Summary
Diabetes is diagnosed using clinical assessment complemented by a few selected basic conventional laboratory tests. We plan to determine, in a representative community-based sample of Australian with diabetes, whether additional knowledge of genetic markers, diabetes-related antibody levels and tests of insulin secretory capacity adds to diagnosis of diabetes type, prediction of therapeutic progression over time, complications and death.

Summary
The insulin-like growth factor (IGF) system, required for normal development and adult life, is often altered in many diseases including cancer. Key regulators of the IGF system are the IGF binding protein (IGFBP) of which IGFBP-2 is the 2nd most abundant. IGFBP-2 may enhance or inhibit the IGFs, but the mechanisms are not clear. This proposal aims to dissect IGFBP-2 action with the ultimate goal to provide knowledge for the development of targeted therapeutic modulators of IGFBP-2 activity.

Summary
Obesity and impaired insulin secretion are significant contributors to Type 2 diabetes. In this project we demonstrate that a protein called RCAN1 contributes to both fat mass and insulin secretion and that this contribution is exacerbated in obesity and in Type 2 diabetes. We will identify how RCAN1 controls these major metabolic pathways with outcomes including the development of new therapeutics for obesity and Type 2 diabetes.
