Abstract: Chlamydiosis, caused by Chlamydia pecorum, is regarded as an important threat to koala populations. Across the koala's geographical range, disease severity associated with C. pecorum infection varies, with pathogen ersity and strain pathogenicity being likely important factors. To examine C. pecorum ersity on a sub-population level a Multi-Locus Sequence Typing (MLST) scheme, containing the housekeeping genes gatA, oppA_3, hflX, gidA, enoA, hemN and fumC, was used to type strains from two sub-populations of koalas from the Liverpool Plains, NSW, Australia, with different disease expressions. Typing of s les from 2015 to 2017, revealed a significant association between sequence type ST 69 and clinical disease and a significant difference in sequence type frequencies between sub-populations. Sequence type ST 69 has previously been identified in both subclinical and clinically diseased koalas indicating that these markers alone are not illustrative of pathogenicity. However, recent emergence of this sequence type in a naïve population may explain the differing disease expressions. Sequence types ST 73 and ST 69 have been described in koalas across a broad geographic range, indicating multiple introduction events and/or a limited veracity of the MLST loci to explore fine scale epidemiological investigations, particularly those examining the interface between pathogenic strain and disease outcome.

Abstract: The philosopher Daniel Dennett argues that complex structures in the natural and cultural worlds emerge from two types of design. Bottom-up design involves the rote action of a simple algorithm in an environment constrained by physical laws. Top-down design involves deliberation and planning, and is unique to modern humans. Identifying the emergence of top-down design in the hominin lineage is an important research challenge, and the archaeological record of stone technology is our best evidence for it. A current view is that artefact types and flaking methods increased in complexity from 3.3 to c. 0.3 million years ago, reflecting improving capacities at spatial cognition and working memory, culminating in top-down design perhaps as early as 1.75 million years ago. Recent experimental work, however, has shown that a simple ‘remove flake’ algorithm constrained by the laws of fracture mechanics—a form of bottom-up design—can produce stone tool attributes thought to be evidence of top-down design. Here, these models are reviewed and critiqued in light of the new experimental evidence. A revised working memory-based model, focusing on the recursive aspects of stone flaking, is proposed.

Abstract: Sulawesi is the largest and oldest island within Wallacea, a vast zone of oceanic islands separating continental Asia from the Pleistocene landmass of Australia and Papua (Sahul). By one million years ago an unknown hominin lineage had colonized Flores immediately to the south, and by about 50 thousand years ago, modern humans (Homo sapiens) had crossed to Sahul. On the basis of position, oceanic currents and biogeographical context, Sulawesi probably played a pivotal part in these dispersals. Uranium-series dating of speleothem deposits associated with rock art in the limestone karst region of Maros in southwest Sulawesi has revealed that humans were living on the island at least 40 thousand years ago (ref. 5). Here we report new excavations at Talepu in the Walanae Basin northeast of Maros, where in situ stone artefacts associated with fossil remains of megafauna (Bubalus sp., Stegodon and Celebochoerus) have been recovered from stratified deposits that accumulated from before 200 thousand years ago until about 100 thousand years ago. Our findings suggest that Sulawesi, like Flores, was host to a long-established population of archaic hominins, the ancestral origins and taxonomic status of which remain elusive.

Abstract: Cryptococcosis, caused by environmental fungi in the Cryptococcus neoformans and Cryptococcus gattii species complexes, affects a variety of hosts, including koalas (Phascolarctos cinereus). Cryptococcal antigenemia and nasal colonization are well characterized in captive koalas, but free-ranging populations have not been studied systematically. Free-ranging koalas (181) from the Liverpool Plains region of New South Wales, Australia, were tested for cryptococcal antigenemia (lateral flow immunoassay) and nasal colonization (bird seed agar culture). Results were related to environmental and in idual koala characteristics. Eucalypt trees (14) were also randomly tested for the presence of Cryptococcus spp. by bird seed agar culture. In sum, 5.5% (10/181) and 6.6% (12/181) of koalas were positive for antigenemia and nasal colonization, respectively, on at least one occasion. And 64.3% (9/14) of eucalypts were culture-positive for Cryptococcus spp. URA5 restriction fragment length polymorphism analysis identified most isolates as C. gattii VGI, while C. neoformans VNI was only found in one koala and one tree. Colonized koalas were significantly more likely to test positive for antigenemia. No associations between antigenemia or colonization, and external environmental characteristics (the relative abundance of Eucalyptus camaldulensis and season), or in idual koala characteristics (body condition, sex, and age), could be established, suggesting that antigenemia and colonization are random outcomes of host-pathogen-environment interactions. The relationship between positive antigenemia status and a relatively high abundance of E. camaldulensis requires further investigation. This study characterizes cryptococcosis in a free-ranging koala population, expands the ecological niche of the C. gattii/C. neoformans species complexes and highlights free-ranging koalas as important sentinels for this disease.

Abstract: The eusporangiate marattialean ferns represent an ancient radiation with a rich fossil record but limited modern ersity in the tropics. The long evolutionary history without close extant relatives has confounded studies of the phylogenetic origin, rooting and timing of marattialean ferns. Here we present new complete plastid genomes of six marattialean species and compiled a plastid genome dataset representing all of the currently accepted marattialean genera. We further supplemented this dataset by compiling a large dataset of mitochondrial genes and a phenotypic data matrix covering both extant and extinct representatives of the lineage. Our phylogenomic and total-evidence analyses corroborated the postulated position of marattialean ferns as the sister to leptosporangiate ferns, and the position of Danaea as the sister to the remaining extant marattialean genera. However, our results provide new evidence that Christensenia is sister to Marattia and that M. cicutifolia actually belongs to Eupodium. The apparently highly reduced rate of molecular evolution in marattialean ferns provides a challenge for dating the key phylogenetic events with molecular clock approaches. We instead applied a parsimony-based total-evidence dating approach, which suggested a Triassic age for the extant crown group. The modern distribution can best be explained as mainly resulting from vicariance following the breakup of Pangaea and Gondwana. We resolved the fossil genera Marattiopsis, Danaeopsis and Qasimia as members of the monophyletic family Marattiaceae, and the Carboniferous genera Sydneia and Radstockia as the monophyletic sister of all other marattialean ferns.

Abstract: In an effort to identify novel drugs targeting fusion-oncogene–induced acute myeloid leukemia (AML), we performed high-resolution proteomic analysis. In AML1-ETO (AE)-driven AML, we uncovered a deregulation of phospholipase C (PLC) signaling. We identified PLCgamma 1 (PLCG1) as a specific target of the AE fusion protein that is induced after AE binding to intergenic regulatory DNA elements. Genetic inactivation of PLCG1 in murine and human AML inhibited AML1-ETO dependent self-renewal programs, leukemic proliferation, and leukemia maintenance in vivo. In contrast, PLCG1 was dispensable for normal hematopoietic stem and progenitor cell function. These findings are extended to and confirmed by pharmacologic perturbation of Ca++-signaling in AML1-ETO AML cells, indicating that the PLCG1 pathway poses an important therapeutic target for AML1-ETO+ leukemic stem cells.

Abstract: Australia was colonized by at least 40,000 bp and scientists agree that the continent was only ever occupied by anatomically and behaviourally modern humans. Australia thus offers an alternative early record for the archaeological expression of behavioural modernity. This review finds that the pattern of change in the Australian archaeological sequence bears remarkable similarity to the pattern from the Lower to Upper Palaeolithic in the Old World, a finding that is inconsistent with the ‘symbolic revolution’ model of the origin of modern behaviour. This highlights the need for archaeologists to rethink the implications of the various criteria and scales of analysis used to identify modern human behaviour.

Abstract: This study examines trends in stone tool reduction technology at Liang Bua, Flores, Indonesia, where excavations have revealed a stratified artifact sequence spanning 95k.yr. The reduction sequence practiced throughout the Pleistocene was straightforward and unchanging. Large flakes were produced off-site and carried into the cave where they were reduced centripetally and bifacially by four techniques: freehand, burination, truncation, and bipolar. The locus of technological complexity at Liang Bua was not in knapping products, but in the way techniques were integrated. This reduction sequence persisted across the Pleistocene/Holocene boundary with a minor shift favoring unifacial flaking after 11ka. Other stone-related changes occurred at the same time, including the first appearance of edge-glossed flakes, a change in raw material selection, and more frequent fire-induced damage to stone artifacts. Later in the Holocene, technological complexity was generated by "adding-on" rectangular-sectioned stone adzes to the reduction sequence. The Pleistocene pattern is directly associated with Homo floresiensis skeletal remains and the Holocene changes correlate with the appearance of Homo sapiens. The one reduction sequence continues across this hominin replacement.

Abstract: Excavations at Saruq al-Hadid, Dubai, UAE, discovered a stone tool technology with backed microliths dating to the Wadi Suq period and Late Bronze Age (ca. 1750–1300 BCE). The stone technology is a contemporary with metal production in the region, and the assemblage was recovered from a thick bone midden deposit at this multi-period site on the edge of the Rub’ al-Khali Desert. Small cobbles of chert were imported to the site and were reduced into flakes by hard-hammer percussion. Cores were frequently rotated during knapping and the reduction strategy was ad hoc, lacking hierarchical reduction stages. Flake tools were used as-is or modified by retouching. Some flakes were selected for backing into geometric microliths, and backing techniques often reflected high levels of stoneworking skill to produce stylised scalene shapes. A review of contemporary archaeological evidence, and the context of the Saruq al-Hadid assemblage, suggest that microliths may have been made as stone armatures for arrows despite the contemporary use of copper-based arrowheads.

Abstract: The archaeology of Sulawesi is important for developing an understanding of human dispersal and occupation of central Island Southeast Asia. Through over a century of archaeological work, multiple human populations in the southwestern region of Sulawesi have been identified, the most well-documented being that of the Mid- to Late Holocene ‘Toalean’ technological period. Archaeological models for this period describe a population with a strong cultural identity, sub ided into groups living on the coastal plains around Maros as well as dispersed upland forest dwellers, hunting endemic wildlife with bow-and-arrow technology. It has been proposed that the Toaleans were capable of vast water-crossings, with possible cultural exchange with northern Australia, Java, and Japan. This Toalean paradigm is built almost exclusively on existing interpretations of distinctive Toalean stone and bone artefact technologies, constructed on out-dated 19 th and 20 th century theory. Moreover, current definitions of Toalean artefact types are inconsistently applied and unsystematic, and the manufacturing sequence has historically been poorly understood. To address these problems in existing artefact models and typologies, we present a clarified typology of the Toalean artefacts of South Sulawesi, and describe the technical aspects of artefact production. This typology provides a tool for standardising research and will facilitate more meaningful assessments of material culture repertoires and more reliable assessment of spatial and temporal changes for the region.

Abstract: We present evidence from the Late Pleistocene of Sulawesi, Indonesia, where an unusually rich and unique symbolic complex was excavated from archaeological deposits spanning 30,000 to 22,000 y ago. Including previously unknown practices of self-ornamentation, used ochre, pigmented artifacts, and portable art, these findings advance our knowledge of the cultural repertoires of modern humans in Pleistocene Wallacea, including the nonparietal artworks and symbolic material culture of some of the world’s earliest known “cave artists.”

Abstract: Recent excavations at the early Middle Pleistocene site of Mata Menge in the So'a Basin of central Flores, Indonesia, have yielded hominin fossils attributed to a population ancestral to Late Pleistocene Homo floresiensis. Here we describe the age and context of the Mata Menge hominin specimens and associated archaeological findings. The fluvial sandstone layer from which the in situ fossils were excavated in 2014 was deposited in a small valley stream around 700 thousand years ago, as indicated by (40)Ar/(39)Ar and fission track dates on stratigraphically bracketing volcanic ash and pyroclastic density current deposits, in combination with coupled uranium-series and electron spin resonance dating of fossil teeth. Palaeoenvironmental data indicate a relatively dry climate in the So'a Basin during the early Middle Pleistocene, while various lines of evidence suggest the hominins inhabited a savannah-like open grassland habitat with a wetland component. The hominin fossils occur alongside the remains of an insular fauna and a simple stone technology that is markedly similar to that associated with Late Pleistocene H. floresiensis.

Abstract: Stone tools have a continuous record extending some 3.3 million years. Our hominin ancestors engaged in relatively simple stone flaking, and stone tools of extreme complexity were produced by cognitively modern humans in the Pleistocene and Holocene. For this reason, stone tools offer a tangible means for tracking the evolution of cognition in our genus. This chapter discusses a recent series of experiments controlled for modern flintknapper intent, the results suggesting that aspects of ancient tool forms sometimes viewed as deliberate can in fact be produced with no more intention than that seen in the removal of in idual flakes. But the removal of in idual flakes is itself a cognitively challenging task, one that places the earliest hominin flintknappers across the “cognitive Rubicon” from their primate relatives.

Abstract: Cryptococcosis caused by yeasts of the Cryptococcus gattii species complex is an increasingly important mycological disease in humans and other mammals. In Australia, cases of C. gattii-related cryptococcosis are more prevalent in the koala (Phascolarctos cinereus) compared to humans and other animals, likely due to the close association that both C. gattii and koalas have with Eucalyptus species. This provides a cogent opportunity to investigate the epidemiology of spontaneous C. gattii infections in a free-living mammalian host, thereby offering insights into similar infections in humans. This study aimed to establish a link between nasal colonisation by C. gattii in free-ranging koalas and the tree hollows of Eucalyptus species, the key environmental source of the pathogen. We (i) detected and genotyped C. gattii from nine out of 169 free-ranging koalas and representative tree hollows within their home range in the Liverpool Plains, New South Wales, and (ii) examined potential environmental predictors of nasal colonisation in koalas and the presence of C. gattii in tree hollows. Phylogenetic analyses based on multi-locus sequence typing (MLST) revealed that the koalas were most likely colonised by the most abundant C. gattii genotypes found in the Eucalyptus species, or closely related genotypes. Importantly, the likelihood of the presence of C. gattii in tree hollows was correlated with increasing hollow size.

Abstract: Excavations at Liang Bua, a large limestone cave on the island of Flores in eastern Indonesia, have yielded evidence for a population of tiny hominins, sufficiently distinct anatomically to be assigned to a new species, Homo floresiensis. The finds comprise the cranial and some post-cranial remains of one in idual, as well as a premolar from another in idual in older deposits. Here we describe their context, implications and the remaining archaeological uncertainties. Dating by radiocarbon (14C), luminescence, uranium-series and electron spin resonance (ESR) methods indicates that H. floresiensis existed from before 38,000 years ago (kyr) until at least 18 kyr. Associated deposits contain stone artefacts and animal remains, including Komodo dragon and an endemic, dwarfed species of Stegodon. H. floresiensis originated from an early dispersal of Homo erectus (including specimens referred to as Homo ergaster and Homo georgicus) that reached Flores, and then survived on this island refuge until relatively recently. It overlapped significantly in time with Homo sapiens in the region, but we do not know if or how the two species interacted.

Abstract: In the Soa Basin of central Flores, eastern Indonesia, stratified archaeological sites, including Mata Menge, Boa Lesa and Kobatuwa (Fig. 1), contain stone artefacts associated with the fossilized remains of Stegodon florensis, Komodo dragon, rat and various other taxa. These sites have been dated to 840-700 kyr bp (thousand years before present). The authenticity of the Soa Basin artefacts and their provenance have been demonstrated by previous work, but to quell lingering doubts, here we describe the context, attributes and production modes of 507 artefacts excavated at Mata Menge. We also note specific similarities, and apparent technological continuity, between the Mata Menge stone artefacts and those excavated from Late Pleistocene levels at Liang Bua cave, 50 km to the west. The latter artefacts, dated to between 95-74 and 12 kyr ago, are associated with the remains of a dwarfed descendent of S. florensis, Komodo dragon, rat and a small-bodied hominin species, Homo floresiensis, which had a brain size of about 400 cubic centimetres. The Mata Menge evidence negates claims that stone artefacts associated with H. floresiensis are so complex that they must have been made by modern humans (Homo sapiens).

Abstract: This study reexamines the current understanding of Pleistocene stone-artifact assemblages in island Southeast Asia. A differentiation has long been made between assemblages of large-sized "core tools" and assemblages of small-sized "flake tools." "Core tool" assemblages are often argued to be the handiwork of early hominin species such as Homo erectus, while small-sized "flake tool" assemblages have been attributed to Homo sapiens. We argue that this traditional Southeast Asian perspective on stone tools assumes that the artifacts recovered from a site reflect a complete technological sequence. Our analyses of Pleistocene-age artifact assemblages from Flores, Indonesia, demonstrate that large pebble-based cores and small flake-based cores are aspects of one reduction sequence. We propose that the Flores pattern applies across island Southeast Asia: large-sized "core tool" assemblages are in fact a missing element of the small-sized flake-based reduction sequences found in many Pleistocene caves and rock-shelters. We conclude by discussing the implications of this for associating stone-artifact assemblages with hominin species in island Southeast Asia.
