    Many heart diseases are associated with impairment of energetics of the heart. Improving the heart's energetics can lead to improved survival and long-term outcomes. Perhexiline is a heart medication that works by improving the way the heart uses energy. Although effective, it is associated with long-term toxicities. Better understanding of this medication may lead to less adverse effects and also provide a basis for further investigation of drug development in the future.

    Heme oxygenase-1 is a protein that recycles our body's iron which itself is essential for life because it is needed for the sensing, transport and use of oxygen we breath. The current project investigates a novel interplay between heme oxygenase-1 and our body’s major response to tissue oxygen becoming limited. By doing so, our project may provide the basis for novel treatments of conditions where arteries are obstructed, such as those that lead to heart attack and amputation.

    Duchenne Muscular Dystrophy is a fatal muscle wasting disorder. We have previously characterised how the heart fails in a mouse model of muscular dystrophy. We now have preliminary data demonstrating that treatment of mice with morpholino oligomers can rescue cardiac function. This project will fully characterise the effect of the treatment on heart function and optimise therapy regimes with the view to utilising the optimised protocol as a guideline in treating cardiomyopathy in Duchenne Muscul ....     Dysregulation of cholesterol metabolism plays a key role in a number of diseases – from diabetes and atherosclerosis to neurological and skin disorders. Mechanisms of regulation of cholesterol metabolism are poorly understood. We have recently discovered a new pathway regulating cholesterol metabolism and in this study we will investigate molecular, cellular and physiological mechanisms of this pathway and will identify the possibilities to target it for therapeutic intervention.

    The process that turns cut fruit brown when it is exposed to air is thought to cause disease of our blood vessels and heart as we age. Despite what we first thought, 'blocking' this oxidation process with antioxidant supplements does not lower heart disease. This is because oxidation not only causes harm but also is useful and essential for normal body function. Our research program aims to show which oxidative processes are needed for blood vessel health or cause vessel disease.

    Molecular Mechanisms Of Inherited Hypocholesterolaemias: Impact Of APOB And MTTP Mutations On Lipoprotein Assembly And Secretion

    Inherited low cholesterol levels can be caused by mutations in either of two genes: APOB and MTTP. These genes encode proteins that are critical for the assembly of fat particles in the body. We plan to use cell lines to study how single amino acid changes out of the 4500 in ApoB and the 900 in the MTTP protein affect protein production, binding with other proteins, and fat particle assembly.
