Summary
Cystic fibrosis is the most common life shortening inherited disease in Caucasians. Lung damage starts in infancy and lung function falls most rapidly in adolescence although why and how this happens and early life determinants are not known. This study takes advantage of a previous study that monitored young children from 3 months to 5 years of life and follows them closely through early adolescence to investigate the protective and risk factors for falling lung function.
Cancers of the skin are the most common tumours in humans, and their diagnosis and treatment impose the largest costs on Australia’s cancer budget. While much has been learned about the roles of sunlight and skin type as risk factors for skin cancer, relatively little is known about the genes conferring risk. This study will compare the genetic profiles of over 6000 patients with skin cancer to 3000 people without skin cancer to pinpoint the genes responsible for skin cancer.

While the burden of dementia in Australia occurs late in life, the underlying brain disease accumulates for decades prior to dementia onset. Disease modifying interventions have the greatest potential to avert later disease burden if introduced during this crucial window, well before the onset of clear cognitive decline. To reduce Australia's future dementia burden, it is thus imperative to identify those Australians at risk of dementia whilst they are still relatively young.

This study aims to investigate the causes of alcohol problems in adult Australians. We will follow-up participants from a birth cohort study who are now in their thirties. We will assess long term outcomes from in-utero exposure to alcohol and biological, developmental and genetic predictors of alcohol disorders in adulthood. This study also aims to study genetic factors which may be important in the development of alcohol abuse and dependence.

Summary
Observational studies suggest that modifiable risk factors such as low vitamin D levels, coffee consumption, alcohol consumption and obesity may be important in cancer risk. However, observational studies can only demonstrate association between a risk factor and cancer, and association does not equal causation. We present an alternative approach to help determine which risk factors actually cause cancer.

Post-traumatic stress occurs in children following traumatic physical injury. However it is not clear to what extent this is so for children who have received a traumatic brain injury. Post-traumatic stress occurs in children following traumatic physical injury. However it is not clear to what extent this is so for children who have received a traumatic brain injury. Furthermore, when there is a traumatic brain injury and traumatic stress, it is not clear how these interact, how they influence long-term outcomes, and what factors such as pre-injury functioning and family support and distress mediate outcomes. These issues are very important since effective rehabilitation of children following traumatic brain injury is essential to maximise long-term functioning and minimise disability. To be effective, rehabilitation must be guided by the knowledge about key factors that determine the recovery process. This study aims to provide answers to these questions by following two cohorts of children (aged 6-14) over 18 months after receiving a traumatic brain injury. In total 240 children will be recruited from Brisbane and Melbourne hospitals. They will be assessed at three, six, twelve and eighteen months post-injury using measures of cognitive, psychological and social functioning. Information on parent distress and behaviours will also be obtained. The aim of this project is to examine the relationship between sleep duration (5, 6, 7, 8, or 9h per day for one week) and glucose metabolism. This will allow us to quantify the amount of harm that different levels of sleep loss cause to the physiological systems that protect people from developing serious health disorders. In particular, the results of the project will be invaluable in the design of effective behavioural interventions for the prevention and/or treatment of type 2 diabetes.

Myopia affects 80% of school leavers in the cities of East Asia, 45% of Asian Australian school leavers and is probably on the rise in European Australian adolescents. Increased levels of education and lack of time outdoors are known to increase the risk of myopia. We will examine 2,000 young adults to find the genes that interact with these risk factors. In addition to confirming when these risk factors are most important, identifying molecular pathways opens the avenue of new treatments.
