Among the most significant but often overlooked conditions in the elderly is depression, which is associated with marked disability, functional decline, risk of hospitalization, diminished quality of life, caregiver burden, increased service utilization, and mortality from comorbid medical conditions or suicide. The World Health Organization predicts that by 2020 depression will be second only to heart disease as a cause of disability and premature death in established market economies. Depression is missed in approximately half of all elderly persons with mood disorder, and this frequency is certainly higher among individuals with dementia. The strategy to diagnose depression in dementia needs to be revised. Patients' reports are often unreliable due to memory problems and lack of full insight into their mood and behavioural changes. Caregivers may overestimate patients' symptoms of depression, especially when they themselves are depressed and overburdened, and clinicians' diagnoses are sometimes based on biased information and short observation periods. The situation in nursing homes is even worse, and there is a shortage of studies on how to diagnose depression in institutionalised patients with dementia. Using specific instruments to assess mood, behaviour and cognition we will develop valid and reliable criteria to diagnose depression in the different stages of dementia, and for patients living in different settings. We have previously made the most widely used animal brain atlas in the world. This atlas based on stained histological sections of the rat brain. In recent years, advances in MRI have made it possible to generate images of the rat brain at very high resolution. We have obtained a very high quality MRI image set from colleagues in Duke University in the USA, and we have begun to map these images in great detail, using our histological atlas as a guide.

Summary
Cardiovascular disease accounts of over one third of all deaths in Australia and is forecasted to become the leading cause of death in Australia and worldwide. Currently, atherosclerosis and thrombosis in coronary arteries are diagnosed by X-ray angiography, an invasive and risk-associated procedure. Our project will explore new contrast agents for the fast, reliable and safe diagnosis of cardiovascular diseases and thrombotic events, which is beneficial to many patients.

Summary
Stroke is a major cause of death and disability in our community and is most often due to a blocked blood vessel. Powerful treatments are available to open blocked blood vessels but there are restrictions on eligibility and some risks. This research aims to improve doctors' understanding of stroke in the individual patient to aid treatment decisions by identifying those with potentially salvageable brain tissue and avoiding those with high risk of bleeding complications.

Summary
In this project we aim to establish a reliable vascular baseline to improve mapping of both small-scale functional architecture and large-scale brain networks in functional human brain mapping using MRI. By mapping the grey matter vasculature with high detail in both humans and animals, and by computing and matching of these atlases across species we will be able to validate this approach in vivo to confirm the better spatial specificity of the newly developed approach.

Atrial fibrillation (AF) is an abnormal heart rhythm which is a common cause of stroke. AF can often go unrecognized as patients may have no symptoms. This project aims to develop computer software which can automatically detect underlying AF based on MRI brain scan pattern, in patients who have had an acute stroke. This project has the potential to offer several benefits, including reduced need for costly investigations, improved AF detection and a larger pool of patients being treated for AF.

Summary
Many people with traumatic brain injury (TBI) experience cognitive problems, including poor memory and concentration. TBI is often referred to as a ‘hidden disability’ because the overwhelming majority of patients with TBI show no abnormalities on standard MRI or CT scans. In my project, I will delineate the mechanisms of secondary injury in finer detail through enhanced neuroimaging techniques, resulting in new assessment and treatment modalities for individuals with TBI.
