Summary
We have a sugar (mannan) that can be used to increase immune responses. We have found that mannan decreases the dose of inactivated virus needed for intranasal immunization. We will investigate if dose sparing is seen when given intramuscularly. This method will be first tried with the human flu virus and if successful will be tried with the bird flu virus. If the preparation can protect mice and ferrets from human or bird flu infection it could develop into a human vaccine against bird flu.

Summary
Periodontitis is a chronic inflammatory disease associated with specific bacteria leading to the destruction of the toothÍs supporting tissues and ultimately tooth loss and is a major public health problem in all societies. Our research is focused on designing a vaccine that targets the major bacteria associated with periodontitis. We aim to produce a triple antigen vaccine and investigate the applicability of a vaccine administered under the tongue as an alternative to needle injections.
Hendra virus (HeV) cause a disease transmitted from bats to horses which in turn infect humans and other horses. There are no drugs or vaccines for HeV. Since humans are infected by inhalation, a vaccine that can generate antibody in the lung and protect from infection will be ideal. We have found that a natural sugar called mannan used with virus proteins and administered via the nostrils to generate such responses. Summary
Despite many candidate vaccines entering clinical development for protection against HIV, none has yet been successful. This proposal centres on late preclinical development for a novel mucosal vaccine strategy for HIV, which combines a preclinically-proven approach to generating strong T cell immune responses, with an existing approach to generating broadly neutralising antibody responses to HIV. Proof of synergy between these approaches will lead directly to clinical development.

Summary
Antibody found in cervicovaginal secretions is predominantly of the IgG subclass. In the acidic environment of the female reproductive tract IgG antibodies against surface antigens on Chlamydia may actually enhance rather than prevent infection, due to uptake of IgG-coated bacteria by the neonatal Fc receptor (FcRn), which is found on the surface of epithelial cells in the reproductive tract. The proposed studies will determine the effects of chlamydial antibodies on the course of infection.

A specialised set of T lymphocytes called Mucosal Associated Invariant T (MAIT) cells protect us from bacteria and yeast at mucosal sites where the body's immune defences are most easily breached, e.g. gut, oral cavity, airways & reproductive tract. This study investigates the role of MAIT cells in health and in diseases like inflammatory bowel disease, peptic ulceration, periodontitis and tuberculosis. Controlling MAIT cells could help in treating these conditions.

Many Australians seek complementary and alternate medicine for the treatment of allergic diseases such as allergic rhinitis. There is evidence that acupuncture treatment is clinically beneficial for the treatment of allergic rhinitis. This project will produce new information that will provide a scientific basis of how acupuncture influences the immune response and modulates inflammation. Broader application to other inflammatory diseases and therapeutic interventions is possible.

Vaccination during pregnancy has health benefits for mothers and their infants; however, it is a relatively new area of research, and the immediate and long-term consequences for children are currently not well understood. As part of this fellowship, I plan to conduct research into the long-term health impacts of vaccination during pregnancy. This fellowship will build my career as a perinatal epidemiologist and establish expertise in Australia related to vaccines given during pregnancy.

Pneumonia is one of the commonest causes of childhood death worldwide. PCV is a vaccine that prevents pneumonia but it is costly; and causes an increase in disease from strains which are not in the vaccine. Our CRE will address 2 outstanding issues: when to switch from a 3 to 2 dose PCV schedule to make it more affordable; and create new understanding of the non-vaccine strains’ impact on disease in low- and middle-income countries in the Asia-Pacific region.
