    Understanding The Role Of CD4 T Cells In Viral Infection: A Means Of Improving Anti-viral Immunotherapy.

    Summary
    Development of therapies to prevent and treat chronic infections is of the highest priority as they cause considerable clinical challenges and on-going health care costs. Efforts to improve treatment of chronic viral infections, such as those caused by HIV, hepatitis C virus and human cytomegalovirus, require a better understanding of the immune responses needed to control these viruses long-term. This proposal will investigate the role of CD4+ T cells in controlling chronic viral infection.

    Despite recent advances in therapeutic options, chronic viral infections, including infection with hepatitis C virus and hepatitis B virus, continue to be a significant cause of morbidity and mortality in Australia and affecting hundreds of millions of people worldwide. This R&D program aims to develop a cheaper drug formulation that is easier to deliver and more stable for transport to remote areas.

    Intestinal helminths (worms) have evolved together with humans and induce strong type 2 immune responses effective at eradicating the parasite, mediating tissue repair and modulating the gut microbiota. My research aims to decipher the roles of type 2 immunity in regulating intestinal health and promoting tissue repair. I also plan to determine how worm infection and type 2 immunity modulate the microbiota the impact of this on health, particularly obesity.

    Defining Immune Regulation And T Cell Responses During Chronic Infectious Disease.

    Chronic infectious diseases such as HIV/AIDS, tuberculosis, malaria and leishmaniasis are responsible for significant morbidity and mortality. They are all characterised by severe immune dysfunction. We will study a parasitic infection to identify important immune cell populations and molecules that promote chronic infectious disease. This knowledge will enable the development of better treatments and vaccines for range of infectious diseases that affect people in many parts of the world.

    Bacterial Outer Membrane Vesicles As Immunomodulatory Agents In Helicobacter Pylori Infection

    Summary
    Chronic inflammation of the stomach is a hallmark of Helicobacter pylori infection, and is a precursor to peptic ulcer disease and cancer. Like many other bacteria, H. pylori sheds spherical blebs from its surface. These blebs bind to stomach cells in vitro and have been found in stomach biopsies of H. pylori-infected subjects. The aims of the work are to investigate the mechanisms whereby H. pylori blebs enter and disseminate within host cells, and how this may contribute to inflammation.

    This project aims to characterise the nature and ontogeny of a novel population of cells with phagocytic capacity that forms a network underlying the capsule of mouse and human liver reminiscent of that formed by Langherans cells in the epidermis of the skin. In this project we will characterise this newly described liver capsular macrophage subset, define their ontogeny and assess their specific functions.

    Harnessing Lipid-reactive Immunity To Combat Mycobacterium Tuberculosis Infection

    Critcial to the survival of Mycobacterium tuberculosis, the causative agent of tuberculosis (TB) is its unique waxy (lipid)-rich cell wall. This proposal aims to target components of its cell wall to devlop novel therapeutic strategies. Specifically, the Australian-Singapore alliance will examine how the immune system "sees" lipid based antigens from M. tuberculosis, and then will ultimately use this information towards the devlopment of novel lipid-based vaccines.

    Understanding The Interplay Between ER Stress And Inflammation

    Chronic inflammatory diseases in the gut and lung affect hundreds of thousands of Australians. We have identified how inflammation causes a type of stress resulting in abnormal protein synthesis in the cells which make the barrier to microbes. Following an infection this process might be the trigger for chronic unresolving inflammatory disease. The further understanding of this process we seek in this project is likely to lead new approaches to treat common inflammatory diseases.
