Development of natural herbal medicines for managing metabolic syndrome in order to prevent type 2 diabetes. Type 2 diabetes affects almost 7% of Australia~{!/~}s population. Its incidence continues to grow, as indicated by high rates of metabolic syndrome, a pre-stage of type 2 diabetes. Using novel and bio-molecular techniques, we will evaluate the effects and mechanisms of three natural compounds from anti-diabetic Chinese herbs on cellular and rodent models with metabolic syndrome. This study will provide scientific evidence as to how and why herbal compounds improve insulin resistance and abnormalities in glucose and lipids metabolism. The outcome of this project will support the development of natural products to manage metabolic syndrome and prevent type 2 diabetes.

Estrogen signalling in gonadotropes. Estrogen action is a normal prerequisite for cyclic function of reproduction in the female, but little is known about how this important hormone acts in the relevant cells of the pituitary gland (gonadotropes). In order to gain information on normal function, we will conduct studies on gonadotropes treated with estrogen in a range of paradigms. The information will be valuable in understanding normal reproduction, but will also form the basis of further studi ....Estrogen signalling in gonadotropes. Blood flow routes in muscle. Ageing well, ageing productively. The Australian population is ageing. The proportion of the population over the age of 65 is expected to greatly increase, reaching 22% by the year 2030. The prevalence of type 2 diabetes in this older population is thought to be ~20%, compared to ~6% in younger populations. An initial cause of type 2 diabetes may be microvascular dysfunction brought on by physical inactivity. Therefore this project addresses the concepts of microvasc ....Blood flow routes in muscle. An initial cause of type 2 diabetes may be microvascular dysfunction brought on by physical inactivity. Therefore this project addresses the concepts of microvascular function and microvascular fitness by using the latest technology to map blood flow routes in muscle under a number of relevant situations. Investigation of the function of Sel S a novel selenoprotein. The long term aim of this project is to find a way to prevent or delay the onset of both Type 1 and Type 2 diabetes. ChemGenex pharmaceuticals, our commercial partners have discovered and patented a selenoprotein with antioxidant properties and have shown in vitro that it protects insulin-producing beta cells from oxidative damage. This project aims to prove, in an in vivo setting, that this protein can prevent or delay the onset of d ....Investigation of the function of Sel S a novel selenoprotein. Microdialysis for monitoring changes in microvascular flow patterns in muscle. Microdialysis is a technique for sampling interstitial fluid. Factors altering vascular delivery and removal of nutrients and hormones can affect muscle metabolism by altering exchange with the interstitium. This project focuses on microdialysis for assessing the impact of microvascular blood flow patterns on skeletal muscle metabolism and contractility. The aim is to develop and refine the technology, including equat ....Microdialysis for monitoring changes in microvascular flow patterns in muscle. The aim is to develop and refine the technology, including equations, specifically for monitoring the nutritive fraction of blood flow in muscle by agents and factors relating to health and disease. This technique could be used for screening drugs in the treatment of diseases such as type 2 diabetes and related conditions.Read moreRead less

There are a number of concerning trends in reproductive health. Women are reporting difficulty conceiving and maintaining pregnancies; while sperm count and quality are declining in men. More concerning is the increase in reproductive cancers. Gonadotropin-releasing hormone (GnRH) antagonist and agonist have been used for decades to treat reproductive cancers (such as breast cancer and prostate cancer), infertility and precocious puberty. Kisspeptin and RF-related peptide may offer more physiological alternatives to GnRH, without detrimental side effects. We will fully explore these two newly defined and major players in reproduction and provide a physiological framework for their progression to clinical use.Read moreRead less

Depression has been reported to be twice as prevalent and more persistent in individuals with diabetes than in the general population. The current research will investigate the point-prevalence of depression in a large sample of Australian diabetic outpatients using a case-finding instrument (CES-D) and follow-up structured clinical interview where appropriate. Demographic, medical, and psychological correlates of depressiono will be investigated. The effects of the screening procedure and diagnosis on help-seeking behaviour, course of the depression, self-care, and glycemic control as well as qualtiy of life will be evaluated after six months.Read moreRead less
