Squamous cell carcinoma of the skin is extremely common in Australia, resulting in disfiguring surgeries and deaths. Although cumulative sun exposure is important, some people are very susceptible, and we do not know why. This project hinges on the notion that skin cancer is a complex (many genes involved). We will utilize novel systems to harness this complexity to understand why some people are resistant and others very susceptible so as to design appropriate control measures and treatments.

Summary
The skin’s immune system is a key defence against skin cancer. Transplant recipients, who are chronically and profoundly immune suppressed to prevent rejection, have a 50-80 fold increase in skin cancer risk. Nicotinamide (vitamin B3) reduced skin cancer in our recent Phase 3 trial in immune competent individuals. A pilot study in renal transplant patients showed similar results. This project will determine at the Phase 3 level whether nicotinamide reduces skin cancers after kidney transplant.

The stem cells of the epidermis or outer lining of the skin are an important group of cells with a role in normal cell replenishment, in wound healing and in skin disorders such as psoriasis and cancer. These primitive cells remain in the skin during the lifetime of an individual and are responsible for the production of mature, functional cells that form a protective barrier against the environment and pathogens. We have recently developed a strategy for the isolation of stem cells of the skin ....The stem cells of the epidermis or outer lining of the skin are an important group of cells with a role in normal cell replenishment, in wound healing and in skin disorders such as psoriasis and cancer. We have recently developed a strategy for the isolation of stem cells of the skin which places us in a unique position to further study these cells and determine possible therapeutic approaches for the future. The aims of this project encompass testing the potency of skin stem cells (i) to reform complex structures such as a mature epidermis, hair follicles and sebaceous glands; and (ii) to give rise to cells from other tissues such as muscle and liver. Schistosoma haematobium causes genitourinary schistosomiasis, a serious disease that affects reproductive health, urinary system health and potentially bladder cancer. This species is the most pathogenic species of all schistosomes, but comparatively less is know about it than other species because of a lack of a suitable model. We need a suitable model host for this important parasite. This project will test whether newborn pigs can be used as laboratory models.
