Summary
Allergic diseases like asthma, eczema and hay-fever, prevent our children from getting a healthy start to life, and we don’t know how to prevent these conditions. Vitamin D levels may be critical in the development of childhood asthma and allergies, and they can be easily modified! Using a group of 620 children who we have followed for 20 years, we will identify the role of vitamin D levels in the development of allergic conditions, and factors that modify these relationships.

Summary
Our recent work has discovered that the Nedd4 gene is crucial for the growth and development of blood vessels and lymphatic vessels. Our data suggest that Nedd4 controls vessel growth by regulating the levels and signalling activity of the key vascular growth factor receptors VEGFR-2 and VEGFR-3. The goals of this proposal are to define precisely how Nedd4-1 regulates the activity of these receptors and how VEGFR signalling could be better targeted to treat vascular disorders.
The D-Health Trial: A Large-scale Population-based Trial Of Vitamin D Supplementation For Improving The Health Of Older Adults

Summary
The D-Health Trial is a randomised placebo-controlled trial of vitamin D supplementation for improving the health of older Australian adults. Over 21,000 people were recruited and randomised to monthly doses of vitamin D or placebo. This grant will enable completion of the planned 5 years of supplementation and long-term follow-up of mortality and cancer. The results of the trial will inform decisions about food fortification and/or supplementation with vitamin D.
There is a need for a greater understanding of the complex relationship between sun exposure and the production of Vitamin D. This study will expand on the recently-completed AusD study to examine how personal, behavioural, and environmental factors impact on intra-individual seasonality in Vitamin D production. The study findings will guide the development of specific, evidence-based public health recommendation that balance the risks and benefits of sun exposure.

Summary
Vitamin D most commonly from sunlight exposure is considered an important determinant of health, including the liability to falls and fractures; not only in the elderly but in adulthood in general. This study aims to distinguish if low vitamin D levels cause adverse health outcomes or relate to ill health, limited sunlight exposure and nutritional change. Only if low vitamin D level is causative of ill health does replacement make good public health sense.

There has been a dramatic and unexplained increase in food allergy over recent decades. The increase in food allergy may relate to a concordant increase in early vitamin D insufficiency, however this hypothesis has never been directly tested.
