Summary
Scabies is an underlying cause of poor health in indigenous communities worldwide. Crusted scabies is a poorly understood, life-threatening form of the disease compromising the success of community control strategies. This research compares the immune response in the skin of scabies patients, and in a world-first animal model of human scabies. This will reveal specific immune defects predisposing to disease, ultimately resulting in improved skin health for disadvantaged communities
Squamous cell carcinoma of the skin is extremely common in Australia, resulting in disfiguring surgeries and deaths. Although cumulative sun exposure is important, some people are very susceptible, and we do not know why. This project hinges on the notion that skin cancer is a complex (many genes involved). We will utilize novel systems to harness this complexity to understand why some people are resistant and others very susceptible so as to design appropriate control measures and treatments.

Australians are subject to high levels of sun exposure , that consequently can lead to skin damage and skin cancer. The specific aims of our reseearch are to investigate the role of skin mast cells in the limitation of skin alterations associated with chronic low-dose ultraviolet B exposure. Understanding the innate mechanisms that protect against excessive skin damage and cancer might aid the development of better treatment modalities in the future.

Summary
Herpes simplex viral causes infections of the skin, eye, brain, genitals and of the newborn. HSV enters via breaks in the skin where it encounters dendritic cells, which are important for local control of the virus locally and for T cell responses. We will use two photon microscopy to visualise fluorescent HSV in mice in which DC appear green under the microscope. This will help us understand the interaction between HSV and DC in vivo which will aid design of topical antivirals and vaccines.

Our goal is to target natural reduction/oxidation (redox) modifiers, i.e. niacin and sulforaphane, to the skin deeper layers to treat photoageing. Both drugs have been separately shown to prevent UV induced skin cancer. The outcomes of this project will be safer and more effective prevention and treatment of sun damaged skin. This project can provide health benefits to Australians from improved treatment of sun damage, and economic benefits to the nation.

Summary
Neutrophils are key defenders against bacterial infections. In this application we will test the hypothesis that perivascular macrophages play a critical role in the recruitment of neutrophils to site of cutaneous infection, and that these cells are targeted and destroyed by bacterial virulence factors. Our studies will gain novel insight into the leukocyte homing paradigm and shed new light on the mechanisms of microbial immuno-evasion.

Summary
Basal and squamous cell skin cancers are the leading cancers in Australia, with about 2% of the population developing them each year. As well as sun exposure, a number of other factors have been thought to effect these cancers. This study will examine if factors such as smoking, alcohol consumption and infection with certain skin related human papillomaviruses also increase their risk. Even a small effect may make a big difference when it comes to preventing these common cancers.

Summary
Harlequin Ichthyosis is a severe inherited skin disease caused by mutations in a protein which regulates how skin cells control their levels of lipids. Treatments for this disease are limited and do little to improve patients condition. We believe we have found a new way to treat this condition by altering tissue inflammation. This grant will undertake important experiments aimed at developing new therapies for this currently incurable disease.
