Abstract: Molecular-based classifications of gastric cancer (GC) were recently proposed, but few of them robustly predict clinical outcomes. While mutation and expression signature of protein-coding genes were used in previous molecular subtyping methods, the noncoding genome in GC remains largely unexplored. Here, we developed the fast long-noncoding RNA analysis (FLORA) method to study RNA sequencing data of GC cases, and prioritized tumor-specific long-noncoding RNAs (lncRNAs) by integrating clinical and multi-omic data. We uncovered 1235 tumor-specific lncRNAs, based on which three subtypes were identified. The lncRNA-based subtype 3 (L3) represented a subgroup of intestinal GC with worse survival, characterized by prevalent TP53 mutations, chromatin instability, hypomethylation, and over-expression of oncogenic lncRNAs. In contrast, the lncRNA-based subtype 1 (L1) has the best survival outcome, while LINC01614 expression further segregated a subgroup of L1 cases with worse survival and increased chance of developing distal metastasis. We demonstrated that LINC01614 over-expression is an independent prognostic factor in L1 and network-based functional prediction implicated its relevance to cell migration. Over-expression and CRISPR-Cas9-guided knockout experiments further validated the functions of LINC01614 in promoting GC cell growth and migration. Altogether, we proposed a lncRNA-based molecular subtype of GC that robustly predicts patient survival and validated LINC01614 as an oncogenic lncRNA that promotes GC proliferation and migration.

Abstract: Initiation of smoking habit often occurs in early teens due to the interaction of teenagers with the social environment. The community has an important role in protecting teenagers from the habit of smoking. The research was aimed to explore the experience of the community in protecting the early-teenagers from smoking habit from the perspective of Indonesian people. The qualitative study used the descriptive phenomenology approach on 100 participants (early-teenagers, parents, teachers, and school nurses) who were selected through snowball s ling method. Data collection was carried out through in-depth interviews using open-ended questions. The data were analysed manually using the data source triangulation method. Six research topics were concluded: 1) screening of habits related to health, 2) smoking bans, 3) punishment for smoking habit, 4) smoke-free counseling, 5) evaluation of efforts done, and 6) follow-up of smoking-free programs. The results of the study indicated the importance of consistent efforts to prevent the initiation of smoking habit in early-teenagers both at school and at home. The communities around them at school and home need to have people with healthy non-smoking habit as good role models for the teens.

Abstract: The aim of this study was to examine the associations of food store choice with food consumption among urban slum women. A cross-sectional survey was carried out among 188 urban slum women (19-50 years old) in Jakarta, Indonesia. A semiquantitative food frequency questionnaire was used to assess food consumption. Associations between food consumption and food store choice were tested by linear regression. This study found that frequencies of buying food from small shops ( warung), street food vendors, and modern food stores were significantly associated with consumption of snacks, mixed dishes, and fruit respectively. In addition, buying food from traditional markets and small cafes ( warung makan) was not significantly associated with particular types of food consumption. As modern food stores are rarely utilized by these women, small shops ( warung) and street food vendors are likely to be important channels to improve slum dwellers’ diet.
