Genetics and evolution of Shigella O antigens. We use genome scale sequencing techniques to sequence 26 O-antigen gene clusters from Shigella. With the seven already known, this will give sequences for every O-antigen of Shigella. This will be the first time that such set is fully sequenced. Shigella are human specific pathogens, have emerged with the evolution of humans. O-antigens are important for their life and pathogenicity. O-antigens are important for their life and pathogenicity. This project will greatly extend our knowledge of the genetic basis and evolution of this important polymorphism. O-antigens are used for typing Shigella and also elicit strong immunity. The molecular data will help establish DNA based typing and vaccine development.Read moreRead less

Infection with Cyclospora cayetanensis is an emerging disease that causes significant morbidity in human populations. Although prevalent in developing countries, recent outbreaks in the USA confirm it is now emerging into the developed world. The research described here will analyse and characterise the antigens on the surface of the oocyst of C. cayetanensis which will add significantly to our sparse knowledge on this important human parasite. We will also develop the first commercial antibodies and technologies which will rapidly and accurately detect the parasite in human patients and our drinking water supplies, thus securing a global market for Australian technology.Read moreRead less
Improved methods for quantitation of acute phase proteins in biological samples. Using monoclonal antibodies and fluorescence polarisation, we aim to develop improved quantitative analytical methods that are superior to the current clinical assays. The initial targets will be C-reactive protein (CRP) and serum amyloid precursor protein (SAP), but the technology should be readily adaptable to other serum proteins. Better assays for CRP and SAP will greatly facilitate improved clinical management of those at risk of heart attack, the single biggest contributor to healthcare costs in Australia. We further aim to adapt this technology to enable "point-of-care" assays that would help medical practitioners, especially in rural areas, to make informed diagnoses immediately.Read moreRead less

West Nile virus (WNV) is transmitted by mosquitoes and causes a fatal disease of the brain in animals and humans. WNV is currently exotic to this country, however a closely related, but benign cousin of this virus (Kunjin) can be found in Northern Australia. Currently it is very difficult to differentiate between infections with WNV and Kunjin using standard laboratory tests, therefore it will be difficult for public health officials to detect WNV if it is introduced to Australia. In this project we aim to develop more accurate laboratory tests to detect infections with the deadly, exotic WNV using new strategies. Read moreRead less
Long Chain Omega-3 Polyunsaturated Fatty Acids and Heart Health in Humans. The primary aims of the project is to assess the effect of DHA rich tuna oil on cholesterol carrying lipoproteins and to determine a quicker method in assessing someone's omega-3 status. The expected outcomes include the determination of: 1) the most appropriate blood sample and a rapid test and for evaluating the omega-3 status; 2) the minimum dose of tuna oil required which will not only improve a person's omega-3 status but also improve their plasma lipid profile and hence reduce their overall risk for cardiovascular disease (CVD); and 3) the use of the Omega-3 Index by medical practitioners. The significance is the increased knowledge of the health benefits of DHA rich tuna oil and that a new CVD risk marker will be generally accepted.Read moreRead less

Gel dosimeters main advantage is dose determination in 3-dimensions. Their main limitation is low spatial-resolution. We propose fabrication of special-type gels and for the first time use the 4.7 Tesla mini-scanner for dose mapping. Increased magnetic field combined with a small aperture for field dissipation will greatly improve spatial resolution down to micrometers (micro-dosimetry). This new technique will render gel-dosimeters suitable for applications in radiotherapy, industrial and all other radiation fields. Moreover, we will employ new parameter for dose mapping, which is expected to be more sensitive to radiation than the currently used relaxation timesRead moreRead less

Using a multidisciplinary approach, this project will determine viral parameters that correlate with the disease status of koalas and lead to improvements in diagnosis and management of disease in koala populations. The planned research will also provide some insight into cross-species transmission of retroviruses.Read moreRead less

Microdialysis is a technique for sampling interstitial fluid. Factors altering vascular delivery and removal of nutrients and hormones can affect muscle metabolism by altering exchange with the interstitium. This project focuses on microdialysis for assessing the impact of microvascular blood flow patterns on skeletal muscle metabolism and contractility. The aim is to develop and refine the technology, including equations, specifically for monitoring the nutritive fraction of blood flow in muscle by agents and factors relating to health and disease. This technique could be used for screening drugs in the treatment of diseases such as type 2 diabetes and related conditions.Read moreRead less
