Summary
Proper functioning of the nonsense mediated mRNA decay (NMD or 'mRNA police') is crucial for any cell to ensure normal development and function. When NMD is compromised the outcome is learning and memory problems, autism or schizophrenia. Under this project we study malfunctioning NMD using stem and neuronal cells derived from patients' skin cells. Some of the affected genes might be considered for therapeutic interventions. The transcriptional co-repressor C-terminal Binding Protein (CtBP) in metabolic control. This project will provide insights into the genes that regulate the storage of fat. We will learn about basic biology but will also discover mechanisms that may be used to influence fat storage in human health. We will also consolidate Australia's expertise in the use of the genetic model organism, the worm C. elegans, and validate the findings in mammalian systems. RNA splicing: factors and mechanisms. Most primary gene transcripts must have their noncoding intronic sequences spliced out before the mRNA can be translated. Moreover, alternative splicing enables cells to generate a far more proteins than there are genes in the nucleus. Based on our proven success with ZNF265 we will isolate novel RNA interactors and their partners, colocalize these in intranuclear compartments, and elucidate their effect on pre-mRNA splicing. The potential impact of this work is widespread, because although it is known that ubiquitlyation has regulatory consequences in multicellular eukaryotes, individual networks have not been completely described in higher eukaryotes. A new mechanism of gene regulation. This project will advance our knowledge of how genes are switched on and off, by focusing on a very common class of gene regulatory proteins known as zinc finger proteins. The results of this study will improve our understanding of the fundamental molecular events that underpin gene regulation and how we might control it in fields such as biotechnology and gene therapy.

Every creature starts out as a single fertilized egg. The genome directs the embryonic development of the egg by regulating the expression of genes each of which must be turned on or off at the correct time and place. This essential balance between the activation or repression of genes is controlled by groups of proteins, including ‘transcriptional co-activators’ and ‘repressors’. This project aims to better understand the role of these proteins during embryonic development.
MicroRNA Networks That Safeguard The Functional Program Of Regulatory T Cells

Summary
A newly discovered group of molecules termed microRNAs are thought to function as rheostats for the activity of genes. We have shown that these molecules are critical for the function of an immune cell type termed regulatory T cells. Without these cells, the immune system is unable to prevent uncontrolled and destructive inflammation. This proposal aims to utilize diverse technologies to uncover the precise molecular mechanisms by which microRNAs safeguard the function of regulatory T cells.
Reducing skeletal malformations in cultured marine fish using gene expression, improved nutrition and advanced system operation. Reducing malformations in farmed fish will benefit the Australian economy and society by providing greater quantities of cheaper, higher quality fish. Increased farmed fish production, currently worth ~$300 million p.a., will increase exports and decrease imports (currently ~50% of all Australian consumed fish). To benefit are the important regional farming operations ....Reducing skeletal malformations in cultured marine fish using gene expression, improved nutrition and advanced system operation. Reducing malformations in farmed fish will benefit the Australian economy and society by providing greater quantities of cheaper, higher quality fish. Increased farmed fish production, currently worth ~$300 million p.a., will increase exports and decrease imports (currently ~50% of all Australian consumed fish). To benefit are the important regional farming operations in QLD, NSW, SA, NT, TAS and WA. In particular, the largest industry in Tasmania will profit by having a viable new species to farm (striped trumpeter) reducing risk due to climate change and global oversupply of salmon.
