Genes that regulate female reproductive organ development are of immense value for Australia as tools for seed improvement. Those from our preliminary screen have convinced our industry partners that they can be agents for engineering of apomixis or creation of fertile seed without fertilisation. This will allow the capture of hybrid vigour in wheat and rice, for which commercial hybrid seed production is not currently available. In wheat alone, apomixis presents for Australia an economic value of more than Aus$ ½ billion per annum. Furthermore, controlled apomixis will accelerate breeding programs that will bring drought resistance and minimal fertiliser requiring varieties to the farmer.Read moreRead less
Identification of nuclear reprogramming factors in oocyte cytoplasm. The mature oocyte contains dominant factors that are capable of erasing tissue specific gene expression profiles of somatic cells. These reprogramming factors would be valuable for dedifferentiation of cells and for nuclear transfer in animal cloning. The research involves determination of reprogramming factors present in active cytoplasm following enucleation of the germinal vesicle, blockage of transcription and translation, ....Identification of nuclear reprogramming factors in oocyte cytoplasm. The mature oocyte contains dominant factors that are capable of erasing tissue specific gene expression profiles of somatic cells. These reprogramming factors would be valuable for dedifferentiation of cells and for nuclear transfer in animal cloning. The research involves determination of reprogramming factors present in active cytoplasm following enucleation of the germinal vesicle, blockage of transcription and translation, and timed cultures. The assays will involve maintenance of reprogramming ability and erasure of somatic gene transcription. By subtractive elimination the function of isolated proteins which are involved in reprogramming will be identified for potential recombinant production.Read moreRead less

Exploiting the lymphatic system for next generation vaccine development . Vaccination is the most successful and cost-effective means of combating infectious diseases. This project will look at how vaccine adjuvants work and will help the development of new vaccines against infections in both animals and man. It will also promote the training of Australian scientists in the field of vaccine research and development.
Pancreatic Differentiation of Cord Blood Stem Cells using Smart Surfaces. Cord blood cells obtained at the time of delivery of a baby are a valuable resource that have the potential to develop into many cell types. This Project entails attaching stem cells derived from cord blood to appropriate 3 dimensional smart surfaces, and examining the ability of such cells to develop into insulin-producing cells. This Project entails attaching stem cells derived from cord blood to appropriate 3 dimensional smart surfaces, and examining the ability of such cells to develop into insulin-producing cells. An understanding of how to coax stem cells, seeded on to smart surfaces, to develop into mature cells with different functions will enhance our ability to understand how cells develop. As well, it enhance the potential usefulness of cord blood for research purposes. Read moreRead less
Pathophysiological mechanisms in equine dyschondroplasia (osteochondrosis). Dyschondroplasia (osteochondrosis) is a developmental orthopaedic disease of horses, which causes significant wastage within the Thoroughbred racing industry. Dyschondroplasia (osteochondrosis) is a developmental orthopaedic disease of horses, which causes significant wastage within the Thoroughbred racing industry. The disease affects growth cartilage beneath joint surfaces in such a way that cartilage fails to be replaced by bone, resulting in defects in the joint surface and lameness. This project will identify differences in gene expression between normal cartilage and early dyschondroplastic lesions, in order to identify the sequence of molecular events leading to induction of disease. The basic understanding of joint development obtained from this work will assist in the development of strategies to prevent and diagnose dyschondroplasia in horses.Read moreRead less
Bacterial Proteomics: From Cell Division to Novel Antibiotic Targets. When a cell divides it is essential that each newborn cell gets a complete copy of the DNA. To ensure that this happens, cell division must be tightly controlled. It is not known how this occurs in bacteria. However, if we knew what molecules were involved in this control, we could target them to kill harmful bacteria. When a cell divides it is essential that each newborn cell gets a complete copy of the DNA. To ensure that this happens, cell division must be tightly controlled. It is not known how this occurs in bacteria. However, if we knew what molecules were involved in this control, we could target them to kill harmful bacteria. This project aims to identify such regulatory molecules as candidate targets for antimicrobial agents, with a view to developing powerful, novel antibiotics to protect us from the imminent threat of bioterrorism and antibiotic-resistant bacteria.

Selective enrichment of proteins using micro-Gradiflow technology and characterisation of proteins using LC-MS/MS. This project describes crucial areas for the technology development of protein enrichment and fractionation. While current techniques are powerful, the usefulness to identify and characterise proteins of low abundance from massively complex samples in the presence of abundant proteins is limited and requires up to mililitre volumes. This project describes crucial areas for the technology development of protein enrichment and fractionation. While current techniques are powerful, the usefulness to identify and characterise proteins of low abundance from massively complex samples in the presence of abundant proteins is limited and requires up to mililitre volumes. We intend to develop the micro-Gradiflow technology in association with Gradipore as a means of protein fractionation using microlitre volumes of sample. This technology will be applicable to all areas of proteomic research and in particular to the study of cell differentiation. The outcome for Australian industry, our competitiveness both industrial and scientific, and the potential for economic advancement is tremendous.Read moreRead less

Eosinophils are involved in parasite immunity and in the pathology of allergic diseases including asthma. Eosinophil recruitment and activation is critical to their functional activity, but the relevant molecular mechanisms have not been fully elucidated. In the present proposal, a unique resource of eosinophils, at different stages of recruitment and activation, will be used for detailed proteomic analysis to identify molecules and molecular pathways involved in eosinophil migration and activation. These studies may lead to novel anti-inflammatory strategies and more targeted drug treatments that will generate significant intellectual property and be of enormous benefit to allergy patients worldwide.Read moreRead less

Embryonic stem (ES) cells are a primitive embryonic cell type that can be maintained and grown in vitro. Mouse ES cells can be instructed to develop into a wide range of specific adult cell types. Research into human ES cells has more recently commenced and has already resulted in the controlled production of specific nerve cells by our group. The following project aims to create respiratory lineages from both mouse and human ES cells. Such an undertaking thus aims to provide a basis for the treatment of respiratory specific diseases such as cystic fibrosis and emphysema.Read moreRead less
