RNA plays many essential roles in cells, from information transfer and regulation of gene expression to scaffolding macromolecular structures and catalysis. Despite these realisations the current approaches to manipulate RNA are limited in many respects. This project will use synthetic biology approaches to engineer synthetic regulators of RNAs in living cells. These studies will provide new tools for use in biological research and provide insights into how natural proteins control gene expression. Furthermore, this project will use these tools to understand the mechanisms of how proteins are synthesised in mammalian mitochondria.Read moreRead less
Physiology and genetics of barley grain germination in the malting and brewing industries. An international research team will provide new scientific information on barley grain germination. This detailed basic knowledge will be immediately applied in breeding programs that are aimed at improving malting and brewing quality in a commercial context. At the same time, the industry's carbon footprint will be significantly reduced.

Confocal and single molecule microscopes for systems microscopy. This project aims to establish Australia’s first system microscopy facility with dedicated live-cell confocal and single-molecule fluorescence microscopes. In systems microscopy, the imaging workflow is automated so that large and unbiased data sets of the spatiotemporal organisation of molecules and cells can be generated. Combined with statistical and bioinformatics analyses, image-derived data provides system-wide information that is not easily obtainable with other approaches. The project will enable Australian researchers to image and analyse the full complexity of biological systems, potentially transforming cell biology, drug development and understanding the molecular basis of disease. It will also demonstrate how the capacity of microscopy facilities can be enhanced and bias in imaging data reduced by automating data acquisition and mining of image-based data.Read moreRead less
Genome Approaches to Investigate Metabolic Coordination in Plant Cells. Metabolism of C and N in legume nodules requires interaction between the symbiotic bacteria and plant organelles, particularly metabolism in plastids and mitochondria. Fixed N is assimilated through the de novo synthesis of purines in both plastids and mitochondria. However, each of the nine pathway enzymes is encoded by a single gene, indicating each protein is targeted to both organelles. Purine metabolism will provide ....Genome Approaches to Investigate Metabolic Coordination in Plant Cells. Purine metabolism will provide a model to assess the more general occurrence of dual-targeted proteins in plants. The aim is to identify and eventually exploit the signalling mechanism(s) that mediate communication between plastids and mitochondria.Read moreRead less
A proteomic approach to identifying the signaling pathway(s) by which acute oxidative stress causes cell death by apoptosis. Oxidative stress following traumatic injury (heart attack or stroke) is known to activate signaling pathways leading to programmed cell death (apoptosis). The aim of this project is to develop methods to identify the signaling proteins involved. Identifying proteins involved in causing cell death will be useful in developing diagnostic tools as well as providing potential ....A proteomic approach to identifying the signaling pathway(s) by which acute oxidative stress causes cell death by apoptosis. Characterising the transport and delivery of oligonucleotides . Short RNA and DNA molecules represent a class of macromolecules that have great potential, but to facilitate their trafficking across cellular and membrane barriers into specific sites of action is challenging. This project aims to develop and apply novel imaging approaches to track them in cells and tissues. Expected outcomes include better understanding of the trafficking across cellular and membrane barriers, and improved imaging ....Characterising the transport and delivery of oligonucleotides . Short RNA and DNA molecules represent a class of macromolecules that have great potential, but to facilitate their trafficking across cellular and membrane barriers into specific sites of action is challenging. This project aims to develop and apply novel imaging approaches to track them in cells and tissues. Expected outcomes include better understanding of the trafficking across cellular and membrane barriers, and improved imaging tools that could be used to further study the molecular mechanisms of accumulation, metabolism and trafficking of these molecules. This project should provide new strategies to target these molecules to specific cells and tissues, which have significant social and economic benefits to the Australian community.Read moreRead less
Probing JNK MAPK function with peptide inhibitors. It has generally been accepted that the JNK MAPK family of protein kinases is rapidly and potently activated following the exposure of mammalian cells to stresses and cytokines. However, their biological role has remained controversial. We believe that this problem reflects the lack of a generally applicable and specific JNK MAPK inhibitor. In this project we continue our characterisation of a small peptide inhibitor developed in our laboratories. We aim to determine its mechanism of inhibition, the specificity of interaction, and to evolve more effective inhibitors. With these new inhibitors, we can effectively address the biological roles of these kinases.Read moreRead less
