Summary
We know very little of the genes that control development of the ovary in female fetuses; most study has focused on the formation of the testis in males. We will use a novel experimental model, a marsupial, where by hormonal treatment of developing males we can switch off testis formation and activate the ovarian pathway. Summary
Levels of abdominal fat are closely correlated with metabolic syndrome. We propose experiments to identify unique characteristics (neurotransmitters or receptors) of neurons deep in the brain that project specifically to this type of fat or other less harmful subcutaneous fat. We can then test the functional significance of these unique elements in animal experimets involving gene knockdown or pharmacological approaches to modify their function and test the effect on fat distribution

Summary
There is a widespread public perception that sperm counts are diminishing. This theory can only be tested by using a representative sample of young men, rather than biased populations (such as men presenting as sperm donors). We have the unique opportunity to test this theory, and to determine any early life events which may lead to reduced sperm counts, such as being growth restricted at birth, exposed to high levels of maternal oestrogens or smoking or being overweight in adolescence.

Summary
Current hormone therapy for breast cancer using inhibitors of oestrogen production results in serious side-effects including bone loss, joint pain and possibly cognitive issues. Our current work is aimed at understanding how oestrogen production is regulated with the goal of developing breast-specific inhibitors of oestrogen production to obviate these problems. In addition, this work is aimed at devising therapeutic intervention to break the linkage between obesity and breast cancer.
