Summary
Diagnosis of Limb-girdle muscular dystrophy (LMGD) is complex with only about 50% success rate. New technology, Next Generation Sequencing (NGS) can identify a diagnosis substantially quicker and cheaper than standard methods, however is only available in research. The results of this study will form a core resource that can be augmented in the future to identify rarer forms of LGMD, so that all patients can be provided with a genetic diagnosis.
Nemaline myopathy is a neuromuscular condition characterised by muscle weakness, low muscle tone and the finding of nemaline bodies or rods on muscle biopsy. This study encompasses a natural history study of nemaline myopathy, genetic diagnosis and gene discovery using new methods of genetic testing, characterisation of a new disease gene for this condition, and reviewing patient experience with tyrosine, a medication commonly used in patients with nemaline myopathy.

Limb loss after tumour, trauma, disease or degeneration is a major cause of disability. Use of a patient’s own nerve signals may offer an intuitive method for controlling a robotic limb to regain independence. Delicate nerves are damaged by the electrodes required for recording nerve signals, but muscles tolerate electrodes well. This project aims to create an artificial muscle construct as a bridge between nerve signals and recording electrodes to enable patient control of robotic limbs.

Amyotrophic Lateral Sclerosis (ALS) is a rapidly progressive, uniformly fatal, neurodegenerative disorder with peak age of onset for the common sporadic variant of the disease being in the middle productive period of 50-60 years. The current research project aims at clarifying the site of onset of disease within the motor system, specifically, the primacy of the corticomotorneuron in pathogenesis. Difficulty taking deep breaths or coughing are two of the breathing complications people with a neuromuscular disease can face. Lung volume recruitment, also known as breath-stacking, is a simple and inexpensive therapy that may help. This research will look at the short and medium-term effects of breath-stacking exercises on the breathing system. If lung volume, chest stiffness and cough effectiveness improve then symptoms, quality of life and potentially survival are likely to be better.

Home mechanical ventilation (HMV) is a treatment that assists someone to breathe using a machine. People may need HMV when their breathing has failed due to conditions such as chronic obstructive pulmonary disease (COPD), neuromuscular disease (NMD) or severe obesity. This project will examine the quality of life of patients receiving HMV in Australia and Canada. It will also determine if we can improve the treatment by using a sleep study to monitor breathing when patients first start HMV.
