    The Structural Basis For Glutamate Transporter Function

    Summary
    Glutamate transporters are vacuum cleaners in the brain that suck the neurotransmitter glutamate into cells. When the glutamate vacuum breaks down or becomes blocked, glutamate levels outside cells increase, leading to cell death in the brain. This process underlies the damage in many brain diseases including Alzheimer’s disease and stroke. The aim of this project is to understand the mechanism of the glutamate vacuum cleaner so we can develop therapeutics to fix it when it breaks down.

    Summary
    Otitis media with effusion (OME) or ’glue ear’ is a common ear problem in Aboriginal and Torres Strait Islander children. In OME, fluid accumulates in the middle ear. This can cause hearing loss and learning difficulties and lead to surgery. We will see if the use of a simple device in which a child blows up a balloon with their nostrils (nasal balloon autoinflation) helps resolve glue ear, improves hearing and is acceptable for the management of this important ear health problem.

    We will investigate the role of the orbitofrontal cortex (OFC) in decision-making, particularly the effect of hyperactivity in the medial vs. ventrolateral orbitofrontal cortex on decision-making. Hyperactivity in these structures has been linked to obsessive compulsive disorder and, in line with the distinct functions of the different regions of OFC, we develop and test a novel hypothesis as to the psychological and neural bases of the obsessions and compulsions distinctive to that disorder.

    Physical inactivity has been described as a global pandemic and only 15% of Australian adolescents are sufficiently active. The pressure to perform in major school assessments drives many older adolescents to sacrifice physical activity to maximise academic performance. Our team has designed a time-efficient solution to increase physical activity in senior high school students using high intensity interval training which will be evaluated using a cluster randomised controlled trial.

    Most treatments for low back pain that are provided by a GP, physiotherapist or chiropractor are only moderately effective. A new approach is needed. We have developed a new treatment, sensorimotor retraining, based on principles from neuroscience, education, psychology and physiotherapy that we are testing in a randomized controlled trial. If successful this new treatment will provide relief for the millions of Australians who suffer from this debilitating condition.

    Summary
    Most lymphomas respond to therapy but then relapse. Immune cells can attack and kill virus related lymphomas. However, most lymphomas are NOT virus related. We will create immune cells targeting these virus negative lymphomas by inserting artificial receptors into the immune cells. These receptors attach to the lymphoma and activate the immune cells. The immune cells will home to the lymphoma, kill lymphoma cells and persist in the body for many years, preventing lymphoma relapse.
