Summary
Asthma and chronic obstructive pulmonary disease (COPD) are characterised by enhanced TGF? expression, which is accompanied by susceptibility to recurrent viral and bacterial infections. Such infections exacerbate lung inflammation in these patients, generally requiring emergency department treatment. This project proposes to clarify the therapeutic potential of TGF? inhibitors to reduce the impact of viral infections in patients with COPD and asthma.

Plasmin is crucial enzyme present in blood plasma that functions in clot dissolution, inflammation, tissue remodeling, and wound healing. We aim to study how this enzyme system is controlled, by studying its interaction with receptors, co-factors and inhibitors. The information we gain will help drive the development of new generation therapeutics for the fine control of plasmin function in clotting disease, bleeding and inflammation.

Summary
Guidelines currently recommend lifelong treatment for patients with chronic hepatitis B, with associated cost and risks of drug resistance and side effects. It has recently been suggested that up to 50% of patients may safely and successfully stop drug after long-term treatment. Our project will identify which patients can safely stop treatment, by performing detailed studies of the human immune system and the hepatitis B virus. This will be an important advance for patient care.
Antibiotic resistance is a looming public health crisis. New antibiotics with new mechanisms of action are desperately needed. The long-term goal of this research is to develop new drugs that disarm bacteria to overcome the problem of antibiotic resistance.

Summary
Chronic infectious diseases such as HIV, hepatitis B and tuberculosis impose a massive global health burden and new treatments are desperately needed. This proposal investigates a new approach to improve immune responses and clear chronic infections. Our multidisciplinary team will define the molecular and cellular biology underlying this approach and translate our findings by re-purposing a drug already approved for other indications in humans.

Lung infections such as pneumonia are common in Indigenous children, and can lead to repeated hospitalisation and permanent lung damage. They are also an important cause of preventable death in children. This study will look at the role of bacteria and viruses in lung infections, and will inform interventions to reduce the burden and consequences of lung infections in Indigenous children.
Targeting Hypermutable ‘superbugs’ In Chronic Respiratory Infections By Optimised Antibiotic Combination Dosage Regimens

Summary
Many bacterial ‘superbugs’ can increase their mutation rate, i.e. become hypermutable, and thus rapidly become resistant to multiple antibiotics. Chronic lung infections with hypermutable bacteria cause increased ill-health and death in patients and current treatments do not work well. We will develop improved treatments using combinations of available antibiotics. This project will provide guidance to doctors on how to treat infections more effectively and minimise emergence of resistance.
This proposal investigates processes that regulate the cell cytoskeleton to control shape and the dynamics membranes, with a view to developing a generic antiviral therapy. As viruses rely upon the cell cytoskeleton to initiate an infection, we posit that enzymes that control the cytoskeleton can be targeted to block infection.

The demand for prosthetic joint replacement surgery will double over the next decade. Complications involving the surgical wound are a devastating complication of this surgery. This randomised controlled trial aims to investigate whether skin cleansing prior to surgical incision with alcoholic iodine reduces the number of patients with superficial wound complications when compared to alcoholic chlorhexidine. The study will be performed in patients undergoing orthopaedic joint replacement surgery ....The demand for prosthetic joint replacement surgery will double over the next decade.
