Summary
Ankylosing spondylitis (AS) and Crohn's disease (CD) are common immune-mediated diseases affecting primarily the joints of the spine and the gut respectively. Genes play a major role in determining the risk of each disease, and it is likely that those genes cause the disease by interaction with some environmental factor, most likely bacteria residing in the gut. This study aims to test that hypothesis by profiling the bacteria in the gut of patients with the diseases and healthy subjects.

Summary
Ankylosing spondylitis is a debilitating arthritic disease, susceptibility to which is conferred by genes of the immune system, particularly HLA-B27, and following gastrointestinal infection. Using mass spectrometry we will identify bacterial peptides bound to HLA-B27 on infected cells that may trigger an autoimmune response. Defining the self peptides that remain the targets of autoimmunity will unravel the molecular and cellular mechanisms if disease and identify peptides for immunotherapy.

Ankylosing spondylitis (AS) is a form of arthritis targeting the spine and pelvis that causes uncontrolled bone formation resulting in complete joint fusion, severe disability and even death for which no therapies are currently available. Using a mouse model that closely replicates the human disease we will characterise the changes causing this joint fusion and identify possible new targets to develop novel treatments.

Ankylosing spondylitis (AS) is the prototypic condition of a group of types of inflammatory arthritis called 'seronegative spondyloarthropathies'. These conditions are the most common form of inflammatory arthritis in white populations and occur worldwide. One third of the risk of developing AS is determined by genes within a region called the 'major histocompatibility complex' (MHC), in addition to the gene HLA-B27, the main gene causing AS. We aim to identify the remaining MHC genes.

Ankylosing spondylitis (AS) is a common inflammatory arthritis which causes primarily back pain and stiffness, and affects 1-250 individuals. Our group identified association between tagging genetic markers in the gene IL23R and AS, and our preliminary data suggests some related genes are involved as well. This study aims to identify the key genetic variants involved and determine the mechanism by which they cause AS.
Ankylosing spondylitis (AS) is a chronic arthritis which causes severe back and joint pain in young men and women. It can be difficult to diagnose as it takes years to show up on x-ray, and by then the joints are already damaged. This study looks at new ways to diagnose people with AS much earlier, which will allow earlier treatment and better outcomes. The study will also look at the role of lifestyle factors such as diet and physical activity which might impact on AS.

Ankylosing spondylitis is a chronic inflammatory disease affecting the spine and causing back pain. The diagnosis of the disease is delayed by up to 10 years due to lack of accurate tests. We aim to identify molecular signatures of the disease that might be used to distinguish inflammatory processes typical of the disease and other causes of back pain. This would allow earlier and more accurate diagnosis of the disease and result earlier patient treatment and better health outcomes.
