Summary
This project will investigate whether or not young people at high risk of developing psychosis show irregularities in the connections between different parts of their brain. It will also investigate whether information about a high-risk individual’s brain connectivity can be used to predict a) whether that person will make the transition to psychosis, and b) their clinical prognosis if they do become psychotic.

Summary
Each year over 10,000 new cases of breast cancer are diagnosed in Australia and over 2500 women die. Tamoxifen, a non-toxic tablet used to control the growth of breast cancer, has safely been taken by more then a million women long term . This project measures tamoxifen's role in preventing breast cancer in high risk women compared to placebo, in a randomised double blind clinical trial. The trial has the potential to benefit many millions of women worldwide. This project measures tamoxifen's role in preventing breast cancer in high risk women compared to placebo, in a randomised double blind clinical trial. The trial has the potential to benefit many millions of women worldwide. The study, IBIS I (International Breast Cancer Intervention Study), is conducted in Australia by the Australian New Zealand Breast Cancer Trials Group (ANZ BCTG), and internationally by Cancer Research UK (CRUK). On the trial women have regular, annual mammography, 6 monthly clinical checks and take a tablet each day called TAMPLAC, which is either tamoxifen or placebo. The accrual phase has been funded by the NHMRC and the target number of women on the trial was reached ahead of schedule - 7154 internationally including 2674 from Australia. By April 2006 all women will have completed the treatment phase. Funding is now being sought, in Australia, for continued follow-up and investigations of additional risk factors (breast density and types of tumors which occur-have occurred on IBIS I). The ANZ BCTG and all the women involved in IBIS I have demonstrated remarkable commitment. It is now vitally important that the essential follow-up is completed to facilitate analysis of the data, including the post-treatment phase, as well as related risk factors and types of tumors which develop. The only other large randomised trial testing tamoxifen (NSABP P-1) ceased and was unblinded in 1998, with an average follow-up of less than 4 years, the trial had to re-commence follow-up to determine longer term tamoxifen effects. The IBIS 1 study remains the only large, blinded trial in the world, and completion of it's follow-up and analysis is of very high international importance.Read moreRead less

Polyunsaturated fatty acids (PUFAs) play an important role in many physiological processes in all organisms. Myelination is the process by which a fatty layer, called myelin, accumulates around nerve cells enabling nerve cells to transmit information faster. PUFAs are essential for myelination, and there is evidence documenting decreased PUFA concentrations and brain white matter (myelin) pathology in people with schizophrenia. The mechanisms underlying these abnormalities are not understood.

To understand who is at risk for depression, we need to understand how both the environment and biological factors promote risk at specific times in the life cycle. This research project will examine such risk factors in different phases of life (from in utero to early adulthood) using a developmental framework. The results of this research will provide a clearer basis for the design of prevention strategies that target individuals, their families and/or broader environmental factors.

Age-related macular degeneration (AMD) is the leading cause of vision loss in older individuals. AMD eyes with reticular pseudo drusen (RPD) are now recognised as at high-risk of faster progression to vision loss. Identifying the underlying mechanisms driving RPD is crucial for to identify specifically targeted therapeutic options. Validating our subthreshold laser trial, and our early endpoint will offer the first proven intervention to slow AMD progression to vision loss.

Summary
People who carry mutations in the HFE gene are pre-disposed to body iron overload but not all of them developed subsequent disease. According to the investigators of the “HealthIron” study in Melbourne, only 28% of men and 1% of women with faulty HFE genes go on to develop disease. This study has recruited more than 1,000 people from the community, and will determine which environmental and genetic risk factors stop people with iron overload from getting symptoms of disease.
