Summary
Defects in penis development are among the most common birth abnormalities, and affect around 1 in every 150 live male births in Australia. Development of the penis is known to be driven by male hormones (androgens), but we have recently shown that oestrogen also plays a role in this process. This project will define the role of estrogen in penis development and how a loss or gain of estrogens can cause developmental defects.
Disorders of sexual development are amongst the most common birth defects in humans. These conditions have profound consequences for the physical and psychological health of affected individuals. The incidence of these disorders is on the rise, and this has been linked to our increased exposure to chemicals in the environment that affect how our hormones function. This project will investigate how these chemicals affect hormonal pathways that control early development.
Breast cancers have diverse characteristics such as their response to treatment and their propensity to relapse. We know that the individual suit of oncogenic lesions probably influences diversity but the characteristics of the cell type from which the cancer arose probably also plays a part. This Application addresses this question and investigates a major new discovery made by the applicant that the ets transcription factor Elf5 plays a key role in specifying the diversity in breast cancer.

We know very little of the genes that control development of the ovary in female fetuses; most study has focused on the formation of the testis in males. We will use a novel experimental model, a marsupial, where by hormonal treatment of developing males we can switch off testis formation and activate the ovarian pathway. Summary
Schizophrenia is characterized by psychosis, social and occupational dysfunction and cognitive deficits. Males are more often diagnosed and more severely impaired than females. Onset in males is most frequent during adolescence, a time of increasing sex hormones. We ask, how do sex hormones act on the adolescent male brain to impact the onset and symptoms of schizophrenia? The answer will allow development of gender and age-specific interventions to prevent onset or ameliorate symptoms.
In men, oestrogen may be important for strong bones and maintaining a healthy weight. Men with prostate cancer are given medical castration treatment to decrease testosterone, because testosterone is required for prostate cancer growth. Because oestrogen is derived from testosterone, they also have very low oestrogen levels. We want to conduct a trial in these men to find out whether giving back oestrogen will prevent bone loss and weight gain, among other health benefits.

Summary
These studies provide insights into the mechanisms and role of oestrogen in regulating whole body and liver fat metabolism. Oestrogen-related medications that modify the action or tissue availability of oestrogen are widely used therapeutics and can predispose to obesity and fat accumulation in the liver. Whether the effect is direct or through interplay with other metabolic hormones is unknown. This proposal examines their metabolic consequences and impact on obesity and liver health.
