Summary
There is a strong association between allergy and asthma. This association been almost universally assumed to be causative. However, recent evidence suggests an alternative explanation ie., that the abnormal epithelium in asthma allows or facilitates sensitization to airborne allergens. This project will test this alternative hypothesis using human lung tissue and an animal model.

Summary
Perthes disease (PD) occurs following loss of blood supply to the hip (femoral head). It is a severe childhood disorder with over 250 new cases per year in Australia. PD results in flattening of the normally round femoral head and painful arthritis. The hip becomes flat because the bone is eaten by cells called osteoclasts. We will test if a medicine aimed at stopping these osteoclasts can prevent hip flattening. This should decrease the risk of arthritis and the need for hip replacement.
The Clinical Impact Of Event-based Motion Correction In Paediatric PET-CT Brain Imaging

Summary
Movement of the head during PET-CT brain imaging can prevent accurate diagnosis by blurring and distorting the image. The problem is perhaps most acute in paediatric patients, many of whom must be anaesthetized or sedated to avoid motion. This work will establish whether a recently developed motion correction method can improve the clinical utility of PET-CT brain images in young patients, and reduce the need for sedation and anaesthesia during the PET scan.

Summary
Recent studies have established that both human genetic susceptibility and viral infections during early childhood are important drivers of asthma development. It has also been noted that asthmatics’ airways are colonized with different bacteria to non-asthmatics. In this project we will examine how genetic susceptibility and interactions between bacteria and viruses in children's airways promote the development of allergy and asthma.
This study will follow up a birth cohort for 25 years with the aim of looking at critical periods for bone development including the role of in utero exposures, early infancy, age 8 and age 16.

Summary
Our previous work indicates that evoked human blood borne factors confer protection against injury, due to loss of blood flow in heart muscle, when a brief stress is remotely applied to a limb (remote ischemic preconditioning). We have identified these proteins that appear to activate genetic and metabolic regulation of adaptive cell survival processes. We will now test their individual and combined capacity, efficiency and mechanisms of protection in the heart using cell and clinical models.

Summary
Cystic fibrosis is the most common life shortening inherited disease in Caucasians. Lung damage starts in infancy and lung function falls most rapidly in adolescence although why and how this happens and early life determinants are not known. This study takes advantage of a previous study that monitored young children from 3 months to 5 years of life and follows them closely through early adolescence to investigate the protective and risk factors for falling lung function.

The Perth Respiratory Birth Cohort Study began in 1987 and has assessed 253 children before birth and at follow-up at 1, 6 & 12 months, and 6, 11 & 18 years of age. The study has resulted in 63 publications and achieved consistent international prominence. The proposed study of the cohort at 24 yrs aims to determine the respiratory and immunological status of the cohort as young adults. This follow-up is essential to determine how early risk factors affect respiratory outcome in young adults.
Cancer Risks From Low-dose Ionising Radiation Following Diagnostic Medical Procedures

Summary
Our study investigates cancer risk following exposure to low doses of ionising radiation from medical procedures. Our first major paper linked over 800,000 CT exposures to cancer outcomes in a cohort of almost 11 million young Australians, and found that CT exposure predicted an increased incidence of leukaemia and most solid cancers. In our ongoing work we will incorporate nuclear medicine and other diagnostic x-rays, and estimate radiation dose for individual procedures and to specific organs.
