Following the success in decoding human genome, i.e. DNA sequence, a major task is to understand how the activity of genes with consequent changes in respective proteins. As proteins are an important component for cell structure and function, such changes in quantity and quality of proteins will play a pivotal role to affect disease development and progression.
HIV infection is a dynamic process, in which the host immune response tries to control viral growth and keep up with the rapid evolution of the virus. This project assembles an interdisciplinary team of mathematicians and biologists to use a modelling approach to understand the dynamics of viral infection, viral evolution, and immune control in the infected individual. The insights gained from this project will help in the development of new drug and vaccination strategies.

Summary
Current therapy for HIV is effective but must be taken for life. If therapy is stopped the virus comes back immediately from reservoirs not affected by current drugs. These fluctuating levels of virus are associated with increased illness and death. We are exploring a method of inducing prolonged viral latency using short double stranded RNA molecules. We propose to understand the mechanism of action of these possible therapeutics and to develop these constructs towards use in clinical trials.

It is estimated that one third of the worlds population is affected by mild to moderate zinc deficiency, and that this predisposes to a range of infectious diseases. The immunomodulatory effects of zinc have been appreciated for many years, and indeed zinc supplementation is used to treat severe diarrhoeal diseases. This project aims to understand the anti-infective mechanisms of zinc by focusing on macrophages, a key cell type involved in killing invading microorganisms.

Follicular lymphoma (FL) is divided into early and advanced stages. Early stage FL is frequently cured, but there is no way to identify who will be cured and who won't. By contrast advanced stage FL is incurable. Our unique access to well-annotated clinical trial and population based cohorts allows us to perform a detailed biological comparison of early and advanced FL, to gain a deeper understanding of the impediments to eradicating the disease, and to predict outcome to conventional therapy.

Specific gene regulatory proteins define functions of various subsets of neurons in the hypothalamus. We will determine how interactions between three such proteins activate the leptin-melanocortin pathway, a hypothalamic signalling circuit that controls appetite. Defects in these proteins are found in obese patients who suffer from excessive eating disorders. The project will improve understanding of the genetic determinants of obesity and provide key points for development of new therapies.
