Summary
Pituitary tumours encompass a number of pathologies. Their cause is not clearly established. Pituitary adenomas are one of the most frequent intracranial tumours. The genetics of sporadic tumours is unknown. Craniopharyngiomas are rare brain tumours arising in the pituitary stalk area that can have profound effects, presenting in childhood or later. To date there is limited knowledge on the cell signaling pathways causing these tumors, which can help to understand cancer in general.
Lung cancer is a leading cause of cancer death globally. Symptoms may not develop until disease is advanced, so it is often incurable at diagnosis. Scientific developments have greatly improved our ability to test for the changes in DNA structure and function responsible for this deadly disease and its progression. This study examines whole lung cancer genomes then uses these findings to develop safer methods for detection based on changes in DNA sequence.

Vitamin D3 levels appear to predict clinical status in multiple sclerosis. The reasons for this are unclear, but may be linked to the effect of the Vitamin D Receptor (VDR) on a subset of immune cells. This project aims to identify key genes which are regulated by this receptor, by using specific gene sequencing technologies combined with knowledge of the genes which confer risk of developing MS. This may help to identify the molecular pathways underlying MS and potential treatment strategies.

This study aims to develop a new antibody therapy for Acute Myeloid Leukaemia (AML), the most common type of leukaemia in adults. We will study the function of CD300f, a new receptor found on the surface of both healthy blood cells and the cancer cells in AML. We will investigate whether blocking this receptor with a newly developed antibody can be used to kill cancer cells. It is hoped that this study will allow development of this antibody for use in a future clinical trial.

Following inhalation, influenza virus infects cells of the immune system and epithelial cells lining the airways. Currently, it is not clear how influenza virus attaches to and infects these cells. This project will use new ways to identify receptors for influenza virus on human epithelial cells and immune cells. The appendix has been regarded as a useless organ, however, there are evidence showing its removal reduces the risk of developing inflammatory bowel disease. We have shown that this may be due to altered intestinal immune regulation. The project plans to explore the mechanisms responsible for this altered immune regulation. With knowledge of specific elements of disease causation gained from these studies, more effective and targeted treatment options will become available.

Summary
Reactive oxygen species (ROS) produced in cancers activate cell surface receptor signalling pathways that drive cancer progression. I will study links between ROS and receptor signalling in cancer cells, and inhibit signalling with ROS scavengers delivered in nanoparticles, targeted to receptor complexes with specific antibodies. These will include antibodies we raised against ADAM10, a protease associated with multiple receptor signalling pathways, to simultaneously inhibit these pathways.
