    Determining The Genetic Basis Of Skeletal Dysplasias Using Next Generation Sequencing

    Summary
    Osteoporosis is a common condition in Australia, yet treatment options are still limited. Study of rare genetic bone conditions known collectively as skeletal dysplasias have already led to the development of two new osteoporosis drug treatments. My project aims to identify the causative gene for several skeletal dysplasias, and to determine how these genes are involved in the development and maintenance of bone. This knowledge may then translate into new osteoporosis therapies.

    The experimental and computational methods (finite element method) are used to predict biomechanical behaviors of osteocytes under normal physiological loading, overloading or under-loading/disuse. This quantitative research will not only help to elucidate the mechanisms of mechanotransduction in osteocytes, it will provide important information that is also relevant to mechanobiology in general.

    In a partnership between Peter MacCallum Cancer Centre, St Vincent's Hospital, and The University of Melbourne, we are investigating the role of PTHrP, a peptide integral to the growth and spread of Cancer. Initially thought to facilitate cancer spread, recent studies suggest it may actually be protective. In a new approach, we will analyse new DNA databases and patient data from around the world. We hope to extend our understanding of PTHrP, and perhaps find novel drug and therapeutic targets.

    Osteonecrosis of the jaw (ONJ) is a debilitating bone condition involving damage and suboptimal healing of bone involving the jaw. This has been associated with bisphosphonate therapy, which is commonly used for the treatment of both cancer and osteoporosis. My research aims to investigate the role of recombinant parathyroid hormone in the stimulation of bone formation and healing and, thus, its potential to reverse ONJ.

    Calcification of the arteries is a ubiquitous finding in patients with end stage renal failure. It is often mirrored by disturbed bone metabolism. Increasing evidence suggests a close relationship between bone and vascular health. This study aims to determine the role of calciprotein particles as a mediator of calcification in bones and arteries. Translating these findings to renal patients will ultimately contribute to providing a scientific basis for developing treatment for these patients.

    The Effect Of Androgen Replacement Therapy On Bone And Muscle Health In Men With Chronic Kidney Disease

    Summary
    Low testosterone (T) levels are common in men with poor kidney function. Low T is known to cause reduced energy, decreased strength and low libido. Normal T is also needed for healthy bones and muscles. Men with kidney disease are already at risk of fractures, poor strength and quality of life. However, there are few studies that look at replacing T to men with kidney failure. We will investigate how low T affects bone and muscle and assess how giving T can benefit bone, muscle and function.

    This project investigates using animal derived and artificial bone in regions of the mouth that are deficient and not suitable for replacing missing teeth using dental implants. Traditional techniques of bone replacement require the use of a patient's own bone. These procedures are associated with post-operative morbidity and this bone is present in limited quantity. The use of these bone replacements may overcome these challenges and eliminate the need for using patient's bone.

    I am a practicing specialist Periodontist focused on growing bone around dental implants used to replace teeth. I will achieve this by comparing available materials to new materials and techniques. Dental implants are a very common treatment modality which results in improved outcomes compared to traditional methods of tooth replacement. This research will improve the outcome of dental implant placement and allow greater utilisation of this treatment modality.

    The Effects Of Zoledronic Acid On Bone Architecture In Premenopausal Women With Breast Cancer Receiving Adjuvant Combined Ovarian Suppression And Aromatase Inhibitor Therapy: A Randomised Controlled Trial.

    Summary
    In premenopausal women, a new treatment method that reduces oestrogen levels to almost zero significantly reduces the risk of breast cancer recurrence. However, this is likely to cause substantial bone loss leading to fractures. Using a new imaging technique (HR-pQCT), the effects of profound oestrogen deprivation on bone structure in premenopausal women will be studied. The ability of zoledronic acid, a drug that reduces bone loss, to prevent these adverse bone effects will also be examined.

    Summary
    The bone marrow (bm) is the birthplace of all blood cells that fight infection in the body. Dendritic cells (DC), essential for starting immune responses, are found in the bm but the exact types and their functions are unknown. I plan to investigate the DC types that reside in the bm and explore their role in inducing immune responses and in influencing the development of cells from the bm with potential attributable benefits for stem cell transplant and vaccine design.
