Abstract: The aim of this study was to determine the effects of a pin‐to‐plate cold atmospheric plasma (CAP) on U‐251 MG three‐dimensional (3D) glioblastoma spheroids under different conditions. 3D tumorspheres showed higher resistance to the CAP treatment compared to 2D monolayer cells. A single CAP treatment was able to induce cytotoxicity, while multiple CAP treatments augmented this effect. CAP was also able to induce cytotoxicity throughout the tumoursphere, and we identified that reactive oxygen species (ROS) plays a major role, while H 2 O 2 plays a partial role in CAP‐induced cytotoxicity in tumourspheres. We conclude that ROS‐dependent cytotoxicity is induced uniformly throughout glioblastoma and epidermoid tumourspheres by direct CAP treatment.

Abstract: Microbial biofilms and bacteria internalised in produce tissue may reduce the effectiveness of decontamination methods. In this study, the inactivation efficacy of in-package atmospheric cold plasma (ACP) afterglow was investigated against Salmonella Typhimurium, Listeria monocytogenes and Escherichia coli in the forms of planktonic cultures, biofilms formed on lettuce and associated bacteria internalised in lettuce tissue. Prepared lettuce broth (3%) was inoculated with bacteria resulting in a final concentration of ~7.0 log10 CFU/ml. For biofilm formation and internalisation, lettuce pieces (5 × 5 cm) were dip-inoculated in bacterial suspension of ~7.0 log10 CFU/ml for 2 h and further incubated for 0, 24 and 48 h at either 4 °C or room temperature (~22 °C) in combination with light/dark photoperiod or at 4 °C under dark conditions. Inoculated s les were sealed inside a rigid polypropylene container and indirectly exposed (i.e. placed outside plasma discharge) to a high voltage (80 kVRMS) air ACP with subsequent storage for 24 h at 4 °C. ACP treatment for 30s reduced planktonic populations of Salmonella, L. monocytogenes and E. coli suspended in lettuce broth to undetectable levels. Depending on storage conditions, bacterial type and age of biofilm, 300 s of treatment resulted in reduction of biofilm populations on lettuce by a maximum of 5 log10 CFU/s le. Scanning electron and confocal laser microscopy pointed to the incidence of bacterial internalisation and biofilm formation, which influenced the inactivation efficacy of ACP. Measured intracellular reactive oxygen species (ROS) revealed that the presence of organic matter in the bacterial suspension might present a protective effect against the action of ROS on bacterial cells. This study demonstrated that high voltage in-package ACP could be a potential technology to overcome bacterial challenges associated with food produce. However, the existence of biofilms and internalised bacteria should be considered for further optimisation of ACP treatment parameters in order to achieve an effective control of the realistic challenges posed by foodborne pathogens.

Abstract: Brown blotch, caused by pathogenic Pseudomonas tolaasii, is the most problematic bacterial disease in Agaricus bisporus mushrooms. Although it does not cause any health problems, it reduces the consumer appeal of mushrooms in the market place, generating important economic losses worldwide. Hyperspectral imaging (HS) is a non-destructive technique that combines imaging and spectroscopy to obtain information from a s le. The objective of this study was to investigate the use of HSI for brown blotch identification and discrimination from mechanical damage on mushrooms. Hyperspectral images of mushrooms subjected to (1) no treatment, (2) mechanical damage or (3) microbiological spoilage were taken during storage and spectra representing each of the classes were selected. Partial least squares-discriminant analysis (PLS-DA) was carried out in two steps: (1) discrimination between undamaged and damaged mushrooms and (2) discrimination between damage sources (i.e. mechanical or microbiological). The models were applied at a pixel level and a decision tree was used to classify mushrooms into one of the aforementioned classes. A correct classification of % was achieved. Results from this study could be used for the development of a sensor to detect and classify mushroom damage of mechanical and microbial origin, which would enable the industry to make rapid and automated decisions to discard produce of poor marketability.

Abstract: In-package nonthermal plasma (NTP) technology is a novel technology for the decontamination of foods and biological materials. This study presents the first report on the potential of the technology for the degradation of pesticide residues on fresh produce. A cocktail of pesticides, namely azoxystrobin, cyprodinil, fludioxonil and pyriproxyfen was tested on strawberries. The concentrations of these pesticides were monitored in priori and post-plasma treatment using GC-MS/MS. An applied voltage and time dependent degradation of the pesticides was observed for treatment voltages of 60, 70 and 80 kV and treatment durations ranging from 1 to 5 min, followed by 24h in-pack storage. The electrical characterisation revealed the operation of the discharge in a stable filamentary regime. The discharge was found to generate reactive oxygen and excited nitrogen species as observed by optical emission spectroscopy.
Synergistic Cytotoxicity Between Cold Atmospheric Plasma and Pyrazolopyrimidinones Against Glioblastoma Cells

Abstract: Pyrazolopyrimidinone is a fused nitrogen-containing heterocyclic system, which acts as a core scaffold in many pharmaceutically relevant compounds. Pyrazolopyrimidinones have been demonstrated to be efficient in treating several diseases, including cystic fibrosis, obesity, viral infection and cancer. We have tested the synergistic anti-cancer effects of 15 pyrazolopyrimidinones, synthesised in a two-step process, combined with cold atmospheric plasma (CAP), a novel innovation generating reactive species with other unique chemical and physical effects. We identify two pyrazolopyrimidinones that act as prodrugs and display enhanced reactive-species dependent cytotoxicity when used in combination with cold atmospheric plasma. Synergistic activation was evident for both direct CAP treatment on prodrug loaded tumour cells and indirect CAP treatment of prodrug in media prior to adding to tumour cells. Our results demonstrate the potential of CAP combined with pyrazolopyrimidinones as a programmable cytotoxic therapy against cancer.

Abstract: Cold plasma is increasingly under research for decontamination of foods, especially fresh fruits and vegetables. The effect of cold plasma on food quality, however, remains under researched. This study investigates the effects of cold plasma generated within a sealed package from a dielectric barrier discharge on the physical quality parameters and respiration rates of cherry tomatoes. Respiration rates and weight loss were monitored continuously, while other parameters are reported at the end of storage period. Differences among weight loss, pH and firmness for control and treated cherry tomatoes were insignificant towards the end of storage life. Changes in respiration rates and colour of tomatoes were recorded as a function of treatment, which were not drastic. The results implicate that cold plasma could be employed as a means for decontamination of cherry tomatoes while retaining product quality.

Anthocyanin and ascorbic acid degradation in sonicated strawberry juice

Abstract: Strawberry juice s les were sonicated at litude levels ranging from 40 to 100% at a constant frequency of 20 kHz for treatment times (2-10 min) and pulse durations of 5 s on and 5 s off. Sonication was found to reduce anthocyanin and ascorbic acid contents by 3.2 and 11%, respectively, at the maximum treatment conditions. Response surface methodology (RSM) based on a two-factor, five-level central composite design was employed to determine the effect of litude level and treatment time on anthocyanins (P3G), ascorbic acid (AA) content, and color values (L*, a*, and b*). The model predictions for the selected nutritional and quality parameters were closely correlated to the experimental results. RSM was demonstrated to be an effective technique to model the effect of sonication on strawberry juice quality while minimizing the number of experiments required.

Abstract: Platinum nanoparticles (PtNPs) have been investigated for their antioxidant abilities in a range of biological and other applications. The ability to reduce off-target CAP cytotoxicity would be useful in Plasma Medicine, however, little has been published to date about the ability of PtNPs to reduce or inhibit the effects of CAP. Here we investigate whether PtNPs can protect against CAP-induced cytotoxicity in cancerous and non-cancerous cell lines. PtNPs were shown to dramatically reduce intracellular reactive species (RONS) production in human U-251 MG cells. However, RONS generation was unaffected by PtNPs in medium without cells. PtNPs protect against CAP induced mitochondrial membrane depolarization, but not cell membrane permeabilization which is a CAP-induced RONS-independent event. PtNPs act as potent intracellular scavengers of reactive species and can protect both cancerous U-251 MG cells and non-cancerous HEK293 cells against CAP induced cytotoxicity. PtNPs may be useful as a catalytic antioxidant for healthy tissue and for protecting against CAP-induced tissue damage. PtNPs are potent catalase and superoxide dismutase mimetics which makes them strong antioxidant candidates for the protection of cells against oxidative stress. CAP was generated using a Dielectric Barrier Device (DBD) system with a voltage output of 75 kV at a frequency of 50 Hz. A range of concentrations of 3nm uncoated PtNPs combined with CAP were examined in human U-251 MG Glioblastoma (GBM) cells and non-cancerous human embryonic kidney HEK293 cells. The protective effects of PtNPs against CAP were explored using several biochemical indicators of oxidative stress and cytotoxicity.

Abstract: In this study, the effect of atmospheric pressure cold plasma and high-pressure processing on the prebiotic orange juice was evaluated. Orange juice containing 7g/100g of commercial fructooligosaccharides (FOS) was directly and indirectly exposed to a plasma discharge at 70kV with processing times of 15, 30, 45 and 60s. For high-pressure processing, the juice containing the same concentration of FOS was treated at 450MPa for 5min at 11.5°C in an industrial equipment (Hyperbaric, model: 300). After the treatments, the fructooligosaccharides were qualified and quantified by thin layer chromatography. The organic acids and color analysis were also evaluated. The maximal overall fructooligosaccharides degradation was found after high-pressure processing. The total color difference was <3.0 for high-pressure and plasma processing. citric and ascorbic acid (Vitamin C) showed increased content after plasma and high-pressure treatment. Thus, atmospheric pressure cold plasma and high-pressure processing can be used as non-thermal alternatives to process prebiotic orange juice.

Abstract: There is a erse range of microbiological challenges facing the food, healthcare and clinical sectors. The increasing and pervasive resistance to broad-spectrum antibiotics and health-related concerns with many biocidal agents drives research for novel and complementary antimicrobial approaches. Biofilms display increased mechanical and antimicrobial stability and are the subject of extensive research. Cold plasmas (CP) have rapidly evolved as a technology for microbial decontamination, wound healing and cancer treatment, owing to the chemical and bio-active radicals generated known collectively as reactive oxygen and nitrogen species. This review outlines the basics of CP technology and discusses the interactions with a range of microbiological targets. Advances in mechanistic insights are presented and applications to food and clinical issues are discussed. The possibility of tailoring CP to control specific microbiological challenges is apparent. This review focuses on microbiological issues in relation to food- and healthcare-associated human infections, the role of CP in their elimination and the current status of plasma mechanisms of action.
Cold Atmospheric Plasma Stimulates Clathrin-Dependent Endocytosis to Repair Oxidised Membrane and Enhance Uptake of Nanomaterial in Glioblastoma Multiforme Cells

Abstract: Cold atmospheric plasma (CAP) enhances uptake and accumulation of nanoparticles and promotes synergistic cytotoxicity against cancer cells. However, the mechanisms are not well understood. In this study, we investigate the enhanced uptake of theranostic nanomaterials by CAP. Numerical modelling of the uptake of gold nanoparticle into U373MG Glioblastoma multiforme (GBM) cells predicts that CAP may introduce a new uptake route. We demonstrate that cell membrane repair pathways play the main role in this stimulated new uptake route, following non-toxic doses of dielectric barrier discharge CAP. CAP treatment induces cellular membrane damage, mainly via lipid peroxidation as a result of reactive oxygen species (ROS) generation. Membranes rich in peroxidised lipids are then trafficked into cells via membrane repairing endocytosis. We confirm that the enhanced uptake of nanomaterials is clathrin-dependent using chemical inhibitors and silencing of gene expression. Therefore, CAP-stimulated membrane repair increases endocytosis and accelerates the uptake of gold nanoparticles into U373MG cells after CAP treatment. We demonstrate the utility of CAP to model membrane oxidative damage in cells and characterise a previously unreported mechanism of membrane repair to trigger nanomaterial uptake. This knowledge will underpin the development of new delivery strategies for theranostic nanoparticles into cancer cells.

Abstract: A plasma source, sustained by the application of a floating high voltage (±15 kV) to parallel-plate electrodes at 50 Hz, has been achieved in a helium/air mixture at atmospheric pressure ( P = 10 5 Pa) contained in a zip-locked plastic package placed in the electrode gap. Some of the physical and antimicrobial properties of this apparatus were established with a view to ascertain its performance as a prototype for the disinfection of fresh produce. The current–voltage ( I – V ) and charge–voltage ( Q – V ) characteristics of the system were measured as a function of gap distance d , in the range (3 × 10 3 ⩽ Pd ⩽ 1.0 × 10 4 Pa m). The electrical measurements showed this plasma source to exhibit the characteristic behaviour of a dielectric barrier discharge in the filamentary mode and its properties could be accurately interpreted by the two-capacitance in series model. The power consumed by the discharge and the reduced field strength were found to decrease quadratically from 12.0 W to 4.5 W and linearly from 140 Td to 50 Td, respectively, in the range studied. Emission spectra of the discharge were recorded on a relative intensity scale and the dominant spectral features could be assigned to strong vibrational bands in the 2+ and 1− systems of N 2 and , respectively, with other weak signatures from the NO and OH radicals and the N + , He and O atomic species. Absolute spectral intensities were also recorded and interpreted by comparison with the non-equilibrium synthetic spectra generated by the computer code SPECAIR. At an inter-electrode gap of 0.04 m, this comparison yielded typical values for the electron, vibrational and translational (gas) temperatures of (4980 ± 100) K, (2700 ± 200) K and (300 ± 100) K, respectively and an electron density of 1.0 × 10 17 m −3 . A Boltzmann plot also provided a value of (3200 ± 200 K) for the vibrational temperature. The antimicrobial efficacy was assessed by studying the resistance of both Escherichia coli K12 its isogenic mutants in soxR , soxS , oxyR , rpoS and dnaK selected to identify possible cellular responses and targets related with 5 min exposure to the active gas in proximity of, but not directly in, the path of the discharge filaments. Both the parent strain and mutants populations were significantly reduced by more than 1.5 log cycles in these conditions, showing the potential of the system. Post-treatment storage studies showed that some transcription regulators and specific genes related to oxidative stress play an important role in the E. coli repair mechanism and that plasma exposure affects specific cell regulator systems.

Optimization of atmospheric air plasma for degradation of organic dyes in wastewater

Abstract: This study optimises the degradation of a cocktail of the dyes methyl orange and bromothymol blue by atmospheric air plasma. Response surface methodology (RSM) was employed to investigate the efficacy of the plasma process parameters on degradation efficiency. A Box–Behnken design (BBD) was employed to optimise the degradation of dyes by air plasma discharge. A second order polynomial equation was proposed to predict process efficiency. It was observed that the predicted values are significant (p& .001) with coefficients of determination 0.98, 0.96, 0.98 for dye degradation, pH value and ozone concentration, respectively. The analysis of variance results showed that the coefficients of the polynomials for the percentage degradation and ozone concentration responses indicated positive linear effects (p& .001), whereas a negative linear effect was found for pH. The positive linear effect of variable emphasises that voltage and treatment time were the most dominant factors (p& .001), meaning that higher degradation efficiencies are achieved with an increase in treatment duration. This study showed that a BBD model and RSM could be employed to optimize the colour degradation parameters of non-thermal plasma treated model dyes while minimising the number of experiments required.

Abstract: Glioblastoma multiforme (GBM) is the most common and biologically aggressive brain tumour. The current standard therapy for GBM consists in surgical resection, followed by radiotherapy and chemotherapy. Yet, the treatment is limited due to the area for the surgical resection and for the inability of some drugs to cross the brain blood barrier, leading to a general prognostic of no more than a year. Cold atmospheric plasma (CAP) is a new approach in the treatment of this challenging disease. CAP interaction with cells is dependent on physical and chemical factors, with different plasma discharges, cell type, and culture conditions leading to different CAP activity. Considering the plasma self-adaptation that different plasma discharge modes can undergo, which leads to different interaction plasma/cells, the characterization of a new device is essential. In this study we analysed the effect of a novel large pin-to-plate non-thermal atmospheric plasma on U-251 MG cells under different conditions. The analysis of reactive oxygen and nitrogen species (RONS) on plasma, media and cells were also assessed. We were able to demonstrate that the pin-to-plate device is cytotoxic to GBM cells in a dose, time and ROS dependent manner. Abstract: Dielectric barrier discharge atmospheric plasma is a novel nonthermal technology for the food and packaging industry. The effects of dielectric barrier discharge plasma on the surface, structural, thermal, and moisture sorption properties of edible zein films have been examined. Plasma treatment increased the surface roughness and equillibrium moisture content of the zein film in a direct relationship with the applied voltage level. No significant difference in the thermal stability of the zein film is also observed after plasma treatment. Dielectric barrier discharge plasma treatments of zein film lead to a change in the protein conformation which is confirmed by X‐ray diffraction and Fourier transform Infrared spectroscopy. The evaluation of films modifications by plasma discharge will contribute to enhance the in‐package decontamination studies of food products by plasma. © 2014 Wiley Periodicals, Inc. J. Appl. Polym. Abstract: This research investigated the efficacy of gaseous ozone on the inactivation of Escherichia coli ATCC 25922 and NCTC 12900 strains in apple juice of a range of pH levels, using an ozone bubble column. The pH levels investigated were 3.0, 3.5, 4.0, 4.5 and 5.0. Apple juice inoculated with E. coli strains (10(6)CFU/mL) was treated with ozone gas at a flow rate of 0.12L/min and ozone concentration of 0.048 mg/min/mL for up to 18 min. Results show that inactivation kinetics of E. coli by ozone were affected by pH of the juice. The ozone treatment duration required for achieving a 5-log reduction was faster (4 min) at the lowest pH than at the highest pH (18 min) studied. The relationship between time required to achieve 5log reduction (t(5d)) and pH for both strains was described mathematically by two exponential equations. Ozone treatment appears to be an effective process for reducing bacteria in apple juice and the required applied treatment for producing a safe apple juice is dependant on its acidity level.
Assessing stress responses to atmospheric cold plasma exposure using Escherichia coli knock-out mutants

Abstract: This study investigated the effect of atmospheric cold plasma (ACP) exposure-induced stress on microbial inactivation patterns and the regulation of genes involved in the microbial stress response in conjunction with key processing parameters of exposure time and post-treatment storage time. Cell suspensions of Escherichia coli BW 25113 and its isogenic knock-out mutants in rpoS, soxR, soxS, oxyR and dnaK genes were treated with high-voltage ACP in a sealed package for 1, 3 and 5 min followed by 0-, 1- and 24-h post-treatment storage. Reactive oxygen species (ROS) densities and colony formation were determined. ΔrpoS strain showed higher microbial reduction and greater cell permeability than other mutants, while ΔoxyR only showed this effect after 5 min of treatment. With increased post-treatment storage time, ΔsoxS and ΔsoxR had increased sensitivity and resistance respectively. ΔdnaK cell suspensions had much higher ROS than other strains and showed increased sensitivity with 24 h post-treatment storage. RpoS and oxyR genes have both short-term and long-term regulatory effects under plasma stress. However, knocking out dnaK gene had an immediate response on ROS scavenging and long-term repairing mechanisms. ΔsoxR and ΔsoxS had different responses to ACP treatment with the increase in post-treatment time in relation to clearance of reactive species implying the different characteristics and functions as subunits. By comparing the response of mutants under ACP exposure to key processing parameters, the mechanism of microbial inactivation was partly revealed with respect to cellular regulation and repairing genes.

Abstract: The exposure of aqueous solutions to atmospheric plasmas results in the generation of relatively long-lived secondary products such as hydrogen peroxide which are biologically active and have demonstrated anti-microbial and cytotoxic activity. The use of plasma-activated solutions in applications such as microbial decontamination or anti-cancer treatments requires not only adequate performance on target cells but also a safe operating window regarding the impact on surrounding tissues. Furthermore the generation of plasma-activated fluids needs to be considered as a by-stander effect of subjecting tissue to plasma discharges. Cytotoxicity and mutagenicity assays using mammalian cell lines were used to elucidate the effects of solutions treated with di-electric barrier discharge atmospheric cold plasma. Plasma-treated PBS inhibited cell growth in a treatment time-dependent manner showing a linear correlation to the solutions’ peroxide concentration which remained stable over several weeks. Plasma-treated foetal bovine serum (FBS) acting as a model for complex bio-fluids showed not only cytotoxic effects but also exhibited increased mutagenic potential as determined using the mammalian HPRT assay. Further studies are warranted to determine the nature, causes and effects of the cyto- and genotoxic potential of solutions exposed to plasma discharges to ensure long-term safety of novel plasma applications in medicine and healthcare.

Abstract: Mapping of vegetation species and communities in sensitive ecosystems is essential for identification and management of anthropogenic impacts. Unmanned aerial vehicle (UAV)-hyperspectral systems are among the latest technologies in remote sensing that hold a potential for obtaining unprecedented quality of remote sensing data for vegetation mapping and health status monitoring applications. In this study, high-resolution (1–1.5 cm) spectral imaging data (15 bands) from a tunable spectrometer is used to map five species of vegetation in a complex upland sw environment. The overall accuracy of classification was found to be 88.9% with a kappa coefficient of 0.83. Three classes (bare earth, sedgeland grass and black sheoak) have achieved higher accuracy (above 78%) and one class (bracken fern) has lower accuracy (58%). UAV-hyperspectral technology is, therefore, an effective tool to identify and map sensitive sw vegetation. The technology can be potentially applied to determine the health status of the species.

Abstract: In this review, antimicrobials from a range of plant, animal, and microbial sources are reviewed along with their potential applications in food systems. Chemical and biochemical antimicrobial compounds derived from these natural sources and their activity against a range of pathogenic and spoilage microorganisms pertinent to food, together with their effects on food organoleptic properties, are outlined. Factors influencing the antimicrobial activity of such agents are discussed including extraction methods, molecular weight, and agent origin. These issues are considered in conjunction with the latest developments in the quantification of the minimum inhibitory (and noninhibitory) concentration of antimicrobials and/or their components. Natural antimicrobials can be used alone or in combination with other novel preservation technologies to facilitate the replacement of traditional approaches. Research priorities and future trends focusing on the impact of product formulation, intrinsic product parameters, and extrinsic storage parameters on the design of efficient food preservation systems are also presented.

Abstract: Silver nanoparticles (AgNP) emerged as a promising reagent for cancer therapy with oxidative stress implicated in the toxicity. Meanwhile, studies reported cold atmospheric plasma (CAP) generation of reactive oxygen and nitrogen species has selectivity towards cancer cells. Gold nanoparticles display synergistic cytotoxicity when combined with CAP against cancer cells but there is a paucity of information using AgNP, prompting to investigate the combined effects of CAP using dielectric barrier discharge system (voltage of 75 kV, current is 62.5mA, duty cycle of 7.5kVA and input frequency of 50-60Hz) and 10nm PVA-coated AgNP using U373MG Glioblastoma Multiforme cells. Cytotoxicity in U373MG cells was -fold greater when treated with both CAP and PVA-AgNP compared with either therapy alone (IC 50 of 4.30 μg/mL with PVA-AgNP alone compared with 0.07 μg/mL after 25s CAP and 0.01 μg/mL 40s CAP). Combined cytotoxicity was ROS-dependent and was prevented using N-Acetyl Cysteine. A novel darkfield spectral imaging method investigated and quantified AgNP uptake in cells determining significantly enhanced uptake, aggregation and subcellular accumulation following CAP treatment, which was confirmed and quantified using atomic absorption spectroscopy. The results indicate that CAP decreases nanoparticle size, decreases surface charge distribution of AgNP and induces uptake, aggregation and enhanced cytotoxicity in vitro .

Abstract: Generation of wastewater is one of the main environmental sustainability issues across food sector industries. The constituents of food process effluents are often complex and require high energy and processing for regulatory compliance. Wastewater streams are the subject of microbiological and chemical criteria, and can have a significant eco-toxicological impact on the aquatic life. Thus, innovative treatment approaches are required to mitigate environmental impact in an energy efficient manner. Here, dielectric barrier discharge atmospheric cold plasma (ACP) was evaluated for control of key microbial indicators encountered in food industry effluent. This study also investigated the eco-toxicological impact of cold plasma treatment of the effluents using a range of aquatic bioassays. Continuous ACP treatment was applied to synthetic dairy and meat effluents. Abstract: Solution pH is a significant parameter that affects the electrical characteristics of gas–liquid discharges and thus potentially produces different plasma chemistries for different plasma-engineered applications. In this study, the discharge characteristics and long-lived aqueous reactive species under different initial pH conditions were investigated. It was found that the discharge contained three phases in one pulse cycle, which occurred at the pulse rising edge, the falling edge, and between the rising and falling edges. The discharge intensity and average power at an initial solution pH of 7.0 are much lower than those obtained at an initial solution pH of 5.0 and 9.0. In contrast, the density ratio of N2 (C3Πu, v = 1)/N2 (C3Πu, v = 0) is much higher under neutral solution conditions, indicating that the relatively high vibrational energy was obtained in the gas–liquid discharge plasma. Concentrations of aqueous species H2O2, NO2–, and NO3– are higher at the initial pH of 9.0, indicating that different plasma intensities and chemical compositions (H+ and OH–) are involved in their generation and consumption processes. This study provides insights into understanding and controlling the characteristics of the gas–liquid discharge under different solution conditions.

Abstract: Non-thermal plasma has received much attention for elimination of microbial contamination from a range of surfaces. This study aimed to determine the effect of a range of dielectric barrier discharge high voltage atmospheric cold plasma (HVACP) parameters for inactivation of Bacillus atrophaeus spores inside a sealed package. A sterile polystyrene Petri dish containing B. atrophaeus spore strip (spore population 2.3 × 10(6)/strip i.e. 6.36 log10/strip) was placed in a sealed polypropylene container and was subjected to HVACP treatment. The HVACP discharge was generated between two aluminium plate electrodes using a high voltage of 70kVRMS. The effects of process parameters, including treatment time, mode of exposure (direct/indirect), and working gas types, were evaluated. The influence of relative humidity on HVACP inactivation efficacy was also assessed. The inactivation efficacy was evaluated using colony counts. Optical absorption spectroscopy (OAS) was used to assess gas composition following HVACP exposure. A strong effect of process parameters on inactivation was observed. Direct plasma exposure for 60s resulted in ≥6 log10 cycle reduction of spores in all gas types tested. However, indirect exposure for 60s resulted in either 2.1 or 6.3 log10 cycle reduction of spores depending on gas types used for HVACP generation. The relative humidity (RH) was a critical factor in bacterial spore inactivation by HVACP, where a major role of plasma-generated species other than ozone was noted. Direct and indirect HVACP exposure for 60s at 70% RH recorded 6.3 and 5.7 log10 cycle reduction of spores, respectively. In summary, a strong influence of process parameters on spore inactivation was noted. Rapid in-package HVACP inactivation of bacterial spores within 30-60s demonstrates the promising potential application for reduction of spores on medical devices and heat-sensitive materials.

Abstract: Hyperspectral imaging (HSI) combines conventional imaging and spectroscopy to simultaneously acquire both spatial and spectral information from an object. This technology has recently emerged as a powerful process analytical tool for rapid, non‐contact and non‐destructive food analysis. In this study, the potential application of HSI for damage detection on the caps of white mushrooms ( Agaricus bisporus ) was investigated. Mushrooms were damaged by controlled vibration to simulate damage caused by transportation. Hyperspectral images were obtained using a pushbroom line‐scanning HSI instrument, operating in the wavelength range of 400–1000 nm with spectroscopic resolution of 5 nm. The effective resolution of the CCD detector was 580 × 580 pixels by 12 bits. Two data reduction methods were investigated: in the first, principal component analysis (PCA) was applied to the hypercube of each s le, and the second PC (PC 2) scores image was used for identification of bruise‐damaged regions on the mushroom surface in the second method PCA was applied to a dataset comprising of average spectra from regions normal and bruise‐damaged tissue. In this case it was observed that normal and bruised tissue were separable along the resultant first principal component (PC 1) axis. Multiplying the PC 1 eigenvector by the hypercube data allowed reduction of the hypercube to a 2‐D image, which showed maximal contrast between normal and bruise‐damaged tissue. The second method performed better than the first when applied to a set of independent mushroom s les. The results from this study could be used for the development of a non‐destructive monitoring system for rapid detection of damaged mushrooms on the processing line. Abstract: Plasma bubbles are regarded as a promising means of interacting plasma discharges and liquids due to their high efficiency in the generation of reactive species. The discharge mode and characteristics are significant factors that should be considered. In this study, the plasmas are initially generated in the gas phase and then driven by the gas flow to diffuse into the solution through the two holes at the lower part of the quartz tube to form plasma bubbles. The discharge modes, characteristics, and plasma–liquid interactions in two different configurations, i.e., bare electrode and dielectric-coated electrode, are investigated. It is found that the discharge mode induced for the two structures is different, with a hybrid-mode operating in the bare electrode design and a filamentary mode operating in the dielectric design. When the applied voltage is increased, a filamentary-to-spark transition occurs in the bare structure, while the discharge remains relatively stable in the dielectric design. Direct and intense contact between the discharge and the solution in the bare structure greatly promotes the physio-chemical reactions and results in obvious changes in H2O2 concentration, solution pH, conductivity, and temperature. This study provides insights into hybrid gas–liquid discharges and reactor design for plasma bubble generation.

Abstract: This study investigated a range of atmospheric cold plasma (ACP) process parameters for bacterial inactivation with further investigation of selected parameters on cell membrane integrity and DNA damage. The effects of high voltage levels, mode of exposure, gas mixture and treatment time against Escherichia coli and Listeria monocytogenes were examined. 10(8) CFU ml(-1) E. coli ATCC 25922, E. coli NCTC 12900 and L. monocytogenes NCTC11994 were ACP-treated in 10 ml phosphate-buffered saline (PBS). Working gas mixtures used were air (gas mix 1), 90% N2 + 10% O2 (gas mix 2) and 65% O2 + 30% CO2 + 5% N2 (gas mix 3). Greater reduction of viability was observed for all strains using higher voltage of 70 kVRMS and with working gas mixtures with higher oxygen content in combination with direct exposure. Indirect ACP exposure for 30 s inactivated below detection level both E. coli strains. L. monocytogenes inactivation within 30 s was irrespective of the mode of exposure. Leakage was assessed using A260 absorbance, and DNA damage was monitored using PCR and gel electrophoresis. Membrane integrity was compromised after 5 s, with noticeable DNA damage also dependent on the target cell after 30 s. Plasma treatment was effective for inactivation of challenge micro-organisms, with a greater sensitivity of L. monocytogenes noted. Different damage patterns were observed for the different bacterial strains attributed to the membrane structure and potential resistance mechanisms. Using atmospheric air as working gas resulted in useful inactivation by comparison with high nitrogen or high oxygen mix. The mechanism of inactivation was a function of treatment duration and cell membrane characteristics, thus offering potential for optimized process parameters specific to the microbial challenge.

Abstract: Contamination of cereal grains as a key global food resource with insects or microorganisms is a persistent concern for the grain industry due to irreversible damage to quality and safety characteristics and economic losses. Atmospheric cold plasma presents an alternative to conventional grain decontamination methods owing to the high antimicrobial potential of reactive species generated during the treatment, but effects against product specific microflora are required to understand how to optimally develop this approach for grains. This work investigated the influence of ACP processing parameters for both cereal grain decontamination and grain quality as important criteria for grain or seed use. A high voltage (HV) (80 kV) dielectric barrier discharge (DBD) closed system was used to assess the potential for control of native microflora and pathogenic bacterial and fungal challenge microorganisms, in tandem with effects on grain functional properties. Response surface modelling of experimental data probed the key factors in relation to microbial control and seed germination promotion. Abstract: The use of plastics has spanned across almost all aspects of day to day life. Although their uses are invaluable, they contribute to the generation of a lot of waste products that end up in the environment and end up polluting natural habitats such as forests and the ocean. By treating low-density polyethylene (LDPE) s les with non-thermal plasma in ambient air and with an addition of ≈4% CO2, the biodegradation of the s les can be increased due to an increase in oxidative species causing better cell adhesion and acceptance on the polymer s le surface. It was, however, found that the use of this slight addition of CO2 aided in the biodegradation of the LDPE s les more than with solely ambient air as the carbon bonds measured from Raman spectroscopy were seen to decrease even more with this change in gas composition and chemistry. The results show that the largest increase of polymer degradation occurs when a voltage of 32 kV is applied over 300 s and with a mixture of ambient air and CO2 in the ratio 25:1.

Abstract: Atmospheric cold plasma (ACP) represents a potential alternative to traditional methods for non-thermal decontamination of foods. In this study, the antimicrobial efficacy of a novel dielectric barrier discharge ACP device against Escherichia coli, Salmonella enterica Typhimurium and Listeria monocytogenes inoculated on cherry tomatoes and strawberries, was examined. Bacteria were spot inoculated on the produce surface, air dried and sealed inside a rigid polypropylene container. S les were indirectly exposed (i.e. placed outside plasma discharge) to a high voltage (70 kVRMS) air ACP and subsequently stored at room temperature for 24 h. ACP treatment for 10, 60 and 120 s resulted in reduction of Salmonella, E. coli and L. monocytogenes populations on tomato to undetectable levels from initial populations of 3.1, 6.3, and 6.7 log10 CFU/s le, respectively. However, an extended ACP treatment time was necessary to reduce bacterial populations attached on the more complex surface of strawberries. Treatment time for 300 s resulted in reduction of E. coli, Salmonella and L. monocytogenes populations by 3.5, 3.8 and 4.2 log10 CFU/s le, respectively, and also effectively reduced the background microflora of tomatoes.

Abstract: Freshly squeezed orange juice s les were ozonated with control variables of gas flow rate (0-0.25 L min (-1)), ozone concentration (0.6-10.0%w/w), and treatment time (0-10 min). Effects of ozone processing variables on orange juice quality parameters of pH, degrees Brix, titratable acidity (TA), cloud value, nonenzymatic browning (NEB), color values ( L*, a*, and b*), and ascorbic acid content were determined. No significant changes in pH, degrees Brix, TA, cloud value, and NEB ( p < 0.05) were found. L*, a*, and b* color values were significantly affected by gas flow rate, ozone concentration, and treatment time. The changes in lightness ( L*) values and total color difference (TCD) values were fitted well to zero-order kinetics, whereas a*, b*, and ascorbic acid degradation followed first-order kinetics. The rate constants for a*, b*, and TCD were linearly correlated with ozone concentration ( R (2) = 0.88-0.99), whereas the rate constants for L* and ascorbic acid were exponentially correlated ( R (2) = 0.94-0.98).

Abstract: This study evaluated the effect of atmospheric cold plasma and ozone treatments on the key compounds (sugars, amino acids and short chain organic acids) in orange juice by NMR and chemometric analysis. The juice was directly and indirectly exposed to atmospheric cold plasma field at 70kV for different treatment time (15, 30, 45 and 60sec). For ozone processing different loads were evaluated. The Principal Component Analysis shown that the groups of compounds are affected differently depending on the processing. The ozone was the processing that more affected the aromatic compounds and atmospheric cold plasma processing affected more the aliphatic compounds. However, these variations did not result in significant changes in orange juice composition as a whole. Thus, NMR data and chemometrics were suitable to follow quality changes in orange juice processing by atmospheric cold plasma and ozone.

Abstract: The past decade has seen a surge in the scientific literature investigating the potential food-related applications of plasma. A multidisciplinary scientific effort has started to demonstrate process efficacy for a range of plasma applications, including antimicrobial, pesticidal, food functionalization, and waste treatment. Insights into the interactions of plasma species with food and the mechanisms of action are also emerging. This review examines the current status of cold plasma technology within the food sector with a particular emphasis on emerging applications. Opportunities and current challenges that need to be addressed for successful adoption of the approach by industry are detailed.
Multipoint NIR spectroscopy for gross composition analysis of powdered infant formula under various motion conditions

Abstract: Powdered infant formula (PIF) is a worldwide, industrially produced, human milk substitute. Manufacture of PIF faces strict quality controls in order to ensure that the product meets all compositional requirements. Near-infrared (NIR) spectroscopy is a rapid, non-destructive and well-qualified technique for food quality assessments. The use of fibre-optic NIR sensors allows measuring in-line and at real-time, and can record spectra from different stages of the process. The non-contact character of fibre-optic sensors can be enhanced by fitting collimators, which allow operation at various distances. The system, based on a Fabry-Perot interferometer, records four spectra concurrently, rather than consecutively as in the "quasi-simultaneous" multipoint NIR systems. In the present study, a novel multipoint NIR spectroscopy system equipped with four fibre-optic probes with collimators was assessed to determine carbohydrate and protein contents of PIF s les under static and motion conditions (0.02, 0.15 and 0.30m/s) to simulate possible industrial scenarios. Best results were obtained under static conditions providing a R(2) of calibration of 0.95 and RMSEP values of 1.89%. Yet, considerably low values of RMSEP, for instance 2.70% at 0.15m/s, were provided with the in-motion predictions, demonstrating the system's potential for in/on-line applications at various levels of speed. The current work also evaluated the viability of using general off-line calibrations developed under static conditions for on/in-line applications subject to motion. To this end, calibrations in both modes were developed and compared. Best results were obtained with specific calibrations however, reasonably accurate models were obtained with the general calibration. Furthermore, this work illustrated independency of the collimator-probe setup by characterizing PIF s les simultaneously recorded according to their carbohydrate content, even when measured under different conditions. Therefore, the improved multipoint NIR approach constitutes a potential in/on-line tool for quality evaluation of PIF over the manufacturing process.

Abstract: To investigate the effect of extrinsic control parameters for ozone inactivation of E. coli in a bubble column. Ozone inactivation of Escherichia coli ATCC 25922 in Tryptic Soya Broth was examined. The parameters studied included temperature (ambient, 20, 25 and 30 degrees C), exposure time (up to 30 min), gas flow rate (0.03, 0.06, 0.12, 0.25, 0.5 and 0.75 l min(-1)) and concentration level (five different levels). The efficacy of ozone treatment was a function of the parameters investigated and optimum control parameters of flow rate (0.12 l min(-1)), temperature (ambient) and ozone concentration (75 microg ml(-1)) resulted in a t(d5) (time required to achieve 5 log reduction) of 20 min. Optimum control parameters of gas flow rate, ozone concentration and temperature are reported for E. coli inactivation within a bubble column. In 2001, the FDA approved use of ozone as a direct additive to food and in 2004, issued guidelines for the use of ozone in liquid systems. However, these guidelines highlighted gaps in the literature for ozonation of liquid foods. This study provides useful information regarding optimum extrinsic control parameters for E. coli inactivation in liquid media using a bubble column to ensure microbiological safety.

Abstract: Many of the documented outbreaks of Cronobacter sakazakii have been linked to infant formula. The aims of this work are to monitor the inactivation kinetics of C.sakazakii NCTC 08155 and ATCC 11467 and to determine quantitatively the effectiveness of ultrasonic treatments as an alternative to heat processing of reconstituted infant milk formula before feeding of infants at highest risk. Inactivation studies of C. sakazakii inoculated in reconstituted infant formula were performed at the combined conditions of temperature, i.e., 25 degrees C, 35 degrees C, 50 degrees C and litude, i.e., 24.4, 30.5, 42.7, 54.9, 61 microm and the kinetics were described by a range of inactivation models. The dependency of the specific inactivation rate with respect to the product of temperature and litude was described by a modified Bigelow type model. Ultrasound combined with temperature was efficient to reduce significantly the microbial levels of C. sakazakii. C. sakazakii strain NCTC 08155 was at the same range of temperature and litude resistance as strain ATCC 11467. Application of ultrasound is an alternative process for the production of safe reconstituted infant formula. This study contributes on the quantitative assessment of the resistance of C. sakazakii.

Abstract: Numerous electrode geometries and power supplies, both commercial and in-house, have been employed for the generation of low-temperature atmospheric plasma jets. In this work, the development and operation of a 12 jet nonthermal atmospheric plasma system is presented. The study is based on optical spectroscopy as a diagnostic method due to its nonintrusive nature. A key focus of this study was the material selection (conductive and nonconductive), with several polymers screened for the jet design leading to polyacetal as the choice material. Their results are compared with other atmospheric plasma jet systems. The results show a significant increase in residence time and the spatial homogeneity for ambient air's main species, including: OH, O I, O2, O3, N2, and N2+. Their densities are studied with respect to treatment time, distance, duty cycle, and discharge frequency, as well as the jets' carrier gas chemistries (argon and helium). For their plasma jet system, the bulk of the chemical reactions occur in the surrounding atmosphere and not in the jet nozzle, which is different from most other plasma jet systems. The electron energy distribution function, for the given chemistries, is also reported.

Abstract: The aim of this study was to investigate whether sonoporation of cancer cells using ultrasound (US) technology could enhance the anticancer effects of cold atmospheric plasma. US‐induced transient sonoporation of cancer cells with little to no cytotoxicity observed on the cell lines tested. Synergistic effects of US were observed when combined with both direct and indirect cold atmospheric plasma. These cytotoxic effects were dependent on reactive species production. To the best of our knowledge that is the first time that the effects of those two nonthermal technologies were evaluated in cancer cells, demonstrating a promising combined strategy for cancer therapies, particularly for those with penetration limitations, such as glioblastoma.

Abstract: Chemical Imaging (CI) is an emerging platform technology that integrates conventional imaging and spectroscopy to attain both spatial and spectral information from an object. Vibrational spectroscopic methods, such as Near Infrared (NIR) and Raman spectroscopy, combined with imaging are particularly useful for analysis of biological harmaceutical forms. The rapid, non-destructive and non-invasive features of CI mark its potential suitability as a process analytical tool for the pharmaceutical industry, for both process monitoring and quality control in the many stages of drug production. This paper provides an overview of CI principles, instrumentation and analysis. Recent applications of Raman and NIR-CI to pharmaceutical quality and process control are presented challenges facing CI implementation and likely future developments in the technology are also discussed.

Abstract: Recent advancements in hyperspectral remote sensing technology now provide improved diagnostic capabilities to assess vegetation health conditions. This paper uses a set of 13 vegetation health indices related to chlorophyll, xanthophyll, blue/green/red ratio and structure from airborne hyperspectral reflectance data collected around a derelict mining area in Yerranderie, New South Wales, Australia. The studied area has ten historic mine shafts with a legacy of heavy metals and acidic contamination in a pristine ecosystem now recognised as Great Blue Mountain World Heritage Area. The forest is predominantly comprised of different species of Eucalyptus trees. In addition to the airborne survey, ground-based spectra of the tree leaves were collected along the two accessible heavy metal contaminated pathways. The stream networks in the area were classified and the geospatial patterns of vegetation health were analysed along the Tonalli River, a major water tributary flowing through the National Park. Despite the inflow of contaminated water from the near-mine streams, the measured vegetation health indices along Tonalli River were found to remain unchanged. The responses of the vegetation health indices between the near-mine and away-mine streams were found similar. Based on the along-stream and inter-stream analysis of the spectral indices of vegetation health, no significant impact of the heavy metal pollution could be noticed. The results indicate the possibility of the vegetation having developed immunity towards the high levels of heavy metal pollution over a century of exposure.

Abstract: Physical stress (i.e., bruising) during harvesting, handling, and transportation triggers enzymatic discoloration of mushrooms, a common and detrimental phenomenon largely mediated by polyphenol oxidase (PPO) enzymes. Hyperspectral imaging (HSI) is a nondestructive technique that combines imaging and spectroscopy to obtain information from a s le. The objective of this study was to assess the ability of HSI to predict the activity of PPO on mushroom caps. Hyperspectral images of mushrooms subjected to various damage treatments were taken, followed by enzyme extraction and PPO activity measurement. Principal component regression (PCR) models (each with three PCs) built on raw reflectance and multiple scatter-corrected (MSC) reflectance data were found to be the best modeling approach. Prediction maps showed that the MSC model allowed for compensation of spectral differences due to s le curvature and surface irregularities. Results reveal the possibility of developing a sensor that could rapidly identify mushrooms with a higher likelihood to develop enzymatic browning, hence aiding produce management decision makers in the industry.

Abstract: Cold plasma science and technology is increasingly investigated for translation to a plethora of issues in the agriculture and food sectors. The ersity of the mechanisms of action of cold plasma, and the flexibility as a standalone technology or one that can integrate with other technologies, provide a rich resource for driving innovative solutions. The emerging understanding of the longer-term role of cold plasma reactive species and follow-on effects across a range of systems will suggest how cold plasma may be optimally applied to biological systems in the agricultural and food sectors. Here we present the current status, emerging issues, regulatory context, and opportunities of cold plasma with respect to the broad stages of primary and secondary food production.
Effect of ultrasound processing on anthocyanins and color of red grape juice

Abstract: Grape juice s les were sonicated with processing variables of litude level (24.4-61.0microm) and treatment time (0-10min) at a constant frequency of 20kHz and pulse durations of 5s on and 5s off. A full factorial experimental design with regression modeling was employed to investigate the main effects of litude level and treatment time on anthocyanins and color parameters. Significant effects of sonication on major anthocyanins cyanidin-3-O-glucosides (CA), malvanidin-3-O-glucosides (MA) and delphinidin-3-O-glucosides (DA), color values (L*, a*, b*) and color index (CI) were observed. Prediction models were found to be significant (p<0.05) with low standard errors and high coefficients of determination (R(2)). Model predictions for critical quality parameters of anthocyanins (CA MA DA), color values (L*, a*, b*), TCD and CI inactivation were closely correlated to the experimental results obtained. Significant retention of anthocyanin content in grape juice was observed for CA (97.5 %) MA (48.2 %) and DA (80.9%) during sonication. CI and other color combinations (L*a*b*, L*a*/b* and L*b*/a*) were found to be strongly correlated with anthocyanin content. This study shows that sonication could be employed for as a preservation technique for fruit juice processing where anthocyanin retention is desired.

Abstract: A range of nonthermal techniques have demonstrated process efficacy in ensuring product safety, extension of shelf life, and in general a retention of key quality attributes. However, various physical, chemical and biochemical effects of nonthermal techniques on macro and micro nutrients are evident, leading to both desirable and undesirable changes in food products. The objective of this review is to outline the effects of nonthermal techniques on food chemistry and the associated degradation mechanisms with the treatment of foods. Oxidation is one of the key mechanisms responsible for undesirable effects induced by nonthermal techniques. Degradation of key macromolecules largely depends on the processing conditions employed. Various extrinsic and intrinsic control parameters of high-pressure processing, pulsed electric field, ultrasound processing, and cold atmospheric plasma on chemistry of processed food are outlined. Proposed mechanisms and associated degradation of macromolecules, i.e., proteins, lipids, and bioactive molecules resulting in food quality changes are also discussed.

This study ascertained the potential of dielectric barrier discharge atmospheric cold plasma (DBD-ACP) for control of B. thermosphacta, taking microbial and food environment factors into consideration, and investigated the shelf-life of lamb chop after in-package plasma treatment in modified atmosphere. Community profiling was used to assess the treatment effects on the lamb microflora. ACP treatment (80 kV) for 30s inactivated B. thermosphacta populations below detection levels in PBS, while 5 min treatment achieved a 2 Log cycle reduction using a complex meat model medium and attached cells. The antimicrobial efficacy of plasma was reduced but still apparent on lamb chop surface-inoculated with high concentrations of B. thermosphacta. Lamb chop treated under modified atmosphere exhibited reduced microbial growth over the product shelf-life and community profiling showed no evident changes to the microbial populations after the treatment. The overall results indicated potential of ACP to enhance microbial control leading to meat storage life extension through adjusting the modality of treatment.

Abstract: Power ultrasound is recognised as a potential non thermal technique to inactivate microorganisms pertinent to fruit juices. In this study, the effect of sonication on the resistance of yeast (Pichia fermentans) in tomato juice was investigated. Tomato juice s les were sonicated at litude levels ranging from 24.4 to 61.0mum at a constant frequency of 20kHz for different treatment times (2 to 10min) and pulse durations of 5s on and 5s off. Significant reductions (p 0.98) and low RMSE (<0.51). The desired 5 log reductions (D(5) value) and shape factors were found to correlate exponentially with litude level. Results presented in this study show that sonication alone is an effective process to achieve the desired level of yeast inactivation in tomato juice.

Abstract: Atmospheric cold plasma (ACP) is a promising nonthermal technology effective against a wide range of pathogenic microorganisms. Reactive oxygen species (ROS) play a crucial inactivation role when air or other oxygen-containing gases are used. With strong oxidative stress, cells can be damaged by lipid peroxidation, enzyme inactivation, and DNA cleavage. Identification of ROS and an understanding of their role are important for advancing ACP applications for a range of complex microbiological issues. In this study, the inactivation efficacy of in-package high-voltage (80 kV [root mean square]) ACP (HVACP) and the role of intracellular ROS were investigated. Two mechanisms of inactivation were observed in which reactive species were found to either react primarily with the cell envelope or damage intracellular components. Escherichia coli was inactivated mainly by cell leakage and low-level DNA damage. Conversely, Staphylococcus aureus was mainly inactivated by intracellular damage, with significantly higher levels of intracellular ROS observed and little envelope damage. However, for both bacteria studied, increasing treatment time had a positive effect on the intracellular ROS levels generated.

Abstract: Cold atmospheric plasma (CAP) has demonstrated synergistic cytotoxic effects with nanoparticles, especially promoting the uptake and accumulation of nanoparticles inside cells. However, the mechanisms driving the effects need to be explored. In this study, we investigate the enhanced uptake of theranostic nanomaterials by CAP. Numerical modelling of the uptake of gold nanoparticle into U373MG Glioblastoma multiforme (GBM) cells predicts that CAP may introduce a new uptake route. We demonstrate that cell membrane repair pathways play the main role in this stimulated new uptake route, following non-toxic doses of dielectric barrier discharge CAP (30 s, 75 kV). CAP treatment induces cellular membrane damage, mainly via lipid peroxidation as a result of reactive oxygen species (ROS) generation. Membranes rich in peroxidated lipids are then trafficked into cells via membrane repairing endocytosis. We confirm that the enhanced uptake of nanomaterials is clathrin-dependent using chemical inhibitors and silencing of gene expression. Therefore, CAP-stimulated membrane repair increases endocytosis and accelerates the uptake of gold nanoparticles into U373MG cells after CAP treatment. Our data demonstrate the utility of CAP to model membrane oxidative damage in cells and characterise a previously unreported mechanism of membrane repair to trigger nanomaterial uptake which will be useful for developing more efficient deliveries of nanoparticles and pharmaceuticals into cancer cells for tumour therapy and diagnosis. This mechanism of RONS-induced endocytosis will also be of relevance to other cancer therapies that induce an increase in extracellular RONS.

Abstract: Due to the attraction of plasma technologies as a clean and efficient means of surface modification, significant research has gone into the physical and chemical aspects of polymer functionalization. In this study, it was shown that the use of an atmospheric plasma jet can efficiently modify the surface of polyethylene terephthalate s les and change their hydrophobic properties to more hydrophilic characteristics. The dependence on the changes with respect to time, distance, and atomic oxygen (O I) intensity were considered as factors. It was found that with closer proximity to the plasma source (without causing thermal degradation) and with increasing levels of O I, that the changes of water contact angle and surface free energy can be maximized. It was also observed that the electron energy distribution function, for a given chemistry, significantly differed with changes in distance from the jet nozzle. This shows that for this type of plasma jet system, the bulk of the chemical reactions occur in the nozzle of the jet and not in the surrounding atmosphere. Therefore, this leads to more efficient energy transfer, higher gas temperatures, and better surface activation of s les when compared to systems that produce external chemical reactions due to more diffusion in the surrounding atmosphere and loss of reactive species to other atoms and molecules that are present.

Abstract: Point-particle direct numerical simulations have been employed to quantify the turbulence modulation and particle responses in a turbulent particle-laden jet in the two-way coupled regime with an inlet Reynolds number based on bulk velocity and jet diameter $({D_j})$ of ~10 000. The investigation focuses on three cases with inlet bulk Stokes numbers of 0.3, 1.4 and 11.2. Special care is taken to account for the particle–gas slip velocity and non-uniform particle concentrations at the nozzle outlet, enabling a reasonable prediction of particle velocity and concentration fields. Turbulence modulation is quantified by the variation of the gas-phase turbulent kinetic energy (TKE). The presence of the particle phase is found to d the gas-phase TKE in the near-field region within $5{D_j}$ from the inlet but subsequently increases the TKE in the intermediate region of (5–20) D j . An analysis of the gas-phase TKE transport equation reveals that the direct impact of the particle phase is to dissipate TKE via the particle-induced source term. However, the finite inertia of the particle phase affects the gas-phase velocity gradients, which indirectly affects the TKE production and dissipation, leading to the observed TKE attenuation and enhancement. Particle response to the gas-phase flow is quantified. Particles are found to exhibit notably stronger response to the gas-phase axial velocity than to the radial velocity. A new dimensionless figure is presented that collapses both the axial and radial components of the particle response as a function of the local Stokes number based on their respective integral length scales.

Prediction of Beef Fat Content Simultaneously under Static and Motion Conditions Using near Infrared Spectroscopy

Abstract: Fat content is one of the most important quality indicators for minced beef products. In this study, a multipoint near infrared (NIR) spectrophotometer system, based on a Fabry–Perot interferometer, combined with a four-point photodiode array detector and flexible collimator–probe arrangement, was used for real-time analysis of beef fat content. The system was employed to predict fat content of mixed minced beef s les concurrently under two different conditions: (a) static and slow motion and (b) static and fast motion. Additionally, a separate measurement was conducted to further test the independency of a collimator–probe arrangement by scanning two s les with different fat percentages concurrently under static and motion conditions. Partial least squares regression was employed, obtaining coefficients of determination in calibration ( R 2 c ) of 0.95, confirming a good fit for the three models. The fat contents of s les in the independent set were predicted with reasonable accuracy: r 2 in the range 0.82–0.92 and standard error of prediction in the range 3.05–3.98%. Moreover, the spectral features observed for the probe independency test clearly illustrated the flexibility and independency of the collimator–probe arrangement. This study showed that the multipoint NIR spectroscopy system can predict beef fat content concurrently under static and motion conditions and illustrates its potential use as an in-line monitoring tool at various junctions in a meat processing plant.

Abstract: Hyperspectral imaging (HSI) integrates conventional imaging and spectroscopy, to obtain both spatial and spectral information from a specimen. This technique enables investigators to analyze the chemical composition of traces and simultaneously visualize their spatial distribution. HSI offers significant potential for the detection, visualization, identification and age estimation of forensic traces. The rapid, non-destructive and non-contact features of HSI mark its suitability as an analytical tool for forensic science. This paper provides an overview of the principles, instrumentation and analytical techniques involved in hyperspectral imaging. We describe recent advances in HSI technology motivating forensic science applications, e.g. the development of portable and fast image acquisition systems. Reported forensic science applications are reviewed. Challenges are addressed, such as the analysis of traces on backgrounds encountered in casework, concluded by a summary of possible future applications.
Surface attachment of active antimicrobial coatings onto conventional plastic-based laminates and performance assessment of these materials on the storage life of vacuum packaged beef sub-primals

Abstract: Two antimicrobial coatings, namely Sodium octanoate and Auranta FV (a commercial antimicrobial composed of bioflavonoids, citric, malic, lactic, and caprylic acids) were used. These two antimicrobials were surface coated onto the inner polyethylene layer of cold plasma treated polyamide films using beef gelatin as a carrier and coating polymer. This packaging material was then used to vacuum pack beef sub-primal cuts and stored at 4 °C. A control was prepared using the non-coated commercial laminate and the same vacuum packaged sub-primal beef cuts. During storage, microbial and quality assessments were carried out. Sodium octanoate treated packages significantly (p < 0.05) reduced microbial counts for all bacteria tested with an increase of 7 and 14 days, respectively compared to control s les. No significant effect on pH was observed with any treatment. The results suggested that these food grade antimicrobials have the potential to be used in antimicrobial active packaging applications for beef products.
Adults with intellectual disabilities affected by cancer: critical challenges for the involvement agenda

Abstract: Cancer incidence and prevalence in people with intellectual disabilities are poorly documented. Changing demographic patterns, however, mirror those in the general population with increased longevity leading to higher numbers of people developing cancer in the future. Little is known about the service and support needs of this group, but there is a precedent to understand their experiences to inform research and service development if appropriately tailored approaches are to be adopted. This article outlines a project that sought to develop an advisory forum of adults with intellectual disabilities affected by cancer, to inform the development of a research agenda. The process of this work is described alongside a critical appraisal of the tensions in involving people with intellectual disabilities as advisors on cancer research and practice. We discuss and challenge the underlying ideologies, which lie in the path of fully engaging with marginalized patients.

Abstract: Traditionally, ozone processing within the food industry has focused on solid foods by either gaseous treatment or washing with ozonized water. However, with the FDA's approval of ozone as a direct additive to food, the potential for liquid applications has emerged. This study investigates the effect of ozone processing on microbial inactivation (E. coli ATCC 25922 and NCTC 12900) and quality parameters (color, phenolic content) of cloudy apple juice. Apple juice s les were ozonated at room temperature (20 ± 1.5 °C) with a generated ozone concentration of 0.048 mg O(3) at a constant flow rate of 0.12 L/min and treatment time of 0 to 10 min. E. coli inactivation kinetics in apple juice were described quantitatively by using the Shoulder log-linear and the Weibull model. Ozone treatment of E. coli in apple juice demonstrate that a desired 5 log reduction can be achieved within 5 min. Apple juice color (L*, a*, and b*) and total phenols were significantly affected by ozone concentration and treatment time.

Abstract: Powdered infant formula (PIF) can be the sole source of nutrition for babies and infants. Monitoring conformational changes in protein during manufacture of PIF is critical in order to maintain its nutritional value. This study presents the development of a calibration model for monitoring conformational changes in PIF protein by applying a novel multipoint near-infrared (NIR) spectrometry. NIR spectra were collected for PIF and PIF proteins, casein and whey protein isolate, before and after heat treatment. Results show that principal component analysis showed discrimination between native protein at room temperature and protein conformational changes caused at elevated temperature. Partial least squares regression analysis showed good calibration models with correlation coefficients ranging between 87% and 99% for the prediction of protein quality. This novel multipoint NIR spectrometry could serve as a simple in-line tool to rapidly monitor protein quality during processing stages, contributing to product nutritional value.
Feasibility of near infrared chemical imaging for pharmaceutical cleaning verification

Abstract: In conformity with the 2011 Food and Drug Administration guidance for industry, Process Validation: General Principles and Practices, there is a requirement for new methodologies that support continued cleaning verification of manufacturing equipment. Near infrared chemical imaging (NIR-CI) represents an attractive alternative to the current methodologies for cleaning verification since it represents a direct, rapid and sensitive technology. The aim of this study was to test the feasibility of NIR-CI for cleaning verification. Stainless steel, glass and Perspex surfaces were soiled with selected active pharmaceutical ingredients (APIs) with the resulting hyperspectral data employed to identify and quantify the dried residues. Correlations to high-performance liquid chromatography (HPLC) swab analysis were also made. Linear models were obtained for the number of API classified pixels and applied soil concentration with R 2 values of 0.90 and 0.98 for caffeine and sulfathiazole sodium salt, respectively, on stainless steel coupons. In the case of caffeine soils on borosilicate glass and Perspex coupons, the obtained R 2 values were 0.95 and 0.96, respectively. These R 2 values suggest a linear relationship between the number of API classified pixels and applied soil concentration. The calculated detection limits for caffeine and sulfathiazole sodium salt soils on stainless steel coupons were 48 μg and 18 μg 50 mm −2 , respectively. For caffeine soils on borosilicate glass and Perspex coupons the calculated detection limits were 33 μg and 27 μg 50 mm −2 , respectively. However, visual inspection of the NIR-CI following the classification function point to limits of detection closer to 1 μg 50 mm −2 or lower. High correlations ( R 2 of 0.91 and 0.93 for caffeine and sulfathiazole sodium salt, respectively) were also obtained between NIR-CI API soil quantification and swabbing/HPLC analysis. This research points to possible future use of portable CI systems for continued cleaning verification of pharmaceutical residues.

Challenges in Model Development for Meat Composition Using Multipoint NIR Spectroscopy from At‐Line to In‐Line Monitoring

Abstract: This study evaluates the efficiency of multipoint near-infrared spectroscopy (NIRS) to predict the fat and moisture content of minced beef s les both in at-line and on-line modes. Additionally, it aims at identifying the obstacles that can be encountered in the path of performing in-line monitoring. Near-infrared (NIR) reflectance spectra of minced beef s les were collected using an NIR spectrophotometer, employing a Fabry-Perot interferometer. Partial least squares regression (PLSR) models based on reference values from proximate analysis yielded calibration coefficients of determination (Rc2) of 0.96 for both fat and moisture. For an independent batch of s les, fat was estimated with a prediction coefficient of determination (Rp2) of 0.87 and 0.82 for the s les in at-line and on-line modes, respectively. All the models were found to have good prediction accuracy however, a higher bias was observed for predictions under on-line mode. Overall results from this study illustrate that multipoint NIR systems combined with multivariate analysis has potential as a process analytical technology (PAT) tool for monitoring process parameters such as fat and moisture in the meat industry, providing real-time spectral and spatial information.

Abstract: Scanning electron microscopy and X-ray microtomography are useful methods for high resolution shape imaging. Visible microscopy is also common, however, developing a low-cost and customizable system for surface and shape investigation of optically active particles is challenging. In this work, we demonstrate an assembly offering good light sensitivity, flexibility of illumination and contrasts from varying angles. The design was applied, together with recent programs for focus-stacking, to analyze crystals of taurine, L-glutamic acid, acetylsalicylic acid, and copper sulfate, along with digital 3D-360° modelling of phosphorescent [Ru(bpy) 3 ]Cl 2 and strontium aluminate particles. We further tested the approach for real time monitoring of size, shape and texture analysis of fat filled milk particles and acid whey powders. The findings show proof of concept for detailed feature imaging of particles directly from the process environment.

Oxidative stress induced by plasma-activated water stimulates astaxanthin production in Phaffia rhodozyma

Abstract: Astaxanthin is used extensively in the nutraceutical, aquaculture, and cosmetic industries. The current market necessitates higher astaxanthin production from Phaffia rhodozyma (P. rhodozyma) due to its higher cost compared to chemical synthesis. In this study, a bubble discharge reactor was developed to generate plasma-activated water (PAW) to produce PAW-made yeast malt (YM) medium. Due to oxidative stress induced by PAW, strains cultured in 15 and 30 min-treated PAW-made medium produced 7.9 ± 1.2 % and 12.6 ± 1.4 % more carotenoids with 15.5 ± 3.3 % and 22.1 ± 1.3 % more astaxanthin, respectively. Reactive oxygen species (ROS) assay results showed that ROS generated by plasma-water interactions elevated intracellular ROS levels. Proteomic analysis revealed increased expression of proteins involved in the cellular response to oxidative stress as well as carotenoid biosynthesis, both of which contribute to higher yields of astaxanthin. Overall, this study supports the potential of PAW to increase astaxanthin yields for industrial-scale production.

Abstract: In recent years, the pharmaceutical industry has focused on a better understanding of the real-time manufacturing process with online measurements. Near infrared (NIR) spectroscopy is perhaps the most used and acceptable technique in such a highly regulated industry. However, one of the big challenges in using NIR systems for in-line manufacturing processes is the acquisition of measurements in real-time simultaneously at different locations. To this end, a series of different optical set-ups were investigated. Results are presented here using a multipoint NIR system based on a Fabry–Pérot Interferometer capable of recording simultaneously four independent spectra in four different points. NIR spectra of cellulose and sucrose granule mixtures were evaluated by the partial least-squares (PLS) regression method. By interpreting score values, loading of the principal components and the root mean square errors of cross-validation ( RMSECV), differences between two sensors composed of fibre optics and the effect of s ling (static or motion) were analysed. Sensors that allow large illumination and detection areas against small granule sizes showed better results in reducing the prediction errors ( RMSECV = 1.8%) than sensors with small illumination and detection areas against s les with large granule sizes ( RMSECV = 9.6%). These results are important to better understand errors associated with optic fibre probes and their configuration in multipoint NIR spectroscopy for monitoring the pharmaceutical manufacturing process.

Controlled cytotoxicity of plasma treated water formulated by open-air hybrid mode discharge

Abstract: Plasma treated liquids (PTLs) provide a means to convey a broad range of effects of relevance for food, environmental, or clinical decontamination, plant growth promotion, and therapeutic applications. Devising the reactive species ingredients and controlling the biological response of PTLs are of great interest. We demonstrate an approach by using an open-air hybrid mode discharge (HMD) to control the principal reactive species composition within plasma treated water (PTW), which is then demonstrated to regulate the cytotoxicity of PTW. The cytotoxicity of HMD produced PTW demonstrates a non-monotonic change over the discharge time. Although hydrogen peroxide and nitrite are not the sole effectors for cell death caused by PTW, using them as principal reactive species indicators, cytotoxicity can be removed and/or enhanced by formulating their concentrations and composition through adjusting the discharge mode and time on-line during PTW generation without the addition of additional working gas or chemical scavengers. This work demonstrates that a hybrid mode discharge can be employed to generate a PTW formulation to control a biological response such as cytotoxicity. This provides insights into how plasma treated liquids may be harnessed for biological applications in a specific and controllable manner.

Abstract: The use of plasma-activated liquids such as PBS, medium or simply plasma-activated water (PAW) has been receiving increasing attention for applications in cancer treatments. Amongst the reactive species contained in these solutions, hydrogen peroxide appears to play a pivotal role in causing cytotoxic effects. H2O2 concentrations can be correlated with reduced cell viability and growth and used as an indicator of the potential efficacy of a plasma-activated liquid. To investigate the cytotoxic mediators generated in water specific to high-voltage DBD-ACP. Using a high-voltage dielectric barrier atmospheric cold plasma (DBD-ACP) system, we examined PAW-mediated cytotoxic effects on different mammalian cell lines employing a set-up where short-lived reactive species can be discounted and activated liquids with long-term stability are generated. The PAW potency could be modulated using voltage level, treatment time and post-treatment storage time and target-related characteristics such as surface to volume ratio. All of these parameters effected cell viability in a hydrogen peroxide concentration correlated manner. The susceptibility of two cancer cell lines to PAW was similar to that observed for two non-cancer cell lines and the toxicity of plasma-activated water exceeded that of the corresponding hydrogen peroxide concentrations. In cytotoxic plasma activated water an essential role for H2O2 has been demonstrated multi-fold, yet further contributing factors are apparent and remain to be identified.

Abstract: Gold nanoparticles (AuNP) have potential as both diagnostic and therapeutic vehicles. However, selective targeting and uptake in cancer cells remains challenging. Cold atmospheric plasma (CAP) can be combined with AuNP to achieve synergistic anti-cancer cytotoxicity. To explore synergistic mechanisms, we demonstrate both rate of AuNP uptake and total amount accumulated in U373MG Glioblastoma multiforme (GBM) cells are significantly increased when exposed to 75 kV CAP generated by dielectric barrier discharge. No significant changes in the physical parameters of AuNP were caused by CAP but active transport mechanisms were stimulated in cells. Unlike many other biological effects of CAP, long-lived reactive species were not involved, and plasma-activated liquids did not replicate the effect. Chemical effects induced by direct and indirect exposure to CAP appears the dominant mediator of enhanced uptake. Transient physical alterations of membrane integrity played a minor role. 3D-reconstruction of deconvoluted confocal images confirmed AuNP accumulation in lysosomes and other acidic vesicles, which will be useful for future drug delivery and diagnostic strategies. Toxicity of AuNP significantly increased by 25-fold when combined with CAP. Our data indicate that direct exposure to CAP activates AuNP-dependent cytotoxicity by increasing AuNP endocytosis and trafficking to lysosomes in U373MG cells.
