    DIET-INDUCED OBESITY ALTERS THE CENTRAL ACTIONS OF GHRELIN

    Ghrelin is a hormone that primarily targets the brain to increase food intake and body weight. It has evolved to prevent starvation during periods of negative energy balance by promoting energy intake and storage. We have identified central ghrelin resistance in diet-induced obesity (DIO) as a novel disturbed neuroendocrine system that restricts excessive food intake.     While the challenge of understanding and managing the global obesity epidemic is well recognised, the role that nutrition plays is more complex than at first glance. Dietary protein may be of central importance in managing weight and small changes in protein consumption may lead to large changes in energy intake and weight. We propose to look at the effects of dietary protein on appetite, its hormonal regulation, and on the risk of developing metabolic diseases such as diabetes.

    Insulin resistance and diabetes are strongly associated with developing heart disease. Changes in the metabolism of a substance in blood vessels called nitric oxide are seen in diabetes, and are related to heart disease. I will study the relationship between the levels of insulin resistance and markers of nitric oxide metabolism in human subjects, and investigate how this relates to their clinical outcomes after cardiac surgery.

    The prevalence of maternal obesity is alarmingly high in Australia and the western world. However, there is little understanding of how obesity of the mother affects the development of the kidney of the fetus and, in turn, how this affects the risk of these offspring developing cardiovascular and renal disease in adulthood. I hope to address the gaps in this knowledge in my studies. This information is critical in order to develop appropriate health care to those born to obese mothers.

    Obesity is a growing epidemic in Australia. Whilst it is well known that obesity has many detrimental cardiovascular effects, our understanding of how it affects the electrical conduction system of the heart remains incomplete. The present study will characterise both the relationship between generalised obesity and heart rhythm disorders, and also the relevance of differing fat deposits - such as the fat layer around the heart _ on the electrical conduction system of the heart.
