Dr. Murphy’s work is centred on examining how different diseases such as obesity, diabetes and other inflammatory conditions increase people’s risk of heart disease. He will explore how these various inflammatory diseases influence the produce increased levels of white blood cells, which is important as increased white blood cells are positively associated with disease severity. He will also explore the relationship between blood cancers, know as leukaemia and how obesity.

Summary
I will develop new therapies for multiple sclerosis and spinal cord injury. I will also evaluate diagnostic and therapeutic uses for macrophage inhibitory cytokine-1 (MIC-1/GDF15) that was discovered in Australia. I will confirm its use in screening for bowel cancer. Also, I will look at using it in the indigenous and wider Australian community to improve health and close the gap in life expectancy. Finally, I will conduct trials of MIC-1/GDF15 therapy of obesity and inflammation.

Summary
Killer cells are white blood cells that destroy cancerous cells. To move to their next target they must quickly detach from a dying target. Failure of detachment results in excessive inflammation and tumour escape. This project will discover the detachment signals required to ‘release’ a locked-on killer cell. This will lead to a deeper understanding of immune pathology and new ways of treating cancer.

Summary
Blood cancers such as acute myeloid leukaemia (AML) are among the most deadly types of cancer and new treatments are desperately needed to improve patient’s survival in these diseases. AML cancer-causing stem cells survive by turning on immortalization programs and we hope to specifically kill these AML stem cells by blocking these crucial pathways. This includes things that control the way the cells divide and the way they respond to genetic damage as well as other novel pathways.

Summary
In our recent Cell paper (January 15, 2015), we reported our discovery of two new adult stem cells. One of these, the osteochondroreticular (OCR) bone stem cell (pronounced “ocker” stem cell) has a particular affinity for developing cartilage. We are testing if this cell could be used as a new treatment for osteoarthritis. The second new stem cell we discovered, is called the iRSC, and lies within the intestine. We are studying this cell in the development and treatment of bowel cancer.

Summary
The human immune system requires T cells for survival. Specialised populations of T cells exist that patrol the body and target unwanted lipid molecules expressed by bacteria or by cells that have become abnormal or cancerous. I will identify these T cells in human blood and skin and determine their role in protection against disease. I will explore the types of lipids molecules recognised by these T cells and use this information to help prevent human diseases.
