    A New Clinical Tool To Assess Fitness-to-drive In Obstructive Sleep Apnea

    Summary
    Obstructive sleep apnoea (OSA) affects 1 in 4 middle aged adults. OSA results in impaired neurobehavioral function, excessive sleepiness, and more than 2-fold increased motor vehicle crash risk and workplace accidents. However, not all patients are impaired and identifying patients at risk is a daily clinical challenge. This project proposes new and simple measures that can accurately identify patients with OSA who are most vulnerable to alertness failure and motor vehicle accident risk.

    Microvascular And Macrovascular Disease In Snoring And Obstructive Sleep Apnoea: A Mechanism For Increased Stroke Risk.

    This project will address the important question of the vascular risk of stroke associated with snoring and obstructive sleep apnoea, by using retinal photography of the eye to directly visualise the vascular changes in the cerebral circulation. Retinal photography may provide a useful clinical tool to help reduce the risk of stroke in patients with snoring and sleep apnoea.

    Summary
    Obstructive sleep apnea (OSA) is a common sleep disorder which significantly impacts daytime functioning leading to excessive sleepiness, and problems with attention and thinking. Currently, the causes for cognitive impairment in OSA (including attentional lapses and performance deficits) are poorly understood. In the awake state, groups of neurons can briefly go “offline” as they do in sleep. These periods of “local sleep” may explain impaired task performance in OSA.

    Disturbances in cardiovascular control underpin many diseases yet little is known about how the brain controls the heart and blood vessels. This project uses brain imaging (fMRI) and concurrent nerve recording in awake human subjects to increase our understanding of how normal blood pressure is maintained and how different disease states influence this control.

    Fear Relapse: Neural Substrates Underlying Its Inhibition And Prevention

    Exposure-based therapies are effective for anxiety disorders such as post traumatic stress, but two challenges remain: 1) patients that have learned to inhibit their fear are likely to relapse, requiring further therapy; 2) many drop out of therapy since it is aversive and anxiety provoking. We use an animal model to: 1) identify the neural substrates underlying fear inhibition; and 2) determine the conditions that prevent relapse and encourage participation in treatment.

    The Effect Of Oxytocin On The Formation, Expression And Inhibition Of Fear Memories

    Oxytocin is a hormone peptide which reduces amygdala activation to threatening stimuli and reduces anxiety in people and laboratory rodents. These results suggest that oxytocin could be a valuable pharmacological adjunct to exposure-based therapy for anxiety disorders. However, several questions must be answered before its therapeutic potential can be determined. As such, this project examines the effects of oxytocin on fear-related behaviours in rats, and neural fear circuits in the amygdala.

    Obesity is a worldwide disease, reflecting an interaction between our environment (diet, physical activity) and genes. We know that a mother's unhealthy diet can predispose offspring to diabetes, and exercise can improve this, but the underlying mechanisms are poorly understood. Here we will examine how exercise can benefit offspring of obese mothers, and test a drug that mimics the effects of exercise. The proposed research will provide insight into ways of reducing the obesity epidemic.

    Low back pain is a leading cause of disability amongst Australians. A critical question is why some people get better after hurting their back while others do not. This project examines whether changes in the brain predict low back pain outcome.
