Role Of Epigenomic Changes In Conferring Hyperglycemic Memory

    Funder
National Health and Medical Research Council
    Funding Amount
    $636,146.00
    Summary
The major burden of type I diabetes remains its vascular complications including diabetes-accelerated athersclerosis. Although the development and progression of these diabetic complications is strongly associated with mean levels of glucose, recent studies suggest that the deleterious effects of early exposure to high levels of glucose persist for years even after treatment has returned glucose levels towards normal.

Developing New Treatments For Brain AVMs

    Funder
National Health and Medical Research Council
    Funding Amount
    $695,269.00
    Summary
Congenital brain vascular malformations are a common cause of stroke and death in young patients. This project aims to develop a new treatment for these lesions that does not require surgery. We will use focussed radiation to change the cells lining the abnormal vessels so that they can be targeted with a new treatment that causes blockage of the vessels and prevents haemorrhage.

Role For Zinc And ZIP2 In The Action Of Nitric Oxide And In Vascular Protection Against Cigarette Smoke And Cardiovascular Disease

    Funder
National Health and Medical Research Council
    Funding Amount
    $685,941.00
    Summary
The NO/cGMP signalling pathway, which is central to cardiovascular physiology and protection against disease, is only fully effective when there are adequate levels of zinc in the vascular endothelium. This is especially important where zinc stores are depleted (elderly, smokers, diabetics and kidney disease). There is an urgent clinical need to implement strategies to monitor vascular Zn status. This application will explore the underlying science and translate these to the clinic.

Uncovering A Novel Genetic Interaction That Governs Blood Vessel Development In Health And Disease

    Funder
National Health and Medical Research Council
    Funding Amount
    $626,557.00
    Summary
Blood vessels are a vital component of the cardiovascular system. Abnormalities in the growth and development of blood vessels are associated with human disorders including cardio-vascular disorders, cancer and inflammatory diseases. The focus of this application is to determine the molecular events that direct the construction of the blood vascular tree, with the aim of identifying targets to which novel therapeutics for the treatment of blood vascular diseases could be generated.

Therapeutic Potential Of Inhibiting Eph/ephrin Signalling To Repair The Vascular Endothelium In Septic Shock

    Funder
National Health and Medical Research Council
    Funding Amount
    $664,734.00
    Summary
Septic shock is a life-threatening condition usually caused by bacterial infection in the bloodstream. More than 5000 people, including 500 children, die from sepsis each year in Australia. Worldwide, it is the most significant cause of death in children. Sepsis is associated with leakage of fluid and proteins through the cells lining the blood vessels. This project will develop and test a novel treatment for sepsis which focuses on reducing this leakage by blocking the Eph/ephrin proteins.

Developing And Testing Pro-thrombotic Conjugates For Brain AVMs

    Funder
National Health and Medical Research Council
    Funding Amount
    $520,256.00
    Summary
Rupture of brain blood vessel abnormalities causes stroke in children and young adults. In this project, we will test new methods for occluding abnormal vessels. We use radiation first to induce changes in the vessels, and then target those changes to occlude the vessels. Successful treatment requires combining targeting and clot forming agents. In this project we will test combinations of targeting and clotting agents. The most effective combination will be progressed towards clinical trials.

Does Manipulation Of Arterial Shear Stress Enhance Cerebrovascular Function And Cognition In The Aging Brain?

    Funder
National Health and Medical Research Council
    Funding Amount
    $707,914.00
    Summary
Globally, 9.2 million less cases of dementia could result from delaying it by as little as 12 months. We will assess the impact, on artery health and cognition, of a novel exercise intervention involving enhanced patterns of brain blood flow and arterial shear stress during exercise, stimuli which should optimise cerebrovascular health. Given that pharmacological approaches to prevention are ineffective, this project has the potential to address the future impact of this common and debilitating .... A New Target For Allergic Inflammation: The Sphingolipid Pathway

    Funder
National Health and Medical Research Council
    Funding Amount
    $588,617.00
    Summary
Collectively, allergic diseases contribute immensely to the burden of health care in Australia. Notably, allergic reactions are symptomatic responses to a normally innocuous environmental antigen. Allergic diseases include asthma, hay fever, food allergy, anaphylaxis, insect sting and drug allergy. This project aims to understand the underlying mechanisms associated with allergic reactions such that it may aid in the identification of novel targets for the development of new treatments.

Functional Characterisation Of A New Surface Adhesion Molecule On Human Vascular Progenitor Cells To Combat Cancer

    Funder
National Health and Medical Research Council
    Funding Amount
    $593,794.00
    Summary
Collectively, diseases of the blood vascular system contribute immensely to the burden of health care in Australia. Notably, abnormal blood vessel formation is a major cause or contributor to many diseases, such as cancer, cardiovascular disease, rheumatoid arthritis, ischemia injury and diabetes. This project aims to understand the underlying mechanisms associated with aberrant angiogenesis such that it may aid in the identification of novel targets for the development of therapeutics.

Role Of Resident Endothelial Progenitor Cells In Melanoma Vascularisation And Progression

    Funder
National Health and Medical Research Council
    Funding Amount
    $952,328.00
    Summary
Melanoma is one of the most frequent cancers in Australia. Its growth depends on the rpoper delivery of nutrients and oxygen through blood vessels. This requires the formation of new blood vessels as the tumour grows. In this project we intend to understand the origin of the blood vessels that form in tumours and identify the stem cells that support them. We will use proof of principle experiments to determine whether removal of these stem cells allows the regression of melanoma tumours.
