Ubiquitous karst hydrological control on speleothem oxygen isotope variability in a global study

Abstract: Speleothem oxygen isotopic (δ 18 O) records are used to reconstruct past hydroclimate yet records from the same cave do not always replicate. We use a global database of speleothem δ 18 O to quantify the replicability of records to show that disagreement is common worldwide, occurs across timescales and is unrelated to climate, depth or lithology. Our global analysis demonstrates that within-cave differences in mean speleothem δ 18 O values are consistent with those of dripwater, supporting a ubiquitous influence of flowpaths. We present a case study of four new stalagmite records from Golgotha Cave, southwest Australia, where the isotopic differences between them are informed by cave monitoring. It is demonstrated that karst hydrology is a major driver of within-cave speleothem and dripwater δ 18 O variability, primarily due to the influence of fractures on flowpaths. Applying our understanding of water movement through fractures assists in quantitative reconstruction of past climate variability from speleothem δ 18 O records.

Abstract: Aluminium (Al) is highly toxic to plant growth, with soluble concentrations being elevated in the ∼40% of arable soils worldwide that are acidic. Determining the distribution of Al in plant tissues is important for understanding the mechanisms by which it is toxic and how some plants tolerate high concentrations. Synchrotron- and laboratory-based X-ray fluorescence microscopy (XFM) is a powerful technique to quantitatively analyse the distribution of elements, including in hydrated and living plants. However, analysis of light elements (z & phosphorus) is extremely challenging due to signal losses in air, and the unsuitability of vacuum environments for (fresh) hydrated plant tissues. This study uses XFM in a helium environment to avoid Al signal loss to reveal the distribution of Al in hydrated plant tissues of Tea (Camellia sinensis). The results show that Al occurs in localised areas across the foliar surface, whereas in cross-sections Al is almost exclusively concentrated in the apoplastic space above and in between adaxial epidermal cells. This distribution of Al is related to the Al tolerance of this species, and accumulation of phytotoxic elements in the apoplastic space, away from sensitive processes such as photosynthesis in the palisade mesophyll cells, is a common tolerance mechanism reported in many different plant species. This study develops an XFM method on both synchrotron and laboratory sources that overcomes the drawbacks of existing analytical techniques, permitting measurement of light elements down to Al in (fresh) hydrated plant tissues.

Spatiotemporal patterns of wheat response to Pyrenophora tritici-repentis in asymptomatic regions revealed by transcriptomic and X-ray fluorescence microscopy analyses

Abstract: Pathogen attacks elicit dynamic and widespread molecular responses in plants. While our understanding of plant responses has advanced considerably, little is known of the molecular responses in the asymptomatic ‘green’ regions adjoining lesions. Here, we explore gene expression data and high-resolution elemental imaging to report the spatiotemporal changes in the asymptomatic green region of susceptible and moderately resistant wheat cultivars infected with a necrotrophic fungal pathogen, Pyrenophora tritici-repentis. We show, with improved spatiotemporal resolution, that calcium oscillations are modified in the susceptible cultivar, resulting in ‘frozen’ host defence signals at the mature disease stage, and silencing of the host’s recognition and defence mechanisms that would otherwise protect it from further attacks. In contrast, calcium accumulation and a heightened defence response were observed in the moderately resistant cultivar in the later stage of disease development. Furthermore, in the susceptible interaction, the asymptomatic green region was unable to recover after disease disruption. Our targeted s ling technique also enabled detection of eight previously predicted proteinaceous effectors in addition to the known ToxA effector. Collectively, our results highlight the benefits of spatially resolved molecular analysis and nutrient mapping to provide high-resolution spatiotemporal snapshots of host–pathogen interactions, paving the way for disentangling complex disease interactions in plants.

Abstract: Measuring the amount of carbon captured in deep-sea limestones is fundamental to understanding the long-term carbon cycle because pelagic limestones represent Earth’s largest carbon sink since the mid-Mesozoic. However, their contribution to the long-term carbon cycle is poorly quantified. Here, we use X-ray fluorescence and scanning X-ray diffraction microscopy for high-resolution chemical and structural analysis of pelagic limestone from the Paleocene Kaiwhata Formation in New Zealand. We identify densely packed diagenetic micro-dissolution seams that are invisible to light and electron-beam microscopes in most cases. Mass-balance calculations indicate that in idual seams remove ~50% of the calcite mud matrix while their bulk-s le carbon loss adds up to ~10%. The liberated carbon is trapped in situ as calcite cement or returned to the ocean during physical compaction or soft-sediment deformation. We suggest micro-dissolution structures may play an important role in the long-term carbon cycle by modulating carbon exchange between the geosphere and hydrosphere.
