This proposal will address a gap in our mass spectrometry capabilities and aid in our understanding of the cellular immune response and investigate the chemical diversity of the targets of immunity. This proposal has broad implications in the basic immunology of antigen presentation, in biomarker discovery as well as in the design of new vaccines in infectious disease and cancer and the development of therapies for autoimmune diseases. In addition to these key scientific outcomes this project will also facilitate the training of several new personnel in a skill area for which there is a critical shortage (mass spectrometry) and promote cross-disciplinary skills (immunology, biochemistry, proteomics).Read moreRead less
New technology for the delivery of peptide-based T-cell vaccines for tumour immunotherapy. This project is dedicated to finding simple methods for vaccinating humans and animals against a wide variety of cancers. Should this be achieved millions of Australians will be protected from the devastating consequences of cancer. Consequently there will be great benefits socially, medically and economically.

Malarial parasite surface proteins: structure and interactions of key merozoite antigens. Malaria remains one the most lethal infectious diseases in the world today, being directly responsible for around 2 million deaths annually, many in children under 5 years of age. Related parasitic diseases affect livestock in malaria-endemic regions and more broadly. There is an urgent need for an improved understanding of how these parasites invade target red blood cells. Knowing the structures of key proteins on the parasite cell surface will provide a deeper understanding of host-parasite interactions, as well as a basis for the design of vaccines or drugs that interfere with parasite invasion of host red blood cells. The role of neutral amino acid transport in normal physiology. Future benefits of these studies include the Promotion and Maintenance of Good Health achieved by providing: (1) a better understanding of brain and balance disorders; (2) insights into the damaging effects of the sun and; (3) existing neonatal screening programmes for Hartnup disorder with greater scientific foundation regarding the implications of inheriting this condition, including dietary advce. We will be able to provide Australians who inherit Hartnup disorder with a better understanding of this disease by enabling individuals and families to make choices that lead to healthy, productive and fulfilling lives.Read moreRead less
Intrinsically Unstructured Proteins (IUPs): NMR characterization, prediction, and application to malarial proteome. Determination of protein structures with longer DR by NMR will enrich the DR dataset and provide a deeper understanding of protein structure-function relationships and protein folding pathways. Determination of protein structures with longer DR by NMR will enrich the DR dataset and provide a deeper understanding of protein structure-function relationships and protein folding pathways. The proposal will also provide valuable information in the key applied area of target selection in structural biology. Not all current web services are freely accessible and available services can be improved further by using more reliable training dataset or more effective algorithms, development of a national DR predictor will help Australian structural biologists increase the success rate of structure determination and provide greater insight into a range of proteomes.Read moreRead less
Cellular uptake of glutathione transferases and their development as cell transfection agents. The function and survival of all cells requires the importation of a vast array of biochemical agents. In order for this to occur, these agents must be transported across the cell membrane wall. We are investigating a novel delivery system involving the enzyme glutathione transferase (GST). The function and survival of all cells requires the importation of a vast array of biochemical agents. In order for this to occur, these agents must be transported across the cell membrane wall. We are investigating a novel delivery system involving the enzyme glutathione transferase (GST). By investigating how GSTs cross membranes, we will be able to develop a new technology for the delivery of biologically active molecules into cells. This exciting new technique will have applications in research and in the delivery of therapeutic drugs for the treatment of a range of diseases. Read moreRead less
