Despite this, there is still much uncertainty about what causes some people to develop this cancer. We believe that some of this uncertainty arises because there are actually different causes of cancer for different sections of the colon and rectum. This study aims to collect information from 1000 people with colorectal cancer and 1000 people without colorectal cancer, and compare various factors such as their genetic makeup, diet, AND lifestyle. We expect to find that some factors are more common in people with cancer in the upper parts of the bowel, while other factors are more common in people with cancer in the lower parts of the bowel. This kind of information can be used to design programs to prevent colorectal cancer. A number of factors, particularly age and family history, influence the risk of prostate cancer but, in contrast to breast cancer, we don't know what is the risk of developing prostate cancer over a period of time for a man with a specific set of risk factors. In fact, while a number of statistical models have been developed that use a woman's risk factor profile to estimate her risk of breast cancer, none is currently a ....Prostate cancer is a major cause of disability and death in Australian men. A number of factors, particularly age and family history, influence the risk of prostate cancer but, in contrast to breast cancer, we don't know what is the risk of developing prostate cancer over a period of time for a man with a specific set of risk factors. In fact, while a number of statistical models have been developed that use a woman's risk factor profile to estimate her risk of breast cancer, none is currently available for prostate cancer. We will apply standard statistical methods to existing data from the Australian Risk Factors for Prostate Cancer study and from the Australian Institute of Health and Welfare to develop a prostate cancer risk prediction model. We will test how factor like age, detailed family history, diet, baldness status and possibly previous PSA tests and prostate biopsies predict the risk. After developing the model, we will test the accuracy of the predictions in three ways. First, using existing data from the Australian Prostate Cancer Family Study, we will see whether the number of cases in a group of men is close to the number predicted by the model (calibration). Second, to test whether the model discriminate well men who develop prostate cancer from those who do not, we will collect family trees in a sample from the Melbourne Collaborative Cohort Study. We will use these data also to estimate the optimal cut point: men above this level of risk will be considered at high risk. Third, we will apply the model to existing data from the Dutch Prostate Cancer Family Study (DPCFS) to test whether the optimal cut point identify high-risk men and to validate the model in a non-Australian population. Automated Mammographic Measures That Predict Breast Cancer Risk

Summary
Mammographic density (MD) is one of the strongest predictors of breast cancer risk but its impractical measurement prevents its use in a clinical setting. An automated measure of MD would allow screening programs to identify and target women at higher risk of breast cancer which could lead to earlier diagnoses and better breast cancer outcomes. We aim to develop an automated measurement, maximized by its ability to predict breast cancer risk, and applicable to both film and digital mammograms.

The prevalence of Depressive Disorders among children and adolescents was estimated in the Australian National Child and Adolescent Mental Health Survey to be 3.7%. This means that at any single point of time, approximately 138,000 Australian children and adolescents are experiencing a Depressive Disorder. Furthermore, the National Survey found that less than half (46%) of those with a Depressive Disorder received any help over a 6 month period prior to the survey, with only 8% attending a menta ....The prevalence of Depressive Disorders among children and adolescents was estimated in the Australian National Child and Adolescent Mental Health Survey to be 3.7%. This means that at any single point of time, approximately 138,000 Australian children and adolescents are experiencing a Depressive Disorder. Furthermore, the National Survey found that less than half (46%) of those with a Depressive Disorder received any help over a 6 month period prior to the survey, with only 8% attending a mental health clinic, and only 4% attending a hospital-based Department of Psychiatry. These findings emphasise the importance of finding alternative approaches to help the large number of young people with depression who do not receive help from professional services. This application seeks funding to evaluate the beyondblue Schools Research Initiative. The key features of the initiative are the development of a strong partnership between the health and education sectors and a focus on both individual-level and school-level risk factors. This has enabled us to provide a much longer duration of intervention than previous studies, to test the intervention in several different Australian States, and to utilise a broadly based intervention that includes a range of approaches, each of which have the potential to help reduce adolescent depression. The significance of the project lies in its potential to identify effective interventions which can reduce rates of depression experienced by adolescents, and the quality of the ongoing research partnership we have established across the education and health sectors. Summary
Staphylococcus aureus infections range from boils to life-threatening diseases and are increasingly resistant to antibiotics and difficult to treat. This study follows patients with community-acquired S. aureus infections, and close contacts, for 24 months to see if they carry S. aureus (nose swabs) or develop infection. Our data on risk factors for colonisation and infection will help doctors decide whether to trace and treat contacts of patients to protect households from further infection.

Although CVD occurs much more frequently in older persons, much of the epidemiological information on CVD risk factors and risk estimation comes from studies of middle-aged populations. Recently there has been an increased focus on whether the established relationships hold or differ in the elderly. This has generated debate about the relative value and effectiveness of treating risk factors in elderly people. This study, which is based on comprehensive and long-term risk factor, mortality and morbidity data from the Busselton Health Study and Health in Men Study, will lead to a better understanding of classic and new CVD risk factors in older persons and will contribute positively to the debate about the relative value and effectiveness of attempting to modify risk factors in the elderly. We know that both environmental factors, such as declining socio-economic conditions, and genetic risk factors such as HLA type contribute to the likelihood of an individual developing disease, but current known factors are insufficient to fully accou ....Tuberculosis remains an enormous global health problem. Some 32% of the world population are infected, with over 1 million persons dying each year. The risk of an infected individual developing clinical disease ranges from 2-23% for their lifetime. We know that both environmental factors, such as declining socio-economic conditions, and genetic risk factors such as HLA type contribute to the likelihood of an individual developing disease, but current known factors are insufficient to fully account for the risk attributed to genetics. The aim of this project is to investigate another potential risk factor involved in the development of tuberculosis, that of P2X7 receptor function. A natural compound, ATP, when added to macrophages is able to kill tuberculosis organisms residing within the macrophage. This process occurs when ATP activates the P2X7 receptor. We have recently identified a mutation in the P2X7 receptor, which causes a loss of receptor function. Individuals who have this mutation are unable to respond to ATP and hence may be unable to kill tuberculosis. Our studies will determine if the mutation we have identified in the P2X7 receptor prevents or inhibits ATP mediated killing of mycobacteria. Furthermore we will determine the frequency of this mutation in TB patients and the general population to determine if this mutation in the P2X7 receptor is a risk factor for the development of tuberculosis disease.Read moreRead less
