CD4 T cell programming by neonatal and early-life infection. T lymphocytes (T cells) are white blood cells that play a critical role in protecting the body from infection. Before T cells can function they need to be programmed so that they can specifically respond to an infectious agent (a type of bacteria or virus). Inappropriate programming can lead to disease. T lymphocytes (T cells) are white blood cells that play a critical role in protecting the body from infection. Before T cells can function they need to be programmed so that they can specifically respond to an infectious agent (a type of bacteria or virus). Inappropriate programming can lead to disease. Whether T cells respond to an infectious agent or foreign substance in a protective or destructive manner may critically depend on the age that an individual first encounters the infection. Our project will identify critical periods in life that direct T cell programming to subsequent protective or destructive responses, providing new insights into the developing immune system that may be exploited to treat disease or develop vaccines.Read moreRead less

This proposal will address a gap in our imaging capabilities, allowing us to visualise the movement of immune cells and infectious agents such as bacteria and viruses within living tissues. This will immensely improve our capacity to understand interactions between the immune system, invading organisms and the rest of our body. The intravital imaging system will provide novel insights into how the immune system works, which will benefit the design of vaccines, the treatment of cancer, and our understanding of allergy. This state-of-the-art facility will also provide vital training in an emerging technology that will have application in many areas of biology.

Foreign DNA is a danger signal for mammalian cells. This project investigates how cells normally respond to foreign DNA, and is relevant to understanding how the body fights infections, particularly by viruses. The results will help us to design more effective treatments for infectious disease. Studying responses to DNA will also promote the design of new treatments for the autoimmune disease lupus, and help improve technologies or treatments where DNA is introduced into cells or tissues. The results will help us to design more effective treatments for infectious disease. Studying responses to DNA will also promote the design of new treatments for the autoimmune disease lupus, and help improve technologies or treatments where DNA is introduced into cells or tissues. This includes gene therapy, new strategies for vaccination, and the production of proteins as drugs by biotechnology. The project will promote National Research Priorities in the areas of preventative healthcare, ageing well ageing productively, breakthrough science and new technologies.Read moreRead less

The survival of our species reflects a delicate balance between infection and immunity. Whether an individual mounts an effective immune response or succumbs to microbial infection is critically dependent on host proteins interacting effectively with microbial antigens, versus microbes developing sophisticated strategies of invasion and immune evasion. This proposal will provide fundamental advancement of knowledge in the areas of infection and immunity. The information gleaned from this research will lead to the rational development of therapeutics. Consequently, the research will potentially have an enormous global impact in the area of biomedical health.Read moreRead less

Multiphoton microscopy of living animals as a tool for immunology and cell biology studies. The multiphoton microscope will enable us to watch the growth, migration and interactions of cells in a living animal in response to changes in the cells' environment will give us better understanding of how we work as living machines, and what can go wrong with that process to make us unwell.

Interleukin-4 (IL-4) is the only immune active product or cytokine that safely skews an aberrant immune response to a healing type of response. Fowlpox viruses (FPV) provide safe and effective human vaccines. Engineering FPV to make both a relevant antigen and a cytokine is proprietary technology. With our commercial partner and using experimental mice, we will test the concept that FPV making a model antigen and IL-4 initiate and/or maintain beneficial Type 2 responses. A successful outcome will guide clinical FPV-based vaccine development for the treatment of important human and veterinary diseases.Read moreRead less

In the developed world infection diseases are the number three killer behind heart disease and cancer, and huge financial effort is put into treatment and prevention. Despite this, results have often been disappointing. One cause of these poor outcomes is the lack of knowledge of how effective immune responses are generated. This project aims to better understand the processes that control the generation of protective lymphocytes. It will deliver information that may enable a more targeted approach to vaccine-development and treatments of infections. As defective differentiation can also be a cause of leukemia it may also lead to targets of cancer treatment.Read moreRead less
