Summary
HDAC inhibitors are anti-cancer drugs that kill rapidly growing cells (like cancer cells). These drugs also have anti-inflammatory properties and so may be beneficial in chronic inflammatory diseases such as as Rheumatoid Arthritis. However, it is unknown how they reduce inflammation. In this project we aim to understand how HDAC inhibitors act as anti-inflammatory agents and to design new HDAC inhibitors with reduced side effects for the treatment of inflammatory diseases.

Secretory phospholipase A2 (sPLA2) is an important mediator of inflammation and is clinically associated with the onset and severity of several immune-mediated diseases including arthritis, asthma, atherosclerosis, psoriasis and recently prostate cancer. These are complex diseases which are poorly understood. We have shown that sPLA2 can by itself and in combination with inflammatory cytokines modulate signalling pathways in cells derived from the joints of patients with arthritis to upregulate ....Secretory phospholipase A2 (sPLA2) is an important mediator of inflammation and is clinically associated with the onset and severity of several immune-mediated diseases including arthritis, asthma, atherosclerosis, psoriasis and recently prostate cancer. How this happens is completely unknown. We plan to work out how this enzyme does this. We have also developed small cyclic peptide inhibitors of sPLA2 which potently block the function of the enzyme in these cells. We plan to determine how this happens and if these inhibitors are effective at blocking inflammation and arthritis. This concern is only partly reduced by our potential to develop new antimicrobial agents and vaccines. This concern is only partly reduced by our potential to develop new antimicrobial agents and vaccines. If we were able to use immunomodulators in a relatively safe and appropriate manner to target and enhance the antimicrobial power of specific components of the immune system then this could be exploited in the treatment of infection. While body proteins formed (cytokines) which modify the behaviour of the immune system are being used as pharmaceuticals, their toxic side effects are problematic to the patient. Our project focusses on one of the cytokines, tumor necrosis factor (TNF), which increases the antimicrobial activity of phagocytic cells but in addition can have quite devastating effects on other tissues in the body. This is because when TNF binds to its receptor on cells and tissues it elicits a multitude of signals inside the cell which can also precipitate illness. The purpose of our investigations is to identify which signals are responsible for increasing resistance against infection and which are not. With this information we will then see if it is feasible to selectively stimulate this signal from outside the cell since this has a better chance of succeeding as a pharmaceutical. This task is likely to be achievable since our research team has made some unique observations about TNF signalling characteristics and we have developed a peptide TNF mimetic which shows only the characteristics of increasing antimicrobial activity.Read moreRead less

Once induced there is a progressive decline in health, which continues even after stopping smoking. There are no treatments that halt this decline. Recently small genes have been discovered that control inflammation. We may be able to control these small genes and stop the induction and progression of emphysema. Immunosuppressive drugs-anti-inflammatory agents have played important roles in treatment of these diseases as well as in helping to prevent rejection of transplanted tissues-organs. There is an ongoing search by the medical community for better immunosuppressive drugs and agents for use for these purposes. Based on studies conducted in the last few years, we have partially identified structural ....Autoimmune diseases are responsible for a high incidence of morbidity and mortality in our community. There is an ongoing search by the medical community for better immunosuppressive drugs and agents for use for these purposes. Based on studies conducted in the last few years, we have partially identified structural elements on polyunsatrated fatty acids (PUFA) which are responsible for specific biological functions. Using this information, we have synthesized a large panel of previously unavailable PUFA which we can use to target T lymphocytes and drive the biological activity of these compounds, preferentially towards immunosuppression. By conducting the research outlined in this project, we are likely to either identify a lead immunosuppressive compound or generate new information for further synthesis of PUFA-based compounds for further examination as potential immunosuppressive agents.Read moreRead less

This project will ascertain whether a naturally occurring compound, Resolvin E1 with potent anti-inflammatory properties, can effectively halt the progression of experimental kidney disease. We will also test whether Resolvin E1 can exert other potential benefits in suppressing progressive fibrosis of the kidney. The outcome of this study will allow us to evaluate the therapeutical potential of Resolvin E1 for the treatment of acute and chronic kidney diseases.

A concerted effort is being made by experts in the field of omega 3 fats (fish oils) to make specific recommendation on their use to improve human health . We have reasoned that the characterisation of a major oxidation product of these oils could be a key to understanding how these fats benefit us. The project is likely to influence the formulation of fish oils to enhance their health promoting properties.

In a small, but significant proportion of asthmatics (5-10%) symptoms persist despite the use of the best combinations of anti-asthma drugs. One of the reasons that acute episodes of asthma occur is that the airway tubes slowly change in structure. These changes involve an increase in the amount of collagen (part of the cement between cells) making the airway stiffer. In this project, we are exploring the impact of the stiffening of the airway on the way that different cells within the airway wall respond to drugs used to treat asthma. Our initial findings suggest that when the airway wall becomes stiffer with more collagen, there is a diminished benefit from the anti-asthma drugs. This new study is designed to identify the molecular mechanisms for the poor response to the anti-asthma drugs. An unfortunate consequence of immune function is that occasionally rogue immune cells are produced that attack the host and lead to the development of so-called autoimmune diseases such as arthritis. Normally a white blood cell called a regulatory T cell suppresses these self reactive immune cells. We have identified factors that govern genetic programs in regulatory T cells. Understanding how these factors work should permit the development of new strategies to combat autoimmune diseases.
