Summary
Australia has experienced a prolonged epidemic of pertussis from 2008 to 2012 and is currently experiencing another epidemic. In this project, we aim to elucidate the evolutionary dynamics of the epidemics by genome sequencing and develop a practical high throughput culture independent method for epidemiological typing. The outcomes will be highly significant for surveillance of pertussis infections and designing strategies for control and prevention of pertussis.

The activity of individual sensory neurons in the brain is surprisingly variable and continuously changing. It is unclear how reliable perception of the world can be generated from the activity of “noisy” neurons, and it remains unclear why neuronal sensitivity should change in the first place! This project will give insights into how groups of sensory neurons collectively overcome their intrinsic variability to support reliable visual perception.

Summary
A possum in a tree is cute to see on a bushwalk, but scary to hear when we are trying to sleep. This illustrates that how we perceive a “target” is affected by “modulators” that are close in space or time to the target, or by the task at hand. Deficits in contextual modulation are apparent in many neurological conditions. This project will investigate the neural circuitry that mediates spatial, temporal and task-related contextual modulation.
In pilot studies, we have found that specific genes are modulated by long-term alcohol intake in a discrete part of the brain that drives habitual behaviours. We will now repeat these studies in post-mortem brain tissue from human alcoholics and age-matched controls. In addition, we will further characterise the functional consequences of long-term alcohol intake within this specific brain region and test new approaches to reduce the urge to consume alcohol.
A New Mouse Model That Determines The Effects Of A Unilateral Vestibular Prosthesis On Vestibular Plasticity.

Summary
Much like a cochlear implant restores auditory function, a vestibular prosthesis restores balance function. It is not clear whether the limited results from vestibular prostheses is due the device not stimulating one component (the otoliths) of the vestibular system essential for self-repair. We will test mutant mice that lack otoliths to determine the importance of stimulating the otoliths in restoring function. This work will shape the future direction of prosthesis development.

HIV diversity observed at the global level is mostly due to the selection pressure exerted on the virus by our adaptive immune response. This process is important in determining infection outcome. This proposal will use a unique single cell approach to understand this complex host-viral interaction to identify important correlates of immune efficacy against HIV that will aid in the rational design of potential vaccine candidates and addresses a fundamental concept in anti-viral immunity.

Summary
Cystic fibrosis (CF) is the most common autosomal recessive disorder in humans, affecting 1:2000 people. Mortality is often caused by Pseudomonas aeruginosa lung infections which have recently been shown to occur not only environmentally but also via person-person contact, usually during CF clinic visits. This project will elucidate the molecular traits responsible for these 'epidemic' P. aeruginosa infections, with the aim of finding novel therapeutics and infection control strategies.
