM-A-Cur). The bioavailability of MSN-A-Cur and MSM-A-Cur was considerably higher than that of 'free curcumin'. These results indicate promising features of amine functionalized MSN as a carrier to deliver low solubility drugs with improved bioavailability via the oral route.

On the Equilibrium of Helical Nanostructures with Ordered Mesopores

Abstract: Helical conformation exists universally at different length scales. We present a new model to explain the energetics of a helical structure with ordered mesopores and successfully predict their equilibrium state. The formation of the helical structure, which is composed of twisted and hexagonally arrayed one-dimensional pore channels, should be understood at the macromorphology level through the competition between surface free energy reduction and torsion strain energy increase. Our model is established by first reverting a helical rod with experimentally defined parameters to a conjectured straight rod without intrinsic pore channel twisting, and then quantitatively calculating the variation of two competitive energies as a function of twist angle in the torsion process starting from the reverted straight rod. Through our model, a free energy curve is achieved, so that the equilibrium state and the helical structural parameters can be predicted, which are in good agreement with experimental results for helical rods synthesized by different surfactant templates. Moreover, our model can be successfully applied to explain the pitch-radius relationships in previous observations. Our achievement provides unique and fundamental understandings for the spontaneous mesoscopic helix formation, which are different from the microscopic helical structures such as DNA chains.

Abstract: Intracellular delivery of proteins is a promising strategy of intervention in disease, which relies heavily on the development of efficient delivery platforms due to the cell membrane impermeability of native proteins, particularly for negatively charged large proteins. This work reports a vesicle supra-assembly approach to synthesize novel amine-functionalized hollow dendritic mesoporous silica nanospheres (A-HDMSN). An amine silica source is introduced into a water-oil reaction solution prior to the addition of conventional silica source tetraethylorthosilicate. A history of FMD research and control programmes in Southeast Asia: lessons from the past informing the future

Abstract: Foot and mouth disease (FMD) is a major animal health problem within Southeast Asia (SEA). Although Indonesia and more recently the Philippines have achieved freedom from FMD, the disease remains endemic on continental SEA. Control of FMD within SEA would increase access to markets in more developed economies and reduce lost productivity in smallholder and emerging commercial farmer settings. However, despite many years of vaccination by in idual countries, numerous factors have prevented the successful control of FMD within the region, including unregulated ‘informal’ transboundary movement of livestock and their products, difficulties implementing vaccination programmes, emergence of new virus topotypes and lineages, low-level technical capacity and biosecurity at national levels, limited farmer knowledge on FMD disease recognition, failure of timely outbreak reporting and response, and limitations in national and international FMD control programmes. This paper examines the published research of FMD in the SEA region, reviewing the history, virology, epidemiology and control programmes and identifies future opportunities for FMD research aimed at the eventual eradication of FMD from the region.

Quantum dots’ size matters for balancing their quantity and quality in label materials to improve lateral flow immunoassay performance for C-reactive protein determination

Abstract: Incorporating quantum dots (QDs) into dendritic mesoporous silica nanoparticles (DMSNs) for signal lification of label materials represents an efficient strategy to improve the performance of lateral flow immunoassays (LFIAs). In this work, it is found that the CdSe/ZnS QD's size matters for balancing their loading amount and quantum yields (QYs) in the DMSNs-QDs based label materials and ultimately determining the performance of LFIA. The impacts of three CdSe/ZnS QDs with diameters of 9.1, 10.5 and 11.7 nm on CdSe/ZnS QDs incorporation and LFIA applications are studied. The increase of CdSe/ZnS QDs size from 9.1 to 11.7 nm results in a decrease in CdSe/ZnS QDs loading amount and an increase in QYs of incorporated CdSe/ZnS QDs. Abstract: Development of veterinary subunit vaccines comes with a spectrum of challenges, such as the choice of adjuvant, antigen delivery vehicle, and optimization of dosing strategy. Over the years, our laboratory has largely focused on investigating silica vesicles (SVs) for developing effective veterinary vaccines for multiple targets. Rhipicephalus microplus (cattle tick) are known to have a high impact on cattle health and the livestock industry in the tropical and subtropical regions. Development of vaccine using Bm86 antigen against R. microplus has emerged as an attractive alternative to control ticks. In this study, we have investigated the biodistribution of SV in a live animal model, as well as further explored the SV ability for vaccine development. Rhodamine-labeled SV-140-C 18 (Rho-SV-140-C 18 ) vesicles were used to adsorb the Cy5-labeled R. microplus Bm86 antigen (Cy5-Bm86) to enable detection and characterization of the biodistribution of SV as well as antigen in vivo in a small animal model for up to 28 days using optical fluorescence imaging. We tracked the in vivo biodistribution of SVs and Bm86 antigen at different timepoints (days 3, 8, 13, and 28) in BALB/c mice. The biodistribution analysis by live imaging as well as by measuring the fluorescent intensity of harvested organs over the duration of the experiment (28 days) showed greater accumulation of SVs at the site of injection. The Bm86 antigen biodistribution was traced in lymph nodes, kidney, and liver, contributing to our understanding how this delivery platform successfully elicits antibody responses in the groups administered antigen in combination with SV. Selected tissues (skin, lymph nodes, spleen, kidney, liver, and lungs) were examined for any cellular abnormalities by histological analysis. No adverse effect or any other abnormalities were observed in the tissues.

Abstract: Photocatalytic oxygen reduction reaction (ORR) for H 2 O 2 production in the absence of sacrificing agents is a green approach and of great significance, where the design of photocatalysts with high performance is the central task. Herein, a spatial specific S‐scheme heterojunction design by introducing a novel semiconducting pair with a S‐scheme mechanism in a purpose‐designed Janus core–shell‐structured hollow morphology is reported. In this design, TiO 2 nanocrystals are grown inside the inner wall of resorcinol‐formaldehyde (RF) resin hollow nanocakes with a reverse bumpy ball morphology (TiO 2 @RF). The S‐scheme heterojunction preserves the high redox ability of the TiO 2 and RF pair, the spatial specific Janus design enhances the charge separation, promotes active site exposure, and reduces the H 2 O 2 decomposition to a large extent. The TiO 2 @RF photocatalyst shows a high H 2 O 2 yield of 66.6 mM g −1 h −1 and solar‐to‐chemical conversion efficiency of 1.11%, superior to another Janus structure (RF@TiO 2 ) with the same heterojunction but a reversed Janus spatial arrangement, and most reported photocatalysts under similar reaction conditions. The work has paved the way toward the design of next‐generation photocatalysts for green synthesis of H 2 O 2 production.

Structure Transition from Hexagonal Mesostructured Rodlike Silica to Multilamellar Vesicles

Abstract: We studied the synthesis of siliceous structures by using a nonionic block copolymer (Pluronic P123) and perfluorooctanoic acid (PFOA) as cotemplates in an acid-catalyzed sol-gel process. Different siliceous structures were obtained through systematically varying the molar ratio (R) of PFOA/P123, and the resultant materials were characterized by X-ray diffraction, scanning electron microscopy, transmission electron microscopy, nitrogen sorption analysis, and Fourier-transform infrared spectroscopy. The results are consistent and reveal a structure transition from a highly ordered 2D hexagonal (HEX) mesostructure with a rodlike morphology to multilamellar vesicles (MLVs) with sharp edges when R is increased. The fact that the MLVs are initiated from the end of hexagonally mesostructured rods provides key evidence in such a novel structure transition. Our finding indicates that, at least in our observations, the MLVs are developed gradually from HEX structures, rather than by a direct cooperative self-assembly mechanism. It is suggested that PFOA molecules with rigid fluorocarbon chains closely interact with PEO. This interaction model may well explain (1) the "wall-thicken" effect in HEX mesostructures by enlarging the hydrophilic PEO moiety (R = 0-1.4), (2) the subsequent HEX to multilamellar structure transition by modifying the hydrophilic/hydrophobic volume ratio (R = 1.4-2.8), and (3) the formation of MLVs with sharp edges by increasing the bending energy. This model provides insight into the fabrication of novel porous materials by the use of block copolymers and fluorinated surfactant mixed templates.

The large cone-shaped pores are formed by silica lamellae closely packed encircling a spherical core, showing a structure similar to the flower dahlia. MSN-CC has a large pore size of 45 nm and a high pore volume of 2.59 cm(3) g(-1). MSN-CC demonstrates a high loading capacity of large proteins and successfully delivers active β-galactosidase into cells, showing their potential as efficient nanocarriers for the cellular delivery of proteins with large molecular weights.

A Phospho-Directed Macroporous Alumina−Silica Nanoreactor with Multi-Functions

Abstract: A phospho-directed nanoreactor with multiple functions is reported. Alumina-functionalized macroporous ordered silica foams (Al-MOSF) have been developed with large pore size, high pore volume (1.6 cm(3)/g), and a surface area of 186 m(2)/g rich in coordination unsaturated Al species, which can be used as phospho-directed nanoreactors for integrated in situ digestion and in situ phosphoisolation. By directly adding Al-MOSF to the conventional in-solution digestion system, both enzymes and proteins are quickly enriched in the macropores of the reactor to achieve a fast proteolysis without increasing the enzyme rotein concentration or using a preimmobilization process, thus the digestion time and the cost can be greatly reduced. Meanwhile, due to the chemo-affinity between alumina and phosphor groups, the Al-MOSF reactor can in situ isolate specific products of the enzymatic reaction (i.e., phosphopeptides) and release the nonspecific peptides to the solution. This strategy is simple, efficient, and successfully applied in the detection of phosphoproteins in real s les.

Abstract: Infectious diseases caused by bacteria have led to a great threat to public health. With the significant advances in nanotechnology in recent decades, nanomaterials have emerged as a powerful tool to boost antibacterial performance due to either intrinsic bactericidal properties or by enhancing the delivery efficiency of antibiotics for effective pathogen killing. Vancomycin, as one of the most widely employed antimicrobial peptides, has a potent bactericidal activity, but at the same time shows a limited bioavailability. Silver nanoparticles have also been extensively explored and were found to have a well-recognized antibacterial activity and limited resistance potential however, how to prevent nanosized Ag particles from aggregation in biological conditions is challenging. In this study, we aimed to combine the advantages of both vancomycin and nano-Ag for enhanced bacterial killing, where both antibacterial agents were successfully loaded onto a silica nanoparticle with a pollen-like morphology. The morphology of nano-Ag-decorated silica nanopollens was characterized using transmission electron microscopy and elemental mapping through energy dispersive spectroscopy. Silver nanoparticles with a size of 10–25 nm were observed as well-distributed on the surface of silica nanoparticles of around 200 nm. The unique design of a spiky morphology of silica nano-carriers promoted the adhesion of nanoparticles towards bacterial surfaces to promote localized drug release for bacterial killing, where the bacterial damage was visualized through scanning electron microscopy. Enhanced bactericidal activity was demonstrated through this co-delivery of vancomycin and nano-Ag, decreasing the minimum inhibition concentration (MIC) towards E. coli and S. epidermidis down to 15 and 10 µg/mL. This study provides an efficient antimicrobial nano-strategy to address potential bacterial infections.

There are numerous reviews on mRNA delivery, but few focus on the underlying causes of mRNA instability and how to tackle the instability issues. Herein, the recent progress in nanobiotechnology-enabled strategies for stabilizing mRNA and better delivery is reviewed. First, factors that destabilize mRNA are introduced. Second, nanobiotechnology-enabled strategies to stabilize mRNA molecules are reviewed, including molecular and nanotechnology approaches. The impact of formulation processing on mRNA stability and shelf-life, including freezing and lyophilization, are also briefly discussed. Lastly, our perspectives on challenges and future directions are presented. This review may provide useful guidelines for understanding the structure–function relationship and the rational design of nanobiotechnology for mRNA stability enhancement and mRNA technology development.

Abstract: A solvothermal-assisted evaporation-induced self-assembly (SA-EISA) approach has been developed for the synthesis of ordered mesoporous alumina (OMA) materials with high thermal stability and improved performance in catalysis. In conventional EISA process, the evaporation step is accompanied by the hydrolysis of organic aluminum precursors, thus the evaporation conditions have significant influences on the reaction and the final structure of OMA. In our approach, the solvothermal treatment step promotes the complete hydrolysis of aluminum precursors and produces partially condensed cluster-like aluminum hydroxyl species, which allows the formation of OMA in a broad range of evaporation conditions. Compared to mesoporous alumina obtained by conventional EISA process, OMA materials prepared by SA-EISA approach exhibit higher specific surface area, pore volume and thermal stability. When used as supporting materials for vanadium oxide catalyst, OMA materials obtained by the SA-EISA approach exhibit excellent activity, selectivity and stability for ethylbenzene dehydrogenation with carbon dioxide as a mild oxidant. Our contribution has provided new understanding in the synthesis of OMA materials with improved performance for catalytic applications.

Abstract: Gene therapy has attracted much attention in modern society and provides a promising approach for treating genetic disorders, diseases and cancers. Safe and effective vectors are vital tools to deliver genetic molecules to cells. This review summarizes recent advances in the rational design of silica-based nanoparticles and their applications in gene therapy. An overview of different types of genetic agents available for gene therapy is provided. The engineering of various silica nanoparticles is described, which can be used as versatile complexation tools for genetic agents and advanced gene therapy. Several challenges are raised and future research directions in the area of gene therapy using silica-based nanoparticles are proposed.

Abstract: Hydrogen peroxide (H 2 O 2 ) production by electrochemical two‐electron water oxidation reaction (2e‐WOR) is a promising approach, where high‐performance electrocatalysts play critical roles. Here, the synthesis of nanostructured CaSnO 3 confined in conductive carbon fiber membrane with abundant oxygen vacancy (O V ) as a new generation of 2e‐WOR electrocatalyst is reported. The CaSnO 3 @carbon fiber membrane can be directly used as a self‐standing electrode, exhibiting a record‐high H 2 O 2 production rate of 39.8 µmol cm −2 min −1 and a selectivity of ≈90% (at 2.9 V vs reversible hydrogen electrode). The CaSnO 3 @carbon fiber membrane design improves not only the electrical conductivity and stability of catalysts but also the inherent activity of CaSnO 3 . Density functional theory calculation further indicates the crucial role of O V in increasing the adsorption free energy toward oxygen intermediates associated with the competitive four‐electron water oxidation reaction pathway, thus enhancing the activity and selectivity of 2e‐WOR. The findings pave a new avenue to the rational design of electrocatalysts for H 2 O 2 production from water.

Abstract: This study systematically investigates periodic mesoporous organosilicas (PMOs) with controlled helical and concentric circular (CC) pore architectures prepared through a basic-catalyzed sol-gel process by using an achiral cationic surfactant trimethyloctadecylammonium bromide (C(18)TAB) as a structure-directing agent, perfluorooctanoic acid (PFOA) as an additive, and 1,2-bis(triethoxysilyl)ethane (BTEE) as a hybrid silica precursor. By increasing the weight ratio of PFOA/C(18)TAB, a pore architecture transition of PMO materials from hexagonal-arrayed, straight longitudinal channels to helical and CC mesostructures is achieved such a transition has not been observed before in PMO materials. Our discovery is helpful in understanding the supramolecular cooperative assembly of hybrid materials and their structural and morphological evolution, which are important in the future applications of PMO materials.

Abstract: Spotted fever group rickettsiae (SFG) are a neglected group of bacteria, belonging to the genus Rickettsia , that represent a large number of new and emerging infectious diseases with a worldwide distribution. The diseases are zoonotic and are transmitted by arthropod vectors, mainly ticks, fleas and mites, to hosts such as wild animals. Domesticated animals and humans are accidental hosts. In Asia, local people in endemic areas as well as travellers to these regions are at high risk of infection. In this review we compare SFG molecular and serological diagnostic methods and discuss their limitations. While there is a large range of molecular diagnostics and serological assays, both approaches have limitations and a positive result is dependent on the timing of s le collection. There is an increasing need for less expensive and easy-to-use diagnostic tests. However, despite many tests being available, their lack of suitability for use in resource-limited regions is of concern, as many require technical expertise, expensive equipment and reagents. In addition, many existing diagnostic tests still require rigorous validation in the regions and populations where these tests may be used, in particular to establish coherent and worthwhile cut-offs. It is likely that the best strategy is to use a real-time quantitative polymerase chain reaction (qPCR) and immunofluorescence assay in tandem. If the specimen is collected early enough in the infection there will be no antibodies but there will be a greater chance of a PCR positive result. Conversely, when there are detectable antibodies it is less likely that there will be a positive PCR result. It is therefore extremely important that a complete medical history is provided especially the number of days of fever prior to s le collection. More effort is required to develop and validate SFG diagnostics and those of other rickettsial infections.

Abstract: An intriguing evolution from a simple internal helix to a hierarchical helical (HH) mesostructure with both internal and external helices or a complicated screwlike and concentric circular (CC) mesostructure is successfully observed. The complicated helical structures are determined by TEM studies and 3D electron tomography. We demonstrate a topological helix-coil transition between the internal and external helices to reveal the origin of the HH mesostructure and the relationship between the straight helical and HH rods. Moreover, the boundary condition of the helix-coil transition is clarified to explain in detail the formation of complex helical structures, such as the screwlike mesostructure. It is proposed that the final structural characteristics are determined exactly by the balance between the decrease in the surface free energy and the maintenance of the hexagonal packing in one in idual rod, which explains the formation of unusual CC, HH, and screwlike morphologies in one pot. Our success has opened new opportunities in the characterization of complex porous architectures, thus paving a way to remarkable advances in the fields of synthesis, understanding, and application of novel porous materials.

Biosafety and biosecurity requirements for Orientia spp. diagnosis and research: Recommendations for risk-based biocontainment, work practices and the case for reclassification to risk group 2

Abstract: Scrub typhus is an important arthropod-borne disease causing significant acute febrile illness by infection with Orientia spp . Using a risk-based approach, this review examines current practice, the evidence base and regulatory requirements regarding matters of biosafety and biosecurity, and presents the case for reclassification from Risk Group 3 to Risk Group 2 along with recommendations for safe working practices of risk-based activities during the manipulation of Orientia spp. in the laboratory. We recommend to reclassify Orientia spp. to Risk Group 2 based on the classification for RG2 pathogens as being moderate in idual risk, low community risk. We recommend that low risk activities, can be performed within a biological safety cabinet located in a Biosafety Level (BSL) 2 core laboratory using standard personal protective equipment. But when the risk assessment indicates, such as high concentration and volume, or aerosol generation, then a higher biocontainment level is warranted. For, the majority of animal activities involving Orientia spp., Animal BSL 2 (ABSL2) is recommended however where high risk activities are performed including necropsies, Animal BSL (ABSL3) is recommended.

Abstract: Assembly of different metal-organic framework (MOF) building blocks into hybrid MOF-on-MOF heterostructures is promising in chemistry and materials science, however the development of ternary MOF-on-MOF heterostructures with controllable architectural and compositional complexity is challenging. Here we report the synthesis of three types of ternary MOF-on-MOF heterostructures via a multiple selective assembly strategy. This strategy relies on the choice of one host MOF with more than one facet that can arrange the growth of a guest MOF, where the arrangement is site-selective without homogenous growth of guest MOF or homogenous coating of guest on host MOF. The growth of guest MOF on a selected site of host MOF in each step provides the opportunity to further vary the combinations of arrangements in multiple steps, leading to ternary MOF-on-MOF heterostructures with tunable complexity. The developed strategy paves the way towards the rational design of intricate and unprecedented MOF-based superstructures for various applications.

Abstract: A new and facile approach has been developed to prepare monodisperse mesoporous silica nanospheres (MMSNs) with controlled particle sizes and pore structures. In our approach, MMSNs were synthesized simply in a sodium acetate solution without adding any other alkali or alcohol additives. MMSNs have a spherical shape and uniform particle sizes, which can be adjusted from 50 to 110 nm by increasing the reaction temperature from 40 to 80 °C. By performing a subsequent hydrothermal treatment (HT) under basic condition (pH=~11.5) at 130 °C, the mesoporous pore volume and surface area can be enhanced, while keeping the mono-dispersion characteristics and the mesopore size almost unchanged. The pore sizes of MMSNs can be adjusted from 2.8 to 4.0 nm under acidic solutions by changing the HT temperature from 100 to 130 °C. The formation process of MMSNs has been investigated by transmission electron microscopy (TEM) and attenuated total reflection Fourier transform infrared (ATR-FTIR) techniques. A spherical micelle templating mechanism is proposed to explain the formation of MMSNs in our system, which is different from that of traditional highly ordered mesoporous silica nanoparticles (MCM-41).

Abstract: We developed an intradermal (ID) challenge cynomolgus macaque (Macaca fascicularis) model of scrub typhus, the leading cause of treatable undifferentiated febrile illness in tropical Asia, caused by the obligate intracellular bacterium, Orientia tsutsugamushi. A well-characterized animal model is required for the development of clinically relevant diagnostic assays and evaluation of therapeutic agents and candidate vaccines. We investigated scrub typhus disease pathophysiology and evaluated two O. tsutsugamushi 47-kDa, Ag-based candidate vaccines, a DNA plasmid vaccine (pKarp47), and a virus-vectored vaccine (Kp47/47-Venezuelan equine encephalitis virus replicon particle) for safety, immunogenicity, and efficacy against homologous ID challenge with O. tsutsugamushi Karp. Control cynomolgus macaques developed fever, classic eschars, lymphadenopathy, bacteremia, altered liver function, increased WBC counts, pathogen-specific Ab (IgM and IgG), and cell-mediated immune responses. Vaccinated macaques receiving the DNA plasmid pKarp47 vaccine had significantly increased O. tsutsugamushi–specific, IFN-γ–producing PBMCs (p = 0.04), reduced eschar frequency and bacteremia duration (p ≤ 0.01), delayed bacteremia onset (p & 0.05), reduced circulating bacterial biomass (p = 0.01), and greater reduction of liver transaminase levels (p & 0.03) than controls. This study demonstrates a vaccine-induced immune response capable of conferring sterile immunity against high-dose homologous ID challenge of O. tsutsugamushi in a nonhuman primate model, and it provides insight into cell-mediated immune control of O. tsutsugamushi and dissemination dynamics, highlights the importance of bacteremia indices for evaluation of both natural and vaccine-induced immune responses, and importantly, to our knowledge, has determined the first phenotypic correlates of immune protection in scrub typhus. We conclude that this model is suitable for detailed investigations into vaccine-induced immune responses and correlates of immunity for scrub typhus.

Abstract: Resoles are resins obtained by base-catalyzed phenol-formaldehyde condensation with a three-dimensional cross-linked framework. They are considered as highly chemical-resistant, and calcination is thus generally used in the treatment of resole-type resins, which significantly limits the ersity of nanostructured materials that can be derived from resole-type resins. Herein, we report that selected metal nitrate solutions can be used to dissolve various types of nanostructured resoles through an oxidative dissolution process. This strategy not only enables the controlled dissolution of resoles, but more importantly provides a new approach to selectively etch resole-based nanocomposites to give rise to a variety of nanostructured materials with unprecedented architectures and great potential in bioapplications.

Abstract: Nanotechnology increasingly plays a significant role in vaccine development. As vaccine development orientates toward less immunogenic "minimalist" compositions, formulations that boost antigen effectiveness are increasingly needed. The use of nanoparticles in vaccine formulations allows not only improved antigen stability and immunogenicity, but also targeted delivery and slow release. A number of nanoparticle vaccines varying in composition, size, shape, and surface properties have been approved for human use and the number of candidates is increasing. However, challenges remain due to a lack of fundamental understanding regarding the in vivo behavior of nanoparticles, which can operate as either a delivery system to enhance antigen processing and/or as an immunostimulant adjuvant to activate or enhance immunity. This review provides a broad overview of recent advances in prophylactic nanovaccinology. Types of nanoparticles used are outlined and their interaction with immune cells and the biosystem are discussed. Increased knowledge and fundamental understanding of nanoparticle mechanism of action in both immunostimulatory and delivery modes, and better understanding of in vivo biodistribution and fate, are urgently required, and will accelerate the rational design of nanoparticle-containing vaccines.

Curcumin-cyclodextrin encapsulated chitosan nanoconjugates with enhanced solubility and cell cytotoxicity

Abstract: Curcumin (CUR), a naturally derived anti-cancer cocktail is arguably the most widely studied neutraceutical. Despite a lot of promises, it is yet to reach the market as an active anti-cancer formulation. In the present study, we have prepared highly soluble (3 mg/ml) CUR-γ-hydroxypropyl cyclodextrin (CUR-CD) hollow spheres. CUR-CD hollow spheres were prepared by a novel and scalable spray drying method. CUR-CD was then encapsulated into positively charged biodegradable chitosan (CUR-CD-CS) nanoparticles. The CUR-CD-CS nanoparticles were characterised by TEM, SEM, DLS, drug loading and in vitro release. We tested the efficacy of these CUR-CD-CS nanoparticles in SCC25 cell lines using MTT assay and investigated its cellular uptake mechanism. We also studied Oligo DNA loading in CUR-CD-CS nanoparticles and its delivery via confocal imaging and FACS analysis. Our results demonstrated that CUR-CD-CS nanoparticles showed superior in vitro release performance and higher cytotoxicity in SCC25 cell line amongst all tested formulations. The cytotoxicity results were corroborated by cell cycle analysis and apoptosis test, showing nearly 100% apoptotic cell death in the case of CUR-CD-CS nanoparticles. Compared to CS nanoparticles, CS-CD nanoformulation showed higher cellular delivery of Cy3-Oligo DNA which was tested quantitatively using flowcytometry analysis, indicating that CD not only enhanced CUR solubility but also boosted the cellular uptake. Our study shows that rationally designed bio-degradable natural biomaterials have great potential as next generation nano-carriers for hydrophobic drug delivery such as CUR with potential of dual drug-gene delivery.

Influence of Novel Nano-Mesoporous Bioactive Glass on the Regulation of IGF-II Gene Expression in Osteoblasts

Abstract: To investigate whether the mesoporous bioactive glass (MBG) exerts any in vitro bioactivity on rat osteoblasts and the potential regulatory mechanism of this bioactivity. Rat osteoblasts were incubated in the presence and absence (control) of ionic dissolution product of MBG in minimal essential medium. The osteoblast proliferation and differentiation were measured using MTT and alkaline phosphatase methods. The IGF-II mRNA expression in osteoblasts was measured by RT-PCR. IGF-II protein and IGFBP were measured by ELISA. The level of alkaline phosphatase activity was increased to 125% of control. Expression of IGF-II mRNA was increased to 125% of control. There was a 175 and 237% increase in the concentration of unbound IGF-II protein and IGFBP, respectively, in the conditioned media of treated osteoblasts. The osteoblast proliferation was 92% of control. The ionic dissolution product of MBG was able to promote the differentiation of osteoblasts, probably by inducing IGF-II expression at both mRNA and protein level.

The Development of an Abattoir-Based Surveillance System in Lao PDR for the Detection of Zoonoses in Large Ruminants: Q Fever and Brucellosis Seroepidemiology as a Pilot Study

Abstract: Although animal health surveillance programmes are useful for gaining information to help improve global health and food security, these programmes can be challenging to establish in developing economies with a low-resource base. This study focused on establishing a national surveillance system initiated by the Lao PDR government using a passive surveillance system of abattoir s les as a pilot model, and to gain information on contagious zoonoses, particularly Q fever and brucellosis, in the large ruminant population. A total of 683 cattle and buffalo s les were collected from six selected provinces of Lao PDR between March–December 2019. Out of 271 s les tested, six s les (2.2%, 95% confidence interval (CI) of 1.0, 4.8) were positive in the Q fever antibody ELISA test. Only one s le (out of 683 0.2%, 95% CI 0.0, 0.8) tested positive to the Brucella antibody ELISA test. Seroprevalence of these important zoonoses in Lao PDR were relatively low in cattle and buffaloes however, extensive animal movement within the country was identified which could increase risks of spreading transboundary diseases. The study highlights the importance of ongoing animal health surveillance and the need to find cost-effective approaches for its long-term sustainability.

Abstract: Enzymes play an indispensable role in biosystems, catalyzing a variety of chemical and biochemical reactions with exceptionally high efficiency and selectivity. These features render them uniquely positioned in developing novel catalytic systems and therapeutics. However, their practical application is largely hindered by the vulnerability, low reusability and the inability to overcome the biological barriers of enzymes. Silica‐based nanoparticles (SNPs) are a classic family of nanomaterials with tunable physicochemical properties, making them ideal candidates to address the intrinsic shortcomings of natural enzymes. SNPs not only improve the activity and durability of enzymes, but also provide precise spatiotemporal control over their intracellular as well as systemic biodistributions for boosting the catalytic outcome. Herein, the recent progress in SNPs for enzyme immobilization and delivery is summarized. The therapeutic applications, including cancer therapy and bacterial inhibition, are particularly highlighted. Our perspectives in this field, including current challenges and possible future research directions are also provided.

Poly-l-lysine Functionalized Large Pore Cubic Mesostructured Silica Nanoparticles as Biocompatible Carriers for Gene Delivery

Abstract: Large pore mesoporous silica nanoparticles (LP-MSNs) functionalized with poly-L-lysine (PLL) were designed as a new carrier material for gene delivery applications. The synthesized LP-MSNs are 100-200 nm in diameter and are composed of cage-like pores organized in a cubic mesostructure. The size of the cavities is about 28 nm with an entrance size of 13.4 nm. Successful grafting of PLL onto the silica surface through covalent immobilization was confirmed by X-ray photoelectron spectroscopy, solid-state (13)C magic-angle spinning nuclear magnetic resonance, Fourier transformed infrared, and thermogravimetric analysis. As a result of the particle modification with PLL, a significant increase of the nanoparticle binding capacity for oligo-DNAs was observed compared to the native unmodified silica particles. Consequently, PLL-functionalized nanoparticles exhibited a strong ability to deliver oligo DNA-Cy3 (a model for siRNA) to Hela cells. Furthermore, PLL-functionalized nanoparticles were proven to be superior as gene carriers compared to amino-functionalized nanoparticles and the native nanoparticles. The system was tested to deliver functional siRNA against minibrain-related kinase and polo-like kinase 1 in osteosarcoma cancer cells. Here, the functionalized particles demonstrated great potential for efficient gene transfer into cancer cells as a decrease of the cellular viability of the osteosarcoma cancer cells was induced. Moreover, the PLL-modified silica nanoparticles also exhibit a high biocompatibility, with low cytotoxicity observed up to 100 μg/mL.

Abstract: The geometry of nanoparticles has a profound effect on their interactions with macrophages. For an elongated geometry, the well-known curvature-dependent phagocytosis mechanism is still under debate, presumably because another important parameter, the probability of orientation, is overlooked. To verify this hypothesis, it is demonstrated that increasing the probability of the preferred vertical orientation is an efficient strategy to significantly enhance macrophage phagocytosis and uptake. This is achieved via a well-designed hexapod nanoparticle in comparison with a monopod counterpart. The hexapod nanoparticle can achieve ≈100% close-to-vertical orientation, thereby favoring phagocytosis. This discovery provides a new insight into the design of nanomaterials for macrophage-oriented bioapplications.

A Bioinspired Route to Various Siliceous Vesicular Structures

Abstract: Various siliceous nanostructures have been successfully synthesized through the co-organization of organic molecules and inorganic silica source under mild pH conditions (pH approximately 5). A biodegradable block copolymer P123 [EO20PO70EO20, EO is poly (ethylene oxide), PO is poly (propylene oxide)] is employed as a marcomolecular template and Na2SiO39H2O as a silica source. By changing the concentrations of the reactants and/or reaction temperature, siliceous multilamellar vesicles, unilamellar nano-foams and multilamellar vesicles with sponge-like walls have been obtained. Our work provides a convenient and bioinspired route to obtain siliceous nanostructured materials with adjustable and multi-level pore structures as well as rich morphologies, which is important to understand the biomineralization mechanism. Such artificial silica nanoporous materials may find potential applications in catalysis, separations, electronics, and photonics, etc.

Abstract: Nano‐immunotherapy has been recognized as a highly promising strategy for cancer treatment in recent decades, which combines nanotechnology and immunotherapy to combat against tumors. Hybrid nanomaterials consisting of at least two constituents with distinct compositions and properties, usually organic and inorganic, have been engineered with integrated functions and enormous potential in boosting cancer immunotherapy. This review provides a summary of hybrid nanomaterials reported for cancer immunotherapy, including nanoscale metal–organic frameworks, metal–phenolic networks, mesoporous organosilica nanoparticles, metallofullerene nanomaterials, polymer–lipid, and biomacromolecule‐based hybrid nanomaterials. The combination of immunotherapy with chemotherapy, chemodynamic therapy, radiotherapy, radiodynamic therapy, photothermal therapy, photodynamic therapy, and sonodynamic therapy based on hybrid nanomaterials is also discussed. Finally, the current challenges and the prospects for designing hybrid nanomaterials and their application in cancer immunotherapy are outlined.

Abstract: Inhibiting the formation of new tumor blood vessels (so-called antiangiogenesis) and obstructing the established ones are two primary strategies in tumor vasculature targeted therapy. However, the therapeutic outcome of conventional methodologies relying on only one mechanism is rather limited. Herein, the first ex le of ultrasmall responsively aggregatable nanochelators that can intrinsically fulfill both antivasculature functions as well as high renal clearable efficiency is introduced. The nanochelators with sub-6 nm sizes exhibit not only systemic copper depletion activity for tumor antiangiogenesis but also, more surprisingly, the capability to transform from a "dispersed" state to an "aggregated" state to form large secondary particles in response to tumor microenvironment with elevated copper and phosphate levels for blood vessel obstruction. Compared to a benchmark antiangiogenic agent that can only inhibit the formation of tumor blood vessels, the nanochelators with unprecedented synergistic functions demonstrate significantly enhanced tumor inhibition activity in both breast cancer and colon cancer tumor models. Moreover, these ultrasmall nanochelators are noncytotoxic and renal clearable, ensuring superior biocompatibility. It is envisaged that the design of nanomaterials with ground-breaking properties and the synergistic antivasculature functions would offer a substantial conceptual advance for tumor vasculature targeted therapy and may provide vast opportunities for developing advanced nanomedicines.
