    The Older Australian Twins Study (OATS) Of Healthy Brain Ageing And Age-related Neurocognitive Disorders

    Summary
    Ageing is associated with cognitive decline and dementia. It is still not completely understood what relative contributions genes and environment play in these. This project is an extension of the Older Australian Twins Study to examine genetic and environmental factors associated with late life brain changes and dementia, and will establish an internationally significant cohort for novel discovery.

    Modelling Of Clinic And Ambulatory Blood Pressure On Cardiovascular Risk And Outcomes

    Summary
    Whilst ambulatory blood pressure monitoring data has been shown to be a good predictor of cardiovascular events, there remains controversy as to its utility in clinical practice. This project will use data from existing population and clinical cohort studies to examine the role of ambulatory blood pressure in risk assessment and hypertension management in Australia and around the globe. The findings are likely to have a major impact on clinical guidelines for hypertension management.

    Young Onset Colorectal Cancer: Genetics Pathology And Environment

    Summary
    There has been a steady increase since 2002, in the age-standardised incidence of CRC in males under 45 years in Australia, contrasting with the stabilisation in incidence of CRC in males of age 45 years and over. Persons under 50 years are not routinely screened unless they have a significant family history of CRC. Young-onset rectal cancer is associated with late presentations and with a higher mortality. This proposal will address the possible risk factors for young-onset CRC.

    Do Exposures Before Conception Influence The Risk Of Asthma In Offspring?

    Asthma and poor lung function are major causes of public health issues. Emerging evidence suggests adverse exposures even before the conception of a child may cause these conditions. The proposed project is part of an international study across generations to identify these factors. This study will provide novel evidence to guide interventions and identify studies to advance this area further. These original findings will be of great importance both nationally and internationally.

    The Predictors Of Asthma And Lung Function Deficits In The Third Decade: Longitudinal Study Of MACS Sibships

    This will be the world’s first birth cohort study to use substantial prospective data to investigate how biological, psychosocial, and environmental markers from birth will predict asthma and lung function in the third decade of life. Our findings will be crucial to the development of new policy and practice for the prevention and management of these conditions and uncover crucial risk factors for young adult asthma.

    Young Adult Myopia: Genetic And Environmental Associations

    Summary
    Myopia affects 80% of school leavers in the cities of East Asia, 45% of Asian Australian school leavers and is probably on the rise in European Australian adolescents. Increased levels of education and lack of time outdoors are known to increase the risk of myopia. We will examine 2,000 young adults to find the genes that interact with these risk factors. In addition to confirming when these risk factors are most important, identifying molecular pathways opens the avenue of new treatments.

    Identifying Glaucoma Risk Variants In The Norfolk Island Genetic Isolate

    Primary open angle glaucoma is the most common form of glaucoma. In this project we will focus on the identification of functional genetic variants influencing development of this disorder, using a powerful whole exome sequencing approach in a large multigenerational pedigree from the Norfolk Island population isolate. The identification of genes influencing glaucoma development would provide invaluable clues to aid in defining the pathophysiology of this common disease.

    Do Breast Cancer Risk Factors Differ According To Underlying Genetic Susceptibility? A Pooled Analysis Of Prospective Studies From The NCI Cancer Cohort Consortium

    We propose to use data from 23 international prospective cohort studies in the Cancer Cohort Consortium organised by the US National Cancer Institute to evaluate gene environment interactions for women who are at increased genetic risk of breast cancer. Our ultimate goal is to enhance the performance of clinical prediction tools and to develop targeted evidence-based strategies to mitigate the high absolute risk of breast cancer for women at increased genetic risk of the disease.

    A Randomised Clinical Trial Of Physical Activity To Delay The Progression Of Cerebrovascular Pathology

    The aim of this randomised clinical trial is to establish whether a 24 months physical activity (PA) program for older adults with memory problems can delay the progression of damage to the blood vessel system in the brain, measured on an MRI scan. It also will investigate whether cognition, mood, quality of life, functional level, fitness and biological markers will improve with the intervention. If successful this PA program could become part of clinical care for adults at risk of dementia.

    Quantifying The Role Of Epigenetic Factors In Neurocognitive Outcomes: A Twin Study

    We aim to identify the environmental factors in early life that contribute towards an individual brain development using MRI brain scans and related psychological skills measured in late childhood.
