    Advancement Of A Personalised Approach To Minimising Infective Complications In Cancer Care

    Summary
    Managing infections in patients with cancer have become more difficult and unpredictable because of new generation cancer therapies. Measuring the response of the immune system (immune profiling) will allow us to predict which patients will develop infection so that action such as vaccination can be taken to reduce their risk. This program will refine immune profiling to personalise infection care for cancer patients and to introduce it into hospital practice.

    Food allergies have emerged as a major public health concern affecting 1 in 10 Australian infants. Hospitals waiting times are in excess of 12-months for specialist services. Recent changes in the environment are driving up rates of food allergy but the mechanisms are unclear. Epigenetics is the science of how the environment influences gene behaviour. This fellowship will address the important and urgent question of how modern environments are changing our genes, leading to food allergy.

    The immune system is comprised of many different cell types, each with a specialised function. Many are short-lived and must be continually replenished throughout life. Abnormalities in this process underlie many human diseases, including immunodeficiency, autoimmunity and cancer. My laboratory seeks to understand the molecular pathways that control development of immune cells and to identify the defects that lead to disease.

    Multistage Vaccines For The Prevention Of Tuberculosis

    Almost two million people die from tuberculosis (TB) each year. The current vaccine, BCG, is ineffective at controlling TB and the type of immune response needed to protect against the disease is poorly understood. We have discovered new antigens of the TB bacterium, and we will combine them with novel delivery strategies to develop new TB vaccines. We will also determine the type of immune response needed to protect against TB, which will aid progression of vaccines into clinical trials.

    The proposed study will involve the Barwon Infant Study (BIS) cohort of 1074 infants to provide the first systematic investigation of the hypotheses that the epidemic of allergic disease and asthma in many parts of the world relates to the paucity of fermentable fibre in the modern diet, and that the protective effect of fermentable fibre is mediated by changes in the organisms that colonise the gut and the metabolites that they produce.

Novel GPCRs expressed selectively on immune cells display a potential target for novel therapies for inflammatory diseases such as Asthma and Rheumatoid arthritis. Outcomes of this project will be further understanding of immune cell development and inflammatory disease development.

    The thymus is an organ dedicated to the production of crucial immune cells, called T lymphocytes. Cancer treatments, such as radiation or chemotherapy, destroy thymic function and impair immune recovery in patients. We aim to uncover molecular processes that govern the life and death decisions of cells in the thymus. Our goal is to then use this information to develop treatments to protect this critical organ from damage and improve immune recovery following radiation or chemotherapy.

    The immune system protects humans against recurrent infections with a wide range of pathogens. Formation of antibodies is a crucial element of the immune response. Defects in the production of antibodies can lead to recurrent and often life-threatening infections. This project seeks to understand a genetic defect in mice resulting in an almost complete absence of antibody producing cells, thereby causing a disease that is similar to some forms of human immunodeficiency.

    The thymus is the organ responsible for producing T cells, a key cell type in the body’s immune system. Certain cancer treatments damage the thymus, compromising the immune system and leaving patients susceptible to opportunistic infections. This proposal will develop clinically applicable strategies for generating functional human thymic mini-organs that could eventually help restore the immune system of people receiving treatment for cancer.

    This will be the world’s first birth cohort study to use substantial prospective data to investigate how biological, psychosocial, and environmental markers from birth will predict asthma and lung function in the third decade of life.
