This project proposes to identify novel components and develop new inhibitors of an epigenetic pathway known to control the immune cells that cause allergic asthma. Our objective is to use these drugs to treat allergic disease.

Summary
Hepatitis C virus is one of the most significant human pathogens. There is no vaccine for HCV, and the antiviral treatment is expensive and does not stop reinfection. This project will study how the immune system of people infected with HCV generates antibodies to clear the virus. This will inform research efforts to design successful preventative vaccine to protect against this viral pathogen.
Natural Treg are dependent on the transcription factor FOXP3, but the mechanism of action of FOXP3 is only now becoming defined for human Treg. Tregs are critical for a balanced, responsive immune system, and deviation from this balance results in autoimmune diseases or persistence of cancers. Summary
Despite knowing a lot about immunity in a mice, functional analysis of the human immune system has been a major challenge. I will study defects of immune cells in humans with gene mutations that cause an antibody deficiency. With new insights from these unique clinical samples, I will functionally dissect human immune responses, directly translate these to chronic inflammatory disease, and provide implications for future vaccine development and cancer treatment.

Interleukin-6 (IL-6) is a member of a family of cytokine proteins that may be causative factors in many neurological disorders where they are involved in diverse processes including inflammation, neuronal injury and repair. In this project we will study how IL-6 affects the brain to bring about these outcomes. The results of this work will advance our understanding of how members of this cytokine family function and how they contribute to neurological disease.
