Summary
Whilst ambulatory blood pressure monitoring data has been shown to be a good predictor of cardiovascular events, there remains controversy as to its utility in clinical practice. This project will use data from existing population and clinical cohort studies to examine the role of ambulatory blood pressure in risk assessment and hypertension management in Australia and around the globe. The findings are likely to have a major impact on clinical guidelines for hypertension management.

Summary
There has been a steady increase since 2002, in the age-standardised incidence of CRC in males under 45 years in Australia, contrasting with the stabilisation in incidence of CRC in males of age 45 years and over. Persons under 50 years are not routinely screened unless they have a significant family history of CRC. Young-onset rectal cancer is associated with late presentations and with a higher mortality. This proposal will address the possible risk factors for young-onset CRC.

Attention deficit hyperactivity disorder is a prevalent behavioural disorder affecting 7.4% of Australian children and adolescents. It has a strong genetic component with high heritability estimates (75–90%) comparable to other serious mental illness such as autism and schizophrenia. Identification and functional characterization of the genetic causes of this disorder will enhance our knowledge of its neurobiology and revolutionise the drug treatment of the disorder.

Summary
CCR5 antagonists are a new class of anti-HIV drug, and maraviroc (MVC) is the only CCR5 antagonists that is licensed for use as a HIV treatment. Like all HIV treatments, drug resistance to MVC can develop in patients. This study will determine the mechanism of how HIV becomes resistant to MVC, which will permit the development of improved, second generation CCR5 antagonists, and will reveal ways to determine which patients are more likely to develop MVC resistance.

This grant will address a major question in HIV cure research - the role of the liver as an HIV reservoir and the impact of HIV persistence in HIV-infected patients on suppressive antiretroviral therapy (ART) on liver disease, in the setting of HIV-HBV co-infection. We will trial a novel intervention to reduce HIV infection of the liver that could potentially reduce chronic liver disease in this setting.

There remains an urgent need for new HIV prevention strategies. New HIV drugs that block the virus by distinct ways are needed to prevent transmission of drug resistant HIV. This study seeks to identify very small molecules called “fragments” that bind to previously undiscovered pockets on the HIV reverse transcriptase to stop its function, and that can be used as building blocks to design more potent HIV drugs to be used solely for HIV prevention.
