Abstract: The Ruwai skarn deposit is the largest polymetallic skarn deposit in Borneo and is located in the Schwaner Mountains. The skarns and massive orebodies are hosted in marble of the Jurassic Ketapang Complex, which was intruded by Cretaceous Sukadana granitoids. The prograde-stage garnet and retrograde-stage titanite yielded U-Pb ages of 97.0 ± 1.8 to 94.2 ± 10.3 Ma and 96.0 ± 2.9 to 95.0 ± 2.0 Ma, respectively. These ages are similar to Re-Os ages obtained on sulfides (96.0 ± 2.3 Ma) and magnetite (99.3 ± 3.6 Ma). The U-Pb zircon ages reveal that magmatism at Ruwai occurred in three phases, including the Early Cretaceous (ca. 145.7 and 106.7–105.7 Ma andesite-dacite), Late Cretaceous (ca. 99.7–97.1 Ma diorite-granodiorite), and late Miocene (ca. 10.94–9.51 Ma diorite-dolerite). Based on geochemical and stable isotopic data (C-O-S) the Ruwai skarn ores are interpreted to have formed from oxidized fluids at ca. 160 to 670°C. The ore-forming fluids and metals were mostly magmatic in origin but with significant crustal input. Ruwai skarn mineralization occurred in the Late Cretaceous, associated with Paleo-Pacific subduction beneath Sundaland after the Southwest Borneo accretion. Ruwai is the first occurrence of Cretaceous mineralization recognized in the Central Borneo metallogenic belt.

Abstract: Drought and salinity are the major factors that limit chickpea production worldwide. We performed whole transcriptome analyses of chickpea genotypes to investigate the molecular basis of drought and salinity stress response/adaptation. Phenotypic analyses confirmed the contrasting responses of the chickpea genotypes to drought or salinity stress. RNA-seq of the roots of drought and salinity related genotypes was carried out under control and stress conditions at vegetative and/or reproductive stages. Comparative analysis of the transcriptomes revealed ergent gene expression in the chickpea genotypes at different developmental stages. We identified a total of 4954 and 5545 genes exclusively regulated in drought-tolerant and salinity-tolerant genotypes, respectively. A significant fraction (~47%) of the transcription factor encoding genes showed differential expression under stress. The key enzymes involved in metabolic pathways, such as carbohydrate metabolism, photosynthesis, lipid metabolism, generation of precursor metabolites/energy, protein modification, redox homeostasis and cell wall component biogenesis, were affected by drought and/or salinity stresses. Interestingly, transcript isoforms showed expression specificity across the chickpea genotypes and/or developmental stages as illustrated by the AP2-EREBP family members. Our findings provide insights into the transcriptome dynamics and components of regulatory network associated with drought and salinity stress responses in chickpea.

Abstract: Cicer arietinum L. (chickpea) is the third most important food legume crop. We have generated the draft sequence of a desi-type chickpea genome using next-generation sequencing platforms, bacterial artificial chromosome end sequences and a genetic map. The 520-Mb assembly covers 70% of the predicted 740-Mb genome length, and more than 80% of the gene space. Genome analysis predicts the presence of 27,571 genes and 210 Mb as repeat elements. The gene expression analysis performed using 274 million RNA-Seq reads identified several tissue-specific and stress-responsive genes. Although segmental duplicated blocks are observed, the chickpea genome does not exhibit any indication of recent whole-genome duplication. Nucleotide ersity analysis provides an assessment of a narrow genetic base within the chickpea cultivars. We have developed a resource for genetic markers by comparing the genome sequences of one wild and three cultivated chickpea genotypes. The draft genome sequence is expected to facilitate genetic enhancement and breeding to develop improved chickpea varieties.

Abstract: The geochemical analysis of trace elements in rutile (e.g., Pb, U, and Zr) is routinely used to extract information on the nature and timing of geological events. However, the mobility of trace elements can affect age and temperature determinations, with the controlling mechanisms for mobility still debated. To further this debate, we use laser-ablation–inductively coupled plasma–mass spectrometry and atom probe tomography to characterize the micro- to nanoscale distribution of trace elements in rutile sourced from the Capricorn orogen, Western Australia. At the & µm scale, there is no significant trace-element variation in single grains, and a concordant U-Pb crystallization age of 1872 ± 6 Ma (2σ) shows no evidence of isotopic disturbance. At the nanoscale, clusters as much as 20 nm in size and enriched in trace elements (Al, Cr, Pb, and V) are observed. The 207Pb/206Pb ratio of 0.176 ± 0.040 (2σ) obtained from clusters indicates that they formed after crystallization, potentially during regional metamorphism. We interpret the clusters to have formed by the entrapment of mobile trace elements in transient sites of radiation damage during upper hibolite facies metamorphism. The entrapment would affect the activation energy for volume diffusion of elements present in the cluster. The low number and density of clusters provides constraints on the time over which clusters formed, indicating that peak metamorphic temperatures are short-lived, & m.y. events. Our results indicate that the use of trace elements to estimate volume diffusion in rutile is more complex than assuming a homogeneous medium.

Abstract: Transcriptomic data of C. roseus offering le sequence resources for providing better insights into gene ersity: large resource of genic SSR markers to accelerate genomic studies and breeding in Catharanthus . Next-generation sequencing is an efficient system for generating high-throughput complete transcripts/genes and developing molecular markers. We present here the transcriptome sequencing of a 26-day-old Catharanthus roseus seedling tissue using Illumina GAIIX platform that resulted in a total of 3.37 Gb of nucleotide sequence data comprising 29,964,104 reads which were de novo assembled into 26,581 unigenes. Based on similarity searches 58 % of the unigenes were annotated of which 13,580 unique transcripts were assigned 5016 gene ontology terms. Further, 7,687 of the unigenes were found to have Cluster of Orthologous Group classifications, and 4,006 were assigned to 289 Kyoto Encyclopedia of Genes and Genome pathways. Also, 5,221 (19.64 %) of transcripts were distributed to 81 known transcription factor (TF) families. In-silico analysis of the transcriptome resulted in identification of 11,004 SSRs in 26.62 % transcripts from which 2,520 SSR markers were designed which exhibited a non-random pattern of distribution. The most abundant was the trinucleotide repeats (AAG/CTT) followed by the dinucleotide repeats (AG/CT). Location specific analysis of SSRs revealed that SSRs were preferentially associated with the 5'-UTRs with a predicted role in regulation of gene expression. A PCR validation of a set of 48 primers revealed 97.9 % successful lification, and 76.6 % of them showed polymorphism across different Catharanthus species as well as accessions of C. roseus. In summary, this study will provide an insight into understanding the seedling development and resources for novel gene discovery and SSR development for utilization in marker-assisted selective breeding in C. roseus.
