    PRESENILINS IN ALZHEIMER'S DISEASE

    Summary
    The presenilin proteins are key components in the development of Alzheimer's disease. Mutations in these proteins can cause early Alzheimer's disease. We will continue our study of the biochemistry and cell biology of these proteins using tools and experimental models that we have already developed. This will provide important information on the mechanism of the disease process and give new leads in the treatment of the disease. The new technologies of genetic screening for presenilin mutations .... The presenilin proteins are key components in the development of Alzheimer's disease.     Progressive Aphasia And Amyloid Deposition: A Multidisciplinary Approach To Improving Dementia Diagnosis

    We aim to understand the role of amyloid deposition in the genesis of symptoms in Alzheimer's disease. Patients presenting with progressive deterioration of language (aphasia) some of whom are known to have Alzheimer's disease will be studied using brain imaging techniques, including a PET method (PiB) that labels amyloid in vivo, and to compare imaging changes with a range of language assessments. This will improve the early diagnosis and management of patients with progressive aphasia.

    Music Therapy Interventions For Dementia: Cluster Randomised Control Trial

    Summary
    Music therapy interventions are increasingly used in aged care facilities across Australia, however Australian-based research that tests its effectiveness is scarce. In this study, we track the impact of group music therapy and group singing on levels of depression, cognitive function, quality of life, and other symptoms of dementia. By providing music therapy programs across sites in Victoria, NSW and Queensland, we will also evaluate the cost effectiveness of the intervention.

    Functional Genomic Analysis Of The Role Of P53 In Early Embryo Death After Assisted Reproductive Technologies (ART).

    Assisted reproductive technologies (ART, such as IVF and related techniques) are successful treatments for most forms of infertility. ART are expensive therapies and much of this cost is related to the relative inefficiency of the technology. Much of this is due to the high mortality of the resulting embryos. Typically, 45-80% of embryos produced by ART do not survive the first week. Consequently the chance of any individual embryo resulting in a successful birth is not high. There has been only .... Assisted reproductive technologies (ART, such as IVF and related techniques) are successful treatments for most forms of infertility. ART are expensive therapies and much of this cost is related to the relative inefficiency of the technology. Much of this is due to the high mortality of the resulting embryos. Typically, 45-80% of embryos produced by ART do not survive the first week. Consequently the chance of any individual embryo resulting in a successful birth is not high. There has been only modest increments in embryo survival in recent years. The low cahnce of individual embryos resulting in a baby means that: (1) generally several treatment cycles are required; (2) superovulation is used to maximise the number of embryos produced giving an accumulation of unwanted cryopreserved embryos; (3) more than one embryo is generally transferred resulting in a significant incidence of multiple pregancies. The high mortality of the early embryo seems to be a general feature of IVF but its causes and effectors are not known. It has recently been established that it largely occurs due to a form of cell 'suicide' known as apoptosis. This form of cell death has important normal functions: its activation allows for cells that are no longer required to be removed, allowing the remodelling of tissues and it also serves to remove cells that are irreversibly damaged. p53 is a protein that has the ability to 'sense' cell stress and damage and to direct the cell to undergo apoptosis if the stress is severe. This project will examine if ART cause increased expression of p53 and whether this elevation of p53 causes embryonic cell death. We will examine the factirs that control p53 expression in the embryo. using mice with mutations that stop the function of p53 and several of its regulatory proteins. Experiments will determine the susceptibility of embryos possessing these mutations and will therefore allow us to define the proteins causing apoptosis after ART.

Results are expected to inform a critical knowledge gap and assist in the development of targeted violence preventive responses which reduce harm and make communities safer.

    Frontotemporal dementia accounts for 12-20% of all dementia cases. It is as common as Alzheimer�s disease in the < 65 year olds but understanding of this disease remains limited. Marked eating disturbance (overeating, snatching food) is a common clinical manifestation. This project will characterise this deficit and define its biological causes using blood tests and magnetic resonance imaging. Results will help in designing therapeutic interventions targeting this disabling feature.

    My research program aims to discover genes that are mutated in dementia, by identifying gene variants present in patients and absent in healthy people, and examining how these variants affect the function of cells. Identifying new dementia genes will reveal the biological processes that lead to brain cell death. Knowledge of these processes is crucial for the development of new treatments for the many people affected worldwide with dementia.
