We have discovered an innovative biochemical strategy whereby our novel compounds exploit and trick a part of the detoxification machinery, that is the transporter, P-glycoprotein, to specifically kill drug resistant cancer cells. Herein, we take advantage of this biochemical mechanism to design novel and safe drugs to selectively target resistant tumours.

Summary
Stillbirth occurs in 35 times as many pregnancies as sudden infant death but the causes are unknown. This project will help to develop tests that can predict the risk of stillbirth so that the obstetrician can deliver the baby before it dies. The investigators hypothesise that stillbirth is due to aging of the placenta and that markers of the aging placenta can be detected in the mother’s blood. The project brings together experts in the placenta, aging and obstetric care of high risk pregnancy.

Summary
Most heart attacks are the consequence of the acute rupture of plaques in arteries that supply our heart with oxygen and nutrients. Current standard tests cannot distinguish plaques that likely rupture from plaques that do not rupture. Similarly, little is known about the processes that determine whether a plaque is vulnerable to rupture or stable. The current project examines the involvement of two processes - either alone or in combination - in determining plaque stability/vulnerability.
Cardiovascular disease is a leading cause of death in Australia, accounting for 36% of all deaths in 2004-05. Diseased blood vessels are its most common form, and the underlying process is atherosclerosis. Atherosclerosis is characterised by plaque formation in blood vessels. Plaque formation is problematic, and may lead to blood vessel blockage. We aim to identify novel targets that prevent plaque formation.

This research project is designed to elucidate how the quality of a father’s spermatozoa can impact upon the health and wellbeing of his children. We hypothesize that factors, such as infertility, heavy smoking or age create a state of oxidative stress in the testes and that this stress influences the genetic structure of spermatozoa in such a way that the incidence of spontaneous mutations and susceptibility-to-cancer are significantly elevated in the offspring.

There is a need for a therapy that can be given before a mother gives birth to protect the baby should ‘oxygen starvation’ threaten the baby’s brain and other organs such as the heart, kidney, lungs, and the ability to breathe properly. We are suggesting that an increased intake of creatine is a very effective treatment against this threat, and its proven safety and ease of use recommends it for wide application, particularly in countries where the access to medical resources is poor.

Plasma non-transferrin bound iron (NTBI) levels are elevated in iron overload disorders. Excess NTBI has serious health consequences as it is toxic and may induce cellular dysfunction and injury. We will investigate the molecular mechanisms by which NTBI transport is regulated, the contribution of NTBI to the development of iron overload and its impact on oxidative-mediated liver and heart injury in iron overload conditions associated with Hereditary Haemochromatosis and thalassaemia.

We are conducting four large clinical trials testing anti-inflammatory treatments like ?-3 PUFAs and aspirin in young people who are at high-risk for psychosis or have depression. This project builds upon our recent findings showing that a protein, called caspase-2, is involved in protecting against oxidative damage and ageing in a mouse model. We propose to carry out further studies to fully understand how caspase-2 regulates cell’s response to oxidative stress. Such studies will be critical in assessing if caspase-2 or its substrates are useful targets for drugs to treat diseases that may arise due to cell’s inability to properly respond to oxidative damage.
