Heart attack and inflammatory diseases such as rheumatoid arthritis und multiple sclerosis either kill or severely disable people. We use the presence of platelets early on in these diseases to develop methods for early diagnosis as well as potential drugs for site-directed therapy. We have developed new biotechnological tools to perform novel high sensitivity imaging in Positron Emission Tomography (PET) and laser light imaging as well as a localised anti-inflammatory therapy.

Asthma is a chronic disorder where airways are remodelled, resulting in poor lung function. Airway remodelling is a consequence of long-term inflammation. As current treatments halt some, but not all, aspects of airway remodelling, new therapeutic approaches are urgently required. In this grant, our aim is to devise novel strategies to boost the function of a critical anti-inflammatory protein - TTP - to reduce inflammation in asthma.

Summary
Heart attacks and strokes are most often caused by rupture of unstable atherosclerotic plaques resulting in clotting and vessel occlusion and ultimately irreversible damage of the heart or brain. We have generated a mouse model that reproduces these plaques as seen in humans. Based on this model and human unstable plaques 1) we have discovered/developed imaging technologies that can identify these dangerous plaques and 2) we will develop drugs that prevent plaque rupture.

Summary
We are conducting four large clinical trials testing anti-inflammatory treatments like ?-3 PUFAs and aspirin in young people who are at high-risk for psychosis or have depression. This proposal adds an important component to this research by investigating inflammatory and oxidative stress markers. We aim to determine if the investigated biomarkers predict the course of illness and response to treatments. The findings will facilitate early intervention and targeted treatment.
During injury or infection, our body’s immune system protects us by launching inflammation. But uncontrolled inflammation drives common diseases such as cancer, diabetes and Alzheimer’s. This project will reveal how the body deactivates inflammasomes - protein complexes at the heart of inflammation and disease – so we can design better strategies for treating patients with inflammation-driven disease.

Inflammation is an underling cause or aggravating factor of many diseases, such as heart attack and atherosclerosis, but there is a lack of effective anti-inflammatory drugs that do not cause side effects. Using advanced biotechnology, we will use viruses and nanoparticles to selectively target gene therapeutics to areas of inflammation, resulting in high efficacy with low side effects. We will test these broadly usable therapeutics in mouse models of heart attack and atherosclerosis.

Cytokines are messenger proteins that function as master regulators of biological processes; thus they play central roles in many diseases. The rare cytokines that block inflammation do so by dampening the immune system’s potentially destructive force, making them attractive targets for drug development. We showed that interleukin 37 is a powerful anti-inflammatory cytokine, and will now evaluate its mechanisms of action and its efficacy against several severe diseases, including cancer.

Despite the widespread and increasing use of opioid analgesics, there is a complete lack of evidence on their efficacy in acute spinal pain. Concerns are also being raised because of the risks of potentially serious adverse events associated with opioid analgesics. In this world-first study, we will establish whether using opioid analgesics can effective reduce pain in people with acute spinal pain and provide rigorous evidence to inform the safe and appropriate use of this medicine.
