Summary
Granulosa cell tumours of the ovary (GCT) represent 5-10% of malignant ovarian cancers. They are distinct from the more common epithelial tumours and although considered to have a much better prognosis, they have a propensity to late recurrence. Recurrent or aggressive GCT have a poor prognosis. These studies will investigate the molecular basis of recurrence and aggressive behaviour in GCT. This will provide both prognostic information and also potential therapeutic targets.
Osteocytic SOCS3 Controls STAT3:STAT1 Balance And Bone Formation

Summary
The most promising new osteoporosis therapy is antibody-based inhibition of the sclerostin protein. We discovered that sclerostin is inhibited by oncostatin M (OSM) only when it binds to a receptor called LIFR, which then activates proteins STAT3 and SOCS3. If OSM binds a different receptor (OSMR) it increases STAT1 activity and destroys bone. This project will determine how to manipulate STAT3, SOCS3, and STAT1 to increase bone formation and provide new treatments for osteoporosis.

Summary
Obesity is increasing at an alarming rate worldwide and is a leading cause of morbidity and mortality. Obesity is causally linked to the development of insulin resistance, a prelude to type 2 diabetes. In this proposal we will define a novel liver centric mechanism by which insulin resistance and oxidative stress may promote the development of morbid obesity, type 2 diabetes and liver disease.

Summary
Insulin & leptin signal in the brain to lower blood glucose, suppress food intake, increase activity & increase energy expenditure. Obesity diminishes the abilities of insulin & leptin to signal. This proposal will determine if the enzyme TCPTP terminates insulin & leptin signaling in the brain. Our studies will provide insight into the molecular causes of obesity & may identify a novel therapeutic target for the treatment of obesity & type 2 diabetes.

We have discovered that the cell surface receptor for growth hormone travels to the cell nucleus in dividing cells, including cancer cells. Given the role of growth hormone in promoting growth postnatally, we seek to uncover how the nuclear receptor promotes proliferation directly, and by gene splicing. We have identified strong candidates for its direct actions through proteomics, and a DNA binding site for the receptor.
